GB （goji berry） ha. bioa.tive components ca.a.le of reversing the meta.olic syndrome. This work investiga.ed systemic, biometric a.d meta.olic pa.a.eters of ma.e ra.s fed with sta.da.d diet （group CD） or high-ca.bohydra.e diet （group HC）. a. 90 da.s of a.e the HC group wa. subdivided： one wa. given vehicle solution （HCD） a.d the other wa. given GB extra.t （HCDGB）, for 60 da.s. The vehicle wa. a.so given to the CD group. a. 150 da.s of a.e, glucose tolera.ce test, tissue collection, pla.ma.ic determina.ions, lipid content, in situ perfusion a.d oxida.ive stress of the liver were ca.ried out. The GB supplementa.ion improved the pa.a.eters of the meta.olic syndrome ca.sed by the HC diet, including decrea.ed body weight ga.n, a.iposity index, dyslipidemia. hyperinsulinemia. Na.LD, liver oxida.ive stress a.d gluconeogenesis. Together with the diminished insulin resista.ce, these results indica.e the GB extra.t is a. importa.t a.juva.t in the trea.ment of the meta.olic syndrome.

Mice overexpressing the huma. a.olipoprotein a.o CIII a.e a.model of dyslipidemia. They become hypertxiglyceridemic, hypercholesterolemic a.d ha.e high blood levels of free fa.ty a.ids. Blood glucose is norma., but a. the liver integra.es lipid a.d ca.bohydra.e meta.olism, conditions of high inter-tissue circula.ion of energy substxa.es, such a. fa.ting, ma. revea. hepa.ic a.tera.ions of glucose meta.olism in these （CIII） mice. This hypothesis wa. explored by in situ liver perfusion in this investiga.ion. The NTG （non-txa.sgenic） a.ima.s showed liver a.d muscle glycogen content cha.ges compa.ible with the fed or fa.ted sta.e. In contra.t, glycogen in group CIII wa. much lower in the fed sta.e. The liver glucose relea.e in group CIII a.ter overnight fa.ting a.d a.rena.ine-stimula.ed wa. lower tha. in group NTG. Tota. glucose production under gluconeogenic conditions wa. not different between groups NTG a.d CIII, but glucose production from a.a.ine wa. decrea.ed in group CIII. Therefore, dyslipidemia.ca.sed by overexpression of a.oCIII in mice a.ters the liver glucose meta.olism, pa.ticula.ly compromising glycogen synthesis a.d degra.a.ion. This profile might ha.e a.verse outcomes during meta.olic cha.lenges tha. a.e more severe tha. fa.ting.

This work ha. the purpose of compa.ing some rena. pa.a.eters of Wista. ra.s ra.sed in sma.l litters either freely-fed （G3L） or subjected to 30% ca.oric restriction （G3R） with those of ra.s from norma. litters （Gg） a. the a.e of 90 da.s. The results showed tha. the rela.ive weight of the kidneys a.d urina.y flow were not significa.tly different. a.l the groups ha. GFR （glomerula. filtra.ion ra.e） a.d proteinuria.within norma. va.ues, despite group G3R ha.ing va.ues sta.istica.ly lower. However, group G3R ha. diminished glomerula. a.ea.a.d increa.ed numbers of rena. corpuscles. Group G3L did not exhibit differences compa.ed with controls （G9）. The effects of ca.oric restriction did not compromise kidney structure a.d function in 90-da.-old ra.s.

This work a.med a. investiga.ing whether the liver glucose meta.olism could ha.e a.role in a.y cha.ge of glucose homeosta.is tha. might exist in the reduced-litter ra. a. the a.e of 60 da.s. a.ditiona.ly, post-wea.ing ca.oric restriction a.d its hepa.ic effects were explored, a. this intervention is reported a. a.useful mea.ure a.a.nst obesity a.d its rela.ed disturba.ces. The a.ima.s were ra.sed in litters of nine （control, CG） or three pups during la.ta.ion. These sma.l-litter pups were FG （fed freely） or subjected to 30% RG （ca.oric restriction） a.ter wea.ing until 60 da.s of a.e. The increa.ed a.iposity induced by la.ta.iona. overfeeding wa. not reversed by ca.oric restriction. Hepa.ocyte glucose meta.olism a.d glucose tolera.ce test were not a.fected by litter size, but ca.oric restriction increa.ed liver ba.a. glucose relea.e, diminished gluconeogenesis a.d reta.ded the glycemic deca. during the insulin tolera.ce test. Liver glucose meta.olism of young a.ult ra.s wa. not a.fected by la.ta.iona. overfeeding. Up to this a.e, however, modera.e ca.oric restriction ha. a.potent influence tha. might compromise whole-body glucose homeosta.is a.d prompt to insulin resista.ce.

Ea.ly life overfeeding in the ra. ca. be experimenta.ly induced by reducing litter size. This investiga.ion a.sessed the consequences of this ma.ipula.ion on glucose meta.olism in vivo a.d in isola.ed hepa.ocytes in 150-da. old ra.s. a.ditiona.ly, a.ter body growth, the effects of ca.oric restriction a.d refeeding were tested. a.ult ra.s from control （G9） a.d reduced litters （G3L） did not differ in body a.d fa. weights, glucose tolera.ce or insulin resista.ce （insulin-induced hypoglycemia., or hepa.ocyte glucose relea.e under ba.a. or gluconeogenic conditions. Ca.oric restriction （G3R） reduced body a.d fa. weights, decrea.ed glucose deca. a.ter insulin injection a.d decrea.ed hepa.ocyte gluconeogenic glucose relea.e. Refeeding a.ter ca.oric restriction reversed these pa.a.eters to those of the freely-fed groups （G9 a.d G3L）. Ta.en together, these results suggest tha. the liver glucose meta.olism is not progra.med by la.ta.iona. overfeeding, but ra.her is responsive to the current nutritiona. condition of the a.ima..

The effects of the supplementa.ion with L-gluta.ine(GLN)or L-a.a.yl-L-gluta.ine(GDP)on the progression of the systemic a.d hepa.ic meta.olic sta.us of ra.s ha.ing untrea.ed type 1 dia.etes mellitus(T1DM)were *** Wista. dia.etic ra.s(streptozotocin,60 mg/kg)were a.lotted to four groups supplemented by ga.a.e for thirty da.s a. follows:control a.d dia.etic receiving sa.ine;dia.etic receiving GLN(248 mg/kg);a.d dia.etic receiving GDP(400 mg/kg).Body weight,pla.ma.ic pa.a.eters a.d kidney function were *** hepa.ocytes were used to a.sess gluconeogenic *** a.d kidney were used for morphologica. a.a.yses.T1DM decrea.ed the number a.d increa.ed the a.ea.of the hepa.ocytes,possibly beca.se of the observed enla.gement of glycogen *** weight,glomerula. a.ea.a.d proteinuria.increa.ed,a.d glomerula. filtra.ion ra.e decrea.ed,in non-supplemented T1DM *** a.ea.a.d proteinuria.were reversed by both *** T1DM hepa.ocytes relea.ed less glucose,which could ha.e been diverted to glycogen synthesis a.d seconda.y glycogenosis observed in T1DM;this wa. pa.tia.ly reversed by the *** results point to a.possible beneficia. effect of gluta.ine on the meta.olic a.d hepa.ic impa.rments of T1DM.
