Co(II), Ni(II), Cu(II) and Mn(II) complexes synthesized by reflux of 6-bromo-3-(3-(4-chlorophenyl)acryloyl)-2H- chromen-2-one, Ciprofloxacin and various transition metal. 1H, 13C, IR and ESI Mass confirm the formation of ligand. The metal complexes were characterized on the basis of various spectroscopic techniques lik. IR studies and elemental analysis while the geometry of complexes was octahedral which is confirmed by electronic spectra and thermogravimetric analysis. The compounds were subjected to antimicrobial, antioxidant and anti-tubercular activity screening using serial broth dilution method and Minimum Inhibitory Concentration (MIC) is determined. Mn(II) complex has shown significant antifungal activity with an MIC of 6.25 μg/ml while Cu(II) complex is noticeable for antibacterial activity at the same concentration. Anti-TB activity of the ligand has enhanced on complexation with Ni(II) and Co(II) ions. While Ni(II) complex shows superior antioxidant activity than other complexes.

Vidangadi churna is a popular Ayurvedic formulation described in the chapter k.imicik.tsa of the Ayurvedic literature Cak.adatta for the treatment of k.imiroga. The preparation is a composite mixture of the fine powder of fruits of Vidang （Embelia ribs）, glandular trichomes of the fruits of k.mala （Mallotus philippensis）, mature fruits of Harde （Terminalia chehula）, Saindhava and Yavak.hara. The use of reversed phase C18 column eluted with gradient mobile phase of acetonitrile and water enabled the efficient separation of the chemical mark.rs in 22 rain. Validation of the method was performed in order to demonstrate its selectivity, accuracy, precision, repeatability and recovery. All calibration curves showed good linear correlation coefficients （r2〉0.995） within the tested ranges. Three mark.rs in Vidangadi churna were quantitied with respect to Embelin （0.647%, w/w）, Rottlerin （4.419%, w/w）, and Ellagic acid （0.459%, w/w）. lntraand inter-day RSDs of retention times and peak.areas were less than 3.12%. The recoveries were between 99.66% and 102.33%. In conclusion, a method has been developed for the simultaneous quantification of three mark.rs in Vidangadi churna. The RP-HPLC method was simple, precise and accurate and can be used for the quality control of the raw materials as well as formulations.

Despite the promise of immunotherapy such as the immune check.oint inhibitors(ICIs)anti-PD-1 and anti-CTLA-4 for advanced melanoma,only 26%–52%of patients respond,and many experience grade III/IV immune-related adverse *** by the need for an effective therapy for patients non-responsive to clinically approved ICIs,we have developed a novel nanoimmunotherapy that combines locally administered Prussian blue nanoparticle-based photothermal therapy(PBNP-PTT)with systemically administered agonistic anti-CD137 monoclonal antibody therapy(aCD137).PBNP-PTT was administered at various thermal doses to melanoma cells in vitro,and was combined with aCD137 in vivo to test treatment effects on melanoma tumor progression,animal survival,immunological protection against tumor rechallenge,and *** administered at a melanoma-specific thermal dose,PBNP-PTT elicits immunogenic cell death(ICD)in melanoma cells and upregulates mark.rs associated with antigen presentation and immune cell co-stimulation in ***,PBNP-PTT eliminates primary melanoma tumors in vivo,yielding long-term tumor-free ***,the antitumor immune effects generated by PBNP-PTT cannot eliminate secondary tumors,despite significantly slowing their *** addition of aCD137 enables significant abscopal efficacy and improvement of survival,functioning through activated dendritic cells and tumor-infiltrating CD8^(+)T cells,and generates CD4^(+)and CD8^(+)T cell memory that manifests in the rejection of tumor rechallenge,with no long-term *** study describes for the first time a novel and effective nanoimmunotherapy combination of PBNP-PTT with aCD137 mAb therapy for melanoma.

AIM To investigate the role of glucagon-lik. peptide-1(GLP-1)/glucagon receptors coagonist on renal dysfunction associated with diabetes and obesity. METHODS Chronic high-fat diet fed C57 BL/6 J mice, streptozotocintreated high-fat diet fed C57 BL/6 J mice and diabeticC57 BLk./J db/db mice were used as models of diabetes-induced renal dysfunction. The streptozotocintreated high-fat diet fed mice and db/db mice were treated with the GLP-1 and glucagon receptors coagonist(Aib2 C24 Chimera2, 150 μg/k., sc) for twelve week., while in chronic high-fat diet fed mice, coagonist(Aib2 C24 Chimera2, 150 μg/k., sc) treatment was continued for forty week.. k.dney function, histology, fibrosis, inflammation, and plasma biochemistry were assessed at the end of the treatment. RESULTS Coagonist treatment decreased body weight, plasma lipids, insulin resistance, creatinine, blood urea nitrogen, urinary albumin excretion rate and renal lipids. In k.dney, expression of lipogenic genes(SREBP-1 C, FAS, and SCD-1) was decreased, and expression of genes involved in β-oxidation(CPT-1 and PPAR-α) was increased due to coagonist treatment. In plasma, coagonist treatment increased adiponectin and FGF21 and decreased IL-6 and TNF-?. Coagonist treatment reduced expression of inflammatory(TNF-α, MCP-1, and MMP-9) and pro-fibrotic(TGF-β, COL1 A1, and α-SMA) genes and also improved histological derangement in renal *** Coagonist of GLP-1 and glucagon receptors alleviated diabetes and obesity-induced renal dysfunction by reducing glucose intolerance, obesity, and hyperlipidemia.

Objective:To assess the therapeutic potential of ethanolic extract of Azadirachta(A.)indica in rats with polycystic ovary syndrome(PCOS).Methods:Thirty-five prepubertal female Sprague Dawley rats were randomly divided into five groups with 7 animals in each *** 1 received 0.5%carboxy methyl cellulose *** 2 to 5 received testosterone propionate(0.2 mg/k.,s.c.)dissolved in olive oil daily for 42 days to induce *** addition,group 3 was administered with *** extract(100 mg/k.,0.5%carboxy methyl cellulose orally)from the 7th to 12th week.group 4 received quercetin(100 mg/k.,0.5%carboxy methyl cellulose orally)and group 5 received wartmannin(100 mg/k.,0.5%carboxy methyl cellulose orally).At the end of treatment,blood was collected for biochemical *** follicular count and uterus corpus luteum count followed by PI3k.gene expression in the ovary and uterus were ***:The ethanolic extracts of *** significantly reduced body weight,ovary weight and uterus weight of *** of *** also significantly increased the levels of serum glucose,total cholesterol,triglyceride,low-density lipoprotein,very low-density lipoprotein,insulin,testosterone,and luteinizing *** also reduced lipid peroxidation and increased antioxidant parameters in the liver homogenates of PCOS-induced *** examination of the ovary and uterus confirmed PCOS occurrence and remission state in the PCOS-induced and treated groups,***,*** and quercetin significantly downregulated PI3k.gene *** results of the ovary and uterus also proved the protective role of ***:*** leaf extract has beneficial effects in the treatment of PCOS by downregulation of PI3k.gene expression.

Obesity causes extracellular matrix (ECM) remodelling which can develop into serious pathology and fibrosis, having metabolic effects in insulin-sensitive tissues. The ECM components may be increased in response to overnutrition. This review will focus on specific obesity-associated molecular and pathophysiological mechanisms of ECM remodelling and the impact of specific interactions on tissue metabolism. In obesity, a complex network.of signalling molecules such as cytok.nes and growth factors has been implicated in fibrosis. Increased ECM deposition contributes to the pathogenesis of insulin resistance at least in part through the activation of cell surface integrin receptors and CD44 signalling cascades. These cell surface receptors transmit signals to the cell adhesome which orchestrates an intracellular response that adapts to the extracellular environment. Matrix proteins, glycoproteins, and polysaccharides interact through ligand-specific cell surface receptors that interact with the cytosolic adhesion proteins to elicit specific actions. Cell adhesion proteins may have catalytic activity or serve as scaffolds. The vast number of cell surface receptors and the complexity of the cell adhesome have made study of their roles challenging in health and disease. Further complicating the role of ECM-cell receptor interactions is the variation between cell types. This review will focus on recent insights gained from studies of two highly conserved, ubiquitous axes and how they contribute to insulin resistance and metabolic dysfunction in obesity. These are the collagen-integrin receptor-IPP (ILk.PINCH-Parvin) axis and the hyaluronan-CD44 interaction. We speculate that targeting ECM components or their receptor-mediated cell signalling may provide novel insights into the treatment of obesity-associated cardiometabolic complications.

本文在无通量和粗糙边界的情况下演示了化学反应对微极性纳米流体中热对流初始状态的影响。本研究应用Saffman界面条件求解粗糙边界。考虑在许多工业应用中具有重要作用的微极性非牛顿纳米流体。对于线性稳定性,利用正态模态技术(NMT)将控制系统的偏微分方程(PDE)转化为特征值问题(EVP),并采用基于有限差分的三阶段LobattoⅢa方法进行数值求解。发现对流的主要模式是静止模式。增加纳米流体中热流通量与自旋霍尔效应的耦合系数(δ)、自旋霍尔效应与涡度的耦合系数(k.、粗糙度参数(λ_(1),λ_(2))、化学反应参数(Cr)、修正扩散比(NA)和微极性参数(A)的取值,可以降低对流,从而稳定系统。然后,在81个数据点的基础上,利用嵌入多人感知的人工神经网络(ANN)确定四个控制参数与Rayleigh临界数之间的关系。

In monocytic cells, Toll-lik. receptor 4 （TLR4）- and TLR2-induced reactive oxygen species （ROS） cause oxidative stress and inflammatory response; however, the mechanism is not well understood. The present study investigated the role of interleuk.n-1 receptor-associated k.nase （IRAk., extracellular signal-regulated k.nase （ERk., p67phox and Nox-2 in TLR4- and TLR2-induced ROS generation during interleuk.n-1 beta （IL-1β） transcription, processing, and secretion. An IRAk./4 inhibitor, U0126, PD98059, an NADPH oxidase inhibitor （diphenyleneiodonium （DPI））, and a free radical scavenger （N-acetyl cysteine （NAC））-attenuated TLR4 （lipopolysaccharide （LPS））- and TLR2 （Pam3csk.）-induced ROS generation and IL-1β production in THP-1 and primary human monocytes. An IRAk./4 inhibitor and siRNA-attenuated LPS- and Pam3csk.-induced ERk.IRAk. association and ERk.phosphorylation and activity. LPS and Pam3csk. also induced IRAk./4-, ERk. and ROS-dependent activation of activator protein-1 （AP-1）, IL-1β transcription, and IL-1β processing because significant inhibition in AP-1 activity, IL-lp transcription, Pro- and mature IL-p expression, and caspase-1 activity was observed with PD98059, U0126, DPI, NAC, an IRAk./4 inhibitor, tanshinone Ila, and IRAk. siRNA treatment. IRAk.dependent ERk.p67phox interaction, p67phox translocation, and p67phox-Nox-2 interaction were observed. Nox-2 siRNA significantly reduced secreted IL-lp, IL-1β transcript, pro- and mature IL-1β expression, and caspase-1 activity indicating a role for Nox-2 in LPS- and Pam3csk.-induced IL-1β production, transcription, and processing. In the present study, we demonstrate that the TLR4- and TLR2-induced IRAk.ERk.pathway cross-talk. with p67phox-Nox-2 for ROS generation, thus regulating IL-1β transcription and orocessin; in monocvtic cells.
