Oxidative stress, cardiolipin and mitochondrial dysfunction in nonalcoholic fatty liver disease

Nonalcoholic fatty liver disease(NAFLD) is today considered the most common form of chronic liver disease, affecting a high proportion of the population worldwide. NAFLD encompasses a large spectrum of liver damage, ranging from simple steatosis to steatohepatitis, advanced fibrosis and cirrhosis. Obesity, hyperglycemia, type 2 diabetes and hypertriglyceridemia are the most important risk factors. The pathogenesis of NAFLD and its progression to fibrosis and chronic liver disease is still unknown. Accumulating evidence indicates that mitochondrial dysfunction plays a key role in the physiopathology of NAFLD, although the mechanisms underlying this dysfunction are still unclear. Oxidative stress is considered an important factor in producing lethal hepatocyte injury associated with NAFLD. mitochondrial respiratory chain is the main subcellular source of reactive oxygen species(ROS), which may damage mitochondrial proteins, lipids and mitochondrial DNA. Cardiolipin, a phospholipid located at the level of the inner mitochondrial membrane, plays an important role in several reactions and processes involved in mitochondrial bioenergetics as well as in mitochondrial dependent steps of apoptosis. This phospholipid is particularly susceptible to ROS attack. Cardiolipin peroxidation has been associated with mitochondrial dysfunction in multiple tissues in several physiopathological conditions, including NAFLD. In this review, we focus on the potential roles played by oxidative stress and cardiolipin alterations in mitochondrial dysfunction associated with NAFLD.

Influence of Surface Finishing on High-Temperature Oxidation of AISI Type 444 Ferritic Stainless Steel Used in SOFC Stacks

This research is focused on the study of the samples, approximatively 15 × 30 mm^2 sized, that were mechanically cut from two sheets （0.4 and 0.2 mm thick, respectively） of AIS1444 Type ferritic stainless steel （FSS）（DIN 1.4521, Eu designation X2CrmoTil8-2）; this material was in the ＇as-rolled＇ state. Part of these specimens were treated superficially on one side using abrasive SiC papers with different average grit sizes （i.e., 46.2, 30.2, 18.3 gm） and diamond suspension （up to 1 tim） in order to obtain various surface roughness. Both the ＇as-rolled＇ and superficially treated samples were then aged in a muffle fumace in static air according to a thermal cycle corresponding to the curing phase of an experimental glass used as sealing in the solid oxide fuel cell stacks. After aging, the chemical compositions and mor- phological peculiarities of the scale formed depending on the thickness of the samples and their surface state were studied by scanning electron microscopy, energy-dispersive spectroscopy, micro-Raman spectroscopy, bright field light optical microscopy. The obtained results show that all scales formed consist of an inner Cr2O3 subscale and an outer （mn,Cr）3O4 spinel layer; the relationship between FSS grain size and scale microstructural features is consistent on the samples with mirror-like surface only; the scale thicknesses on SiC grinded samples are comparable; the scales covering the ＇as-rolled＇ surfaces are morphologically similar to those grown on the surfaces finished with the 30.2 and 18.3 μm SiC papers, and their thicknesses show an intermediate situation between the abraded and the mirror-like specimens. The last described characteristics depend mainly on the surface and microstructural peculiarities resulting from the rolling process.

Plasma synthesis of various polymorphs of tungsten trioxide nanoparticles using gliding electric discharge in humid air:characterization and photocatalytic properties

Pupal emergence pattern in cactophilic Drosophila and the effect of host plants

Aquaporins are water channel proteins that mediate the fine-tuning of cell membrane water permeability during development or in response to environmental stresses. The present work focuses on the oxidative stress-induced redistribution of plasma membrane intrinsic protein （PIP） aquaporins from the plasma membrane （Pm） to intracellular membranes. This process was investigated in the Arabidopsis root. Su- crose density gradient centrifugation showed that exposure of roots to 0.5 mm H2O2 induces significant depletion in Pm fractions of several abundant PIP homologs after 15 rain. Analyses by single-particle tracking and fluorescence correlative spectroscopy showed that, in the Pm of epidermal cells, H2O2 treat- ment induces an increase in lateral motion and a reduction in the density of a fluorescently tagged form of the prototypal AtPIP2;1 isoform, respectively. Co-expression analyses of AtPIP2;1 with endomembrane markers revealed that H2O2 triggers AtPIP2;1 accumulation in the late endosomal compartments. Life- time analyses established that the high stability of PIPs was maintained under oxidative stress conditions, suggesting that H2O2 triggers a mechanism for intracellular sequestration of Pm aquaporins without further degradation. In addition to information on cellular regulation of aquaporins, this study provides novel and complementary insights into the dynamic remodeling of plant internal membranes during oxida- tive stress responses.

Evidence for a con served microbiota across the different developmental stages of Plodia interpunctella

Diversity and composition of lepidopteran microbiotas are poorly investigated, especially across the different developmental stages. To improve this knowledge, we characterize the microbiota among different developmental stages of the Indian meal moth, Plodia interpunctella, which is considered one of the major pest of commodities world-wide. Using culture-independent approach based on Illumina 16S rRNA gene sequencing we characterized the microbiota of four developmental stages: eggs, first-, and last-instar larvae, and adult. A total of 1022 bacterial OTUs were obtained, showing a quite diversified microbiota associated to all the analyzed stages. The microbiotas associated with P. interpunctella resulted almost constant throughout the developmental stages, with approximately 77% of bacterial OTUs belonging to the phylum of Proteobacteria. The dominant bacterial genus is represented by Burkholderia (?64%), followed by Propionibacterium, Delftia, Pseudomonas, and Stenotrophomonas. A core bacterial community, composed of 139 OTUs, was detected in all the developmental stages, among which 112 OTUs were assigned to the genus Burkholderia. A phylogenetic reconstruction, based on the 16S rRNA, revealed that our Burkholderia OTUs clustered with Burkholderia cepacia complex, in the same group of those isolated from the hemipterans Gossyparia spuria and Acanthococcus aceris. The functional profiling, predicted on the base of the bacterial 16S rRNA, indicates differences in the metabolic pathways related to metabolism of amino acids between preimaginal and adult stages. We can hypothesize that bacteria may support the insect host during preimaginal stages.

Natural history of Valentin’s rock lizard(Darevskia valentini) in Armenia

Valentin's rock lizard (Darevskia valentini) is suggested to be the pare nt for several parthenoge netic species (e.g., D. armeniaca, D. bendimahiensis, D. sapphirina, and D. unisexualis) that evolved through hybridization. Complex evolutionary processes (ineluding reticulate evolution) are occurring within the areas where Valentin' s rock lizard coexists with these and other rock lizards. Hence, a detailed biological specification of this species is important for understanding how vertebrates evolve. Valentin's rock lizard is a longlived (up to 9 years), small diurnal lizard with larger females than males, which is unlike other species of the genus. Their relatively large eggs and early reproducti on period, which occurs just after emerge nee from winter shelters, are adaptatio ns for living in a high elevation climate (higher than 2 000 m a.s.l.). Their body temperatures (31 -32℃) are comparable to body temperatures of rock lizards living in milder climates, though female body temperature is more depe ndent on substrate temperature and basking due to their lower activity than that found in males. Population density fluctuates from several individuals to several hundred per hectare and is not affected by parthenogen coexistenee, although hybrids do occur in sexually biased populations where males are more common than females. The male home range is larger than that of females, though these home ranges broadly overlap. Prey is not limited in the mountain meadows and Valentin's rock lizards feed on a great variety of arthropods. Infan ticide occurs in high-de nsity populati ons.
