Proline as a biochemical marker in relation to the ecology of two halophytic Juncus species

Aims osmolytes,used for maintaining osmotic balance and as‘osmoprotectants’,are synthesized in plants as a general,con-served response to abiotic stress,although their contribution to stress-tolerance mechanisms remains ***,the most common osmolyte,accumulates in many plant species in parallel with increased external salinity and is considered a reliable bio-chemical marker of salt *** have measured proline levels in two halophytic,closely related Juncus species under laboratory and field conditions to assess the possible relevance of proline biosynthesis for salt tolerance and therefore for the ecology of these two *** Proline was quantified in plants treated with increasing NaCl con-centrations and in plants sampled in two salt marshes located in the provinces of Valencia and alicante,respectively,in southeast *** conductivity,pH,Na+and Cl−concentrations were measured in soil samples collected in parallel with the plant *** Findings Treatment with NaCl inhibited growth of *** plants in a concentration-dependent manner,but only under high salt conditions for *** treatments led to proline accumulation in both species,especially in the more salt-tolerant *** results,obtained under laboratory conditions,were confirmed in plants sam-pled in the *** all the samplings,proline contents were signifi-cantly lower in *** than in the more tolerant *** growing in the same *** direct correlation between soil salinity and proline levels could be established,but seasonal variations were detected,with increased proline contents under accentuated water deficit *** results suggest that proline biosynthesis is not only an induced,general response to salt stress but also an important contributing factor in the physiological mechanisms of salt tolerance in Juncus,and that it therefore correlates with the ecology of both species.

Cutting-edge technologies are extremely useful to develop new workflows in studying ecological data,particularly to understand animal behavior and movement trajectories at the individual *** parental care is a well-studied phenomenon,most studies have been focused on direct observational or video recording data,as well as experimental ***,what happens out of our sight still remains *** high-frequency GPS/GSM dataloggers and tri-axial accelerometers we monitored 25 Bonelli’s eagles Aquila fasciata during the breeding season to understand parental activities from a broader *** used recursive data,measured as number of visits and residence time,to reveal nest attendance patterns of biparental care with role specialization between *** data interpreted as the overall dynamic body acceleration,a proxy of energy expenditure,showed strong differences in parental effort throughout the breeding season and between ***,males increased substantially their energetic requirements,due to the increased workload,while females spent most of the time on the ***,during critical phases of the breeding season,a low percentage of suitable hunting spots in eagles’territories led them to increase their ranging behavior in order to find food,with important consequences in energy consumption and mortality *** results highlight the crucial role of males in raptor species exhibiting biparental ***,we exemplify how biologging technologies are an adequate and objective method to study parental care in raptors as well as to get deeper insight into breeding ecology of birds in general.

Sub-1 GHz Network-Based Wireless Bridge-Monitoring System:Feature and Verification

Traditional bridge monitoring systems often require wired connections between sensors,a data acquisition system,and data *** use of extension wires,conduits,and other costly accessories can dramatically increase the total cost of bridge *** the development of wireless technologies and the notable cost benefits,many researchers have been integrating wireless networks into bridge monitoring *** this study,a wireless bridge monitoring system has been developed based on the Sub-1 GHz *** main functional components of this system include sensors,wireless nodes,gateway and data *** nodes can convert analog signals obtained from the sensors to digital signals,then transmit the collected data to the gateway using the Sub1 GHz *** gateway receives and sorts data from different wireless nodes and then forwards these data to the data center *** collected data are processed in the data center using the data processing software developed in this *** order to validate the performance of the wireless system developed in this study,a steel girder bridge was monitored in the field during the concrete deck *** field results were also compared with the theoretical values obtained from finite element models to ensure the accuracy and reliability of the wireless *** results indicate that the wireless bridge monitoring system developed in this study is effective and *** Sub-1 GHz network can be a better solution for bridges with complicated site conditions because of the extended data transmission *** the power consumption can be controlled by using low-power consumption components,including the power control in software design can also dramatically reduce the system’s power consumption.

New perspectives in occult hepatitis C virus infection

Occult hepatitis C virus (HCV) infection, defined as the presence of HCV RNA in liver and in peripheral blood mononuclear cells (PBMCs) in the absence of detectable viral RNA in serum by standard assays, can be found in anti-HCV positive patients with normal serum levels of liver enzymes and in anti-HCV negative patients with persistently elevated liver enzymes of unknown etiology. Occult HCV infection is distributed worldwide and all HCV genotypes seem to be involved in this infection. Occult hepatitis C has been found not only in anti-HCV positive subjects with normal values of liver enzymes or in chronic hepatitis of unknown origin but also in several groups at risk for HCV infection such as hemodialysis patients or family members of patients with occult HCV. This occult infection has been reported also in healthy populations without evidence of liver disease. Occult HCV infection seems to be less aggressive than chronic hepatitis C although patients affected by occult HCV may develop liver cirrhosis and even hepatocellular carcinoma. Thus, anti-HCV negative patients with occult HCV may benefit from antiviral therapy with pegylated-interferon plus ribavirin. The persistence of very low levels of HCV RNA in serum and in PBMCs, along with the maintenance of specific T-cell responses against HCV-antigens observed during a long-term follow-up of patients with occult hepatitis C, indicate that occult HCV is a persistent infection that is not spontaneously eradicated. This is an updated report on diagnosis, epidemiology and clinical implications of occult HCV with special emphasis on anti-HCV negative cases.

Prefoldins Negatively Regulate Cold Acclimation in Arabidopsis thaliana by Promoting Nuclear Proteasome-Mediated HY5 Degradation

The process of cold acclimation is an important adaptive response whereby many plants from temperate regions increase their freezing tolerance after being exposed to low non-freezing temperatures. The correct development of this response relies on proper accumulation of a number of transcription factors that regulate expression patterns of cold-responsive genes. Multiple studies have revealed a variety of molecular mechanisms involved in promoting the accumulation of these transcription factors. Interestingly, however, the mechanisms implicated in controlling such accumulation to ensure their adequate levels remain largely unknown. In this work, we demonstrate that prefoldins （PFDs） control the levels of HY5, an Arabidopsis transcription factor with a key role in cold acclimation by activating anthocyanin biosynthesis, in response to low temperature. Our results show that, under cold conditions, PFDs accumulate into the nucleus through a DELLA-dependent mechanism, where they interact with HY5, triggering its ubiquitination and subsequent degradation. The degradation of HY5 would result, in turn, in anthocyanin biosynthesis attenuation, ensuring the accurate development of cold acclimation. These findings uncover an unanticipated nuclear function for PFDs in plant responses to abiotic stresses.

In educational environments an improvement in the quality of interior lighting has a direct benefit in increasing productivity and alertness of students and teachers, as well as very important implications for the energy efficiency of the education facilities but when a replacement with different lighting is analyzed, research may be tarnished by users’ pre-formed opinions, influenced by manufacturers and advertising. Consequently, it is necessary to understand the users’ point of view, even before of being subjected to any change in the lighting *** on the Kansei Engineering framework, the general objective of this paper is to evaluate and compare the subjective evaluation of students’ pre-formed opinions to lighting provided by two types of lamps (fluorescent and LED). The subjective assessment of 427 university students has been compared over four years. The results show significant differences in students’ subjective evaluation. This finding highlights the existence of symbolic or functional attributes of the usefulness perceived by the student that could influence on investigations in which different types of lighting are compared.

Role of 9-Lipoxygenase and α-Dioxygenase Oxylipin Pathways as Modulators of Local and Systemic Defense

Plant 9-1ipoxygenases （9-LOX） and α-dioxygenases （α-DOX） initiate the synthesis of oxylipins after bacterial infection. Here, the role of these enzymes in plants＇ defense was investigated using individual Arabidopsis thaliana Ioxl and doxl mutants and a double Ioxl doxl mutant. Studies with Pseudomonas syringae pv. tomato （Pst） revealed the enhanced susceptibility of Ioxl to the virulent strain Pst DC3000 and the partial impairment of Ioxl and doxl mutants to activate systemic acquired resistance. Notably, both defects were enhanced in the Ioxl doxl plants as compared with individual mutants. We found that pre-treatment with 9-LOX- and α-DOX-generated oxylipins protected plant tissues against bacterial infection. The strongest effect in this respect was exerted by 9-ketooctadecatrienoic acid （9-KOT）, which is produced from linolenic acid by 9-LOX. Quantification of 9-KOT revealed its accumulation after bacterial infection. The levels were reduced in Ioxl and Ioxl doxl plants but strongly increased in the doxl mutant due to metabolic interaction of the two pathways. Transcriptional analyses indicated that 9-KOT pre-treatment modifies hormone homeostasis during bacterial infection. The nature of the changes detected suggested that 9-KOT interferes with the hormonal changes caused by bacterial effectors. This notion was substantiated by the finding that 9-KOT failed to reduce the growth of PstDC3OOOhrpA, a mutant compromised in effector secretion, and of the avirulent strain Pst DC3000 avrRpml. Further support for the action of the 9-LOX- and α-DOX-oxylipin pathways as modulators of hormone homeostasis was the ob- servation that Ioxl doxl seedlings are hypersensitive to the growth-inhibitory effect of ABA and showed enhanced ac- tivation of ABA-inducible marker genes as compared with wild-type plants.

In this work three fuel consumption and exhaust emission models,ADVISOR,VT-MICRO and the European Environmental Agency Emission factors,have been used to obtain fuel consumption(FC)and exhaust *** models have been used at micro-scale,using the two signal treatment methods *** manuscript presents:1)a methodology to collect data in real urban driving cycles,2)an estimation of FC and tailpipe emissions using some available models in literature,and 3)a novel analysis of the results based on delivered wheel *** results include Fuel Consumption(FC),CO_(2),NO_(x) and PM_(10) emissions,which are derived from the three *** the first part of the paper we present a new procedure for incomplete drive cycle data treatment,which is necessary for real drive cycle acquisition in high density *** the models are used to obtain second by second FC and exhaust ***,a new methodology named Cycle Analysis by Ordered Power(CAbOP)is presented and used to compare the *** method consists in the re-ordering of time dependant data,considering the wheel mechanical power domain instead of the standard time *** new strategy allows the 5 situations in drive cycles to be clearly visualized:hard breaking zone,slowdowns,idle or stop zone,sustained speed zone and acceleration *** complete methodology is applied in two real drive cycles surveyed in Barcelona(Spain)and the results are compared with a standardized WLTC urban cycle.

Use of micellar liquid chromatography for rapid monitoring of fungicides post harvest applied to citrus wastewater

A method based on micellar liquid chromatography has been developed to simultaneously monitor four pesticides largely post-harvest applied to citrus：thiabendazole,pyrimethanil,o-phenylphenol and *** samples were filtered and directly injected without other treatment,thus avoiding extraction *** composition of the mobile phase was optimized using a chemometrical approach to achieve and excellent resolution to 0.07 mol/L SDS/5%,V/V 1-pentanol buffered at p H *** phase run through a C18 column at 1 m L/min at room *** detection was performing by UV–Visible absorbance using a wavelength program：0–10 min,305 nm（for thiabendazole）;10–12;265 nm（for pyrimethanil）and 12–18,220 nm（o-phenylphenol and imazalil）.The developed method was validated following the guidelines of the US Environmental Protection Agency in terms of：quantitation range,（0.5–4 to 15μg/m L）,linearity（r2〉0.9995）,sensitivity（LOD,0.18–1.4μg/m L）,precision（〈9.2%）,trueness（93.9%–103.7%）,and ruggedness（〈9.9%）.It was found that the fungicides remain up to eight days in surface water at outdoor *** method was used to screen the presence of the analytes in several waste water samples,and was proved to be useful in routine analysis.
