Background:It remains unclear if the vascular and connective tissue structures of primary and metastatic tumors are intrinsically determined or whether these characteristics are defined by the host *** we examined the microanatomical steps of vasculature and connective tissue development of C38 colon carcinoma in different ***:Tumors produced in mice at five different locations(the cecal wall,skin,liver,lung,and brain)were ana-lyzed using fluorescent immunohistochemistry,electron microscopy and quantitative real-time polymerase chain ***:We found that in the cecal wall,skin,liver,and lung,resident fibroblasts differentiate into collagenous matrix-producing myofibroblasts at the tumor *** activated fibroblasts together with the produced matrix were incorporated by the *** connective tissue development culminated in the appearance of intratumoral tissue columns(centrally located single microvessels embedded in connective tissue and smooth muscle actin-expressing myofibroblasts surrounded by basement membrane).Conversely,in the brain(which lacks fibroblasts),C38 metasta-ses only induced the development of vascularized desmoplastic tissue columns when the growing tumor reached the fibroblast-containing ***:Our data suggest that the desmoplastic host tissue response is induced by tumor-derived fibrogenic molecules acting on host tissue *** concluded that not only the host tissue characteristics but also the tumor-derived fibrogenic signals determine the vascular and connective tissue structure of tumors.

背景与目的关于原发和转移性肿瘤的血管和结缔组织结构是由本身决定还是由宿主组织决定,目前尚不清楚。因此,我们研究了不同组织中C38结直肠癌血管和结缔组织发展的显微解剖过程。方法采用免疫荧光染色、电镜和实时定量聚合酶链反应等方法对小鼠盲肠壁、皮肤、肝脏、肺和大脑5个不同部位生成的肿瘤进行分析。结果我们发现在盲肠壁、皮肤、肝脏和肺的成纤维细胞分化成胶原蛋白基质,在肿瘤周围产生肌成纤维细胞。这些活化的成纤维细胞和生成的基质被肿瘤吸收利用。结缔组织的发展最终引起瘤内组织柱的出现(位于中心位置的单个微血管包埋在结缔组织和表达平滑肌肌动蛋白的肌成纤维细胞中,最外围由基底膜包围)。相反,在缺乏成纤维细胞的大脑中,仅当增生的肿瘤接触到含有成纤维细胞的脑膜时,C38转移才能诱导血管生成的促结缔组织增生性纤维柱的形成。结论我们的数据表明,作用于宿主组织成纤维细胞的肿瘤源性纤维生成分子诱导了促结缔组织增生的宿主组织反应。我们认为宿主组织特征和肿瘤来源的纤维生成信号共同决定了肿瘤的血管和结缔组织的结构。

Association between autoimmune pancreatitis and systemic autoimmune diseases

AIM: To investigate the association between autoimmune pancreatitis (AIP) and systemic autoimmune diseases (SAIDs) by measurement of serum immunoglobulin G4 (IgG4). METHODS: The serum level of IgG4 was measured in 61 patients with SAIDs of different types who had not yet participated in glucocorticosteroid treatment. Patients with an elevated IgG4 level were examined by abdominal ultrasonography (US) and, in some cases, by computer tomography (CT). RESUlTS: Elevated serum IgG4 levels (919 ± 996 mg/l) were detected in 17 (28%) of the 61 SAID patients. 10 patients had Sj gren's syndrome (SS) (IgG4: 590 ± 232 mg/l), 2 of them in association with Hashimoto's thyroiditis, and 7 patients (IgG4: 1388 ± 985.5 mg/l) had systemic lupus erythematosus (SlE). The IgG4 level in the SlE patients and that in patients with SS were not significantly different from that in AIP patients (783 ± 522 mg/l). Abdominal US and CT did not reveal any characteristic features of AIP among the SAID patients with an elevated IgG4 level. CONClUSION: The serum IgG4 level may be elevated in SAIDs without the presence of AIP. The determination of serum IgG4 does not seem to be suitable for the differentiation between IgG4-related diseases and SAIDs.

Characterization of functional biliary pain and dyspeptic symptoms in patients with sphincter of Oddi dysfunction:Effect of papillotomy

AIM： To characterize functional biliary pain and other gastrointestinal （GI） symptoms in postcholecystectomy syndrome （PCS） patients with and without sphincter of Oddi dysfunction （SOD） proved by endoscopic sphincter of Oddi manometry （ESOM）, and to assess the postendoscopic sphincterotomy （EST） outcome. METHODS： We prospectively investigated 85 cholecystectomized patients referred for ERCP because of PCS and suspected SOD. On admission, all patients completed our questionnaire. Physical examination, laboratory tests, abdominal ultrasound, quantitative hepatobiliary scintigraphy （QHBS）, and ERCP were performed in all patients. Based on clinical and ERCP findings 15 patients had unexpected bile duct stone disease and 15 patients had SOD biliary type Ⅰ. ESOM demonstrated an elevated basal pressure in 25 patients with SOD biliary-type Ⅲ. In the remaining 30 cholecystectomized patients without SOD, the liver function tests, ERCP, QHBS and ESOM were all normal. As a control group, 30 ‘asymptomatic＇ cholecystectomized volunteers （attended to our hospital for general cardiovascular screening） completed our questionnaire, which is consisted of 50 separate questions on GI symptoms and abdominal pain characteristics. Severity of the abdominal pain （frequency and intensity） was assessed with a visual analogue scale （VAS）. In 40 of 80 patients having definite SOD （i.e. patients with SOD biliary type Ⅰ and those with elevated SO basal pressure on ESOM）, an EST was performed just afl：er ERCP. In these patients repeated questionnaires were filled at each follow-up visit （at 3 and 6 too） and a second look QHBS was performed 3 mo after the EST to assess the functional response to EST. RESUlTS： The analysis of characteristics of the abdominal pain demonstrated that patients with common bile duct stone and definite SOD had a significantly higher score of symptomatic agreement with previously determined biliary-like pain features than patient groups of PCS without SOD and controls. The potential application of red mud and soil mixture as additive to the surface layer of a landfill cover system

Red mud, the by-product of aluminum production, has been regarded as a problematic residue all over the world. Its storage involves risks as evidenced by the Ajka red mud spill,an accident in Hungary where the slurry broke free, flooding the surrounding areas. As an immediate remediation measure more than 5 cm thick red mud layer was removed from the flooded soil surface. The removed red mud and soil mixture（RMSM） was transferred into the reservoirs for storage. In this paper the application of RMSM is evaluated in a field study aiming at re-utilizing waste, decreasing cost of waste disposal and providing a value-added product. The purpose was to investigate the applicability of RMSM as surface layer component of landfill cover systems. The field study was carried out in two steps： in lysimeters and in field plots. The RMSM was mixed at ratios ranging between 0 and 50% w/w with low quality subsoil（lQS） originally used as surface layer of an interim landfill cover. The characteristics of the lQS ＋ RMSM mixtures compared to the subsoil（lQS） and the RMSM were determined by physical–chemical, biological and ecotoxicological methods. The addition of RMSM to the subsoil（lQS） at up to 20% did not result any ecotoxic effect, but it increased the water holding capacity. In addition, the microbial substrate utilization became about triple of subsoil（lQS） after 10 months. According to our results the RMSM mixed into subsoil（lQS） at20% w/w dose may be applied as surface layer of landfill cover systems.

Climate Change Impacts on Environmental Hazards on the Great Hungarian Plain, Carpathian Basin

The potential impacts of climate change on the Great Hungarian Plain based on two regional climate models,REMO and AlADIN,were analyzed using indicators for environmental *** the climate parameters(temperature,precipitation,and wind)will change in the two investigated periods(2021–2050 and 2071–2100),their influences on drought,wind erosion,and inland excess water hazards are modeled by simple predictive *** hazards on arable lands will increasingly affect the productivity of agriculture compared to the reference period(1961–1990).The models predict an increase between 12.3%(REMO)and 20%(AlADIN)in the first period,and between 35.6%(REMO)and 45.2%(AlADIN)in the second *** increase of wind erosion hazards is not as obvious(+15%for the first period in the REMO model).Inland excess water hazards are expected to be slightly reduced(-4 to 0%)by both model predictions in the two periods without showing a clear tendency on *** three indicators together give a first regional picture of potential hazards of climate *** predictive model and data combinations of the regional climate change models and the hazard assessment models provide insights into regional and subregional impacts of climate change and will be useful in planning and land management activities.

Changes of plasma fasting carnitine ester profile in patients with ulcerative colitis

AIM： To determine the plasma carnitine ester profile in adult patients with ulcerative culitis （UC） and compared with healthy control subjects. METHOD： Using ESI triple quadrupole tandem mass spectrometry, the carnitine ester profile was measured in 44 patients with UC and 44 age- and sex-matched healthy controls. RESUlTS： There was no significant difference in the fasting free carnitine level between the patients with UC and the healthy controls. The fasting propionyl- （0.331 ± 0.019 vs 0.392 ± 0.017 μmol/l）, butyryl- （0.219 ± 0.014 vs 0.265 ± 0.012）, and isovalerylcarniUne （0.111 ± 0.008 vs 0.134 ± 0.008） levels were decreased in the UC patients. By contrast, the level of octanoyl- （0.147 ± 0.009 vs 0.114 ± 0.008）, decanoyl- （0.180 ± 0.012 vs 0.137 ± 0.008）, myristoyl- （0.048 ± 0.003 vs 0.039 ± 0.003）, palmitoyl- （0.128 ± 0.006 vs 0.109 ± 0.004）, palmitoleyl- （0.042±0.003 vs 0.031 ± 0.002） and oleylcarnitine （0.183 ± 0.007 vs 0.163 ± 0.007; P 〈 0.05 in all comparisons） were increased in the patients with UC. CONClUSION： Our data suggest selective involvement of the carnitine esters in UC patients, probably due to their altered metabolism.

Bone disorders in experimentally induced liver disease in growing rats

AIM： To investigate the change of bone parameters in a new model of experimentally induced liver cirrhosis and hepatocellular carcinoma （HCC） in growing rats. METHODS： Fischer-344 rats （n = 55） were used. Carbon tetrachloride （CCh）, phenobarbital （PB）, and a single diethylnitrosamine （DEN） injection were used. Animals were killed at wk 8 and 16. Bone mineral content, femoral length, cortical index （quotient of cortical thickness and whole diameter） and ultimate bending load （Fmax） of the femora were determined. The results in animals treated with DEN＋PB＋CCh （DPC, n = 21） were com- pared to those in untreated animals （UNT, n = 14） and in control group treated only with DEN＋PB （DP, n = 20）. RESUlTS： Fatty liver and cirrhosis developed in each DPC-treated rat at wk 8 and HCC was presented at wk 16. No skeletal changes were found in this group at wk 8, but each parameter was lower （P〈0.05 for each） at wk 16 in comparison to the control group. Neither fatty liver nor cirrhosis was observed in DP-treated animals at any time point. Femoral length and Fmax values were higher （P〈0.05 for both） in DP-treated animals at wk 8 compared to the UNT controls. However, no difference was found at wk 16. CONClUSION： Experimental liver cirrhosis and HCC are accompanied with inhibited skeletal growth, reduced bone mass, and decreased mechanical resistance in growing rats. Our results are in concordance withthe data of other studies using different animal models. A novel finding is the transiently accelerated skeletal growth and bone strength after a 8-wk long phenobarbital treatment following diethylnitrosamine injection.

Novel Method to Monitor the De-Swelling of PNIPAAm Hydrogels of Different Cross-link Density

The effect of cross-linker (methylene-bis-acrylamide) (MBA) on the volume phase transition, mechanical properties and de-swelling of Poly(N-isopropyl acrylamide-co-methylene-bis-acrylamide) hydrogel (PNIPAAm/MBA hydrogel) was investigated. A new method, namely isothermal thermo-gravimetry was developed for monitoring de-swelling of PNIPAAm/MBA hydrogel. Monomer/ Cross-linker ratio of the initial monomer composition R = moleNIPAAm/moleMBA was introduced. It has been proven earlier that initial monomer composition is close to the copolymer composition;hence R values may be used to express cross-link density. Hydrogels from R10 to R150 were investigated. The results of DSC analysis revealed that the less the cross-linker ratio in the gel (from R10 to R150) the more sharp the temperature range of volume phase transition and the higher its enthalpy. Cross-link density, namely increasing cross-linker content in the copolymer (R from 150 to 10) does not significantly affect the temperature range of volume phase transition. It sets on at 33°C - 34°C, and ends between 35?C and 38?C. Cross-link density has significant effect on compression modulus. By decreasing the ratio of cross-linker (by increasing R from 10 to 150), the compression modulus increases, goes through a maximum, and then decreases. The highest compression modulus was measured for PNIPAAm/MBA hydrogel R20. Hydrogels with cross-linker content between R100 and 30 are strong enough and have their thermoresponsivity. Isothermal thermograms of de-swelling are of similar character for all the gels with different cross-linker content. During the initial stage of de-swelling for gels with higher cross-linker content (R10 - R15) the solute release is quicker than for gels R20 - 150 and the thermograms are drawn out. Antioxidant potential,tannin and polyphenol contents of seed and pericarp of three Coffea species

Objective:To investigate the antioxidant activity,total phenolic and total tannin content of the pericarp and the seed of Coffea benghalensis(***) and Coffea liberica compared to Coffea arabica(***).Methods:The antioxidant potential,total tannin and polyphenol contents of the immature and mature seed and pericarp of *** and Coffea liberica were quantified and compared to *** chemiluminescence(ECl).2,2-diphenyl-1-picrylhydruzyl(DPPH).oxygen radical absorbance capacity,Folin-Ciocaltcau method and total tannin content assays were ***:Trolox equivalent(TE/g plant material) values obtained by ECl and DPPH methods showed loose correlation(r^2=0.587)while those measured by oxygen radical absorbance capacity assay were higher without correlation in each plant.A closer correlation was detected between the ECl method and the percentage antioxidant activity of the DPPH technique(r^2=0.610 7) in each species,however the immature pericarp of *** showed much higher DPPH scavenging potential than was seen in the ECl *** immature pericarp of *** expressed the highest tannin and polyphenol content,and a high polyphenol level was also detected in the immature seed of *** immature pericarp of Bengal and liberian coffees showed the largest amount of phenolic ***:The obtained data highlight the potential role of *** as a new source of natural antioxidants and polyphenols compared to ***.
