3D culturing of human adipose-derived stem cells enhances their pluripotency and differentiation abilities

Human mesenchymal stem cells,such as human adipose-derived stem cells(hASCs),are typically cultured on a two-dimensional(2 D)monolayer material surface,on which 2 D culturing methods are easily performed and ***,hASCs usually suffer from decreased pluripotency and differentiation ability when cultured with a 2 D monolayer culturing method compared to hASCs cultured with a three-dimensional(3 D)culturing method,such as suspension *** this study,we evaluated whether the pluripotency and differentiation ability of hASCs can be reversibly changed during sequential cultivation with 2 D and 3 D culturing *** hASCs cultivated with a 3 D culturing process after 2 D culture showed at least 2-fold enhanced pluripotency(Sox2,Nanog,and OCT4)compared with that of hASCs cultured with the 2 D culture process ***,hASCs obtained from the 3 D culture process expressed increased levels of differentiation markers of chondrocytes and osteoblasts compared with hASCs obtained from the 2 D culture process when hASCs were induced to ***,their pluripotency and differentiation ability were extensively reduced when hASCs were shifted from 3 D culture to 2 D culture and vice versa,which indicates that hASCs show reversibility in terms of their pluripotency and differentiation ability depending on their environment in 2 D and 3 D *** reversibility of pluripotency and differentiation ability were found to last for at least 5 passages in culture during the alternative and sequential culture of cells with 2 D and 3 D culturing *** study revealed the importance of the culture microenvironment in maintaining the pluripotency and differentiation ability of hASCs,which may reduce the effects of the aging process in *** discuss whether the environment of stem cell culture(i.e.,2 D or 3 D cultivation)can affect stem cell fate in terms of pluripotency and differentiation reversibility.

Aim： To assess the efficacy and safety of tadalafil in comparison to a placebo, when taken on demand for 12 weeks by East/Southeast Asian men with erectile dysfunction （ED）, Methods： This multicenter, randomized, double-blind, parallel group, placebo-controlled study was conducted at 17 centers across East and Southeast Asia between August 2002 and February 2003. Men more than 18 years of age with mild to severe ED of various etiologies were randomized to receive a placebo or 20 mg of tadalafil taken as needed （maximum once daily）. Efficacy assessments included the International Index of Erectile Function, the Sexual Encounter Profile diary and Global Assessment Questions. Results： Tadalafil significantly improved erectile function as compared to the placebo （P 〈 0.001）. At the endpoint, the patients receiving 20 mg of tadalafil reported a greater mean per patient percentage of successful intercourse attempts （Sexual Encounter Profile question 3： 70.9% compared to 33.5% in the placebo） and a greater proportion of improved erections （Global Assessment Question： 86.2% compared to 30.1%）. Most （≥3%） treatment emergent adverse events were mild or moderate. The most common treatment emergent adverse events were headache, back pain, dizziness and dyspepsia. Conclusion： Tadalafil was an effective and well-tolerated treatment for ED in East and Southeast Asian men.

BACKGROUND Cholangiocarcinoma is a highly lethal disease that had been underestimated in the past two decades. Many risk factors are well documented for in cholangiocarcinoma, but the impacts of advanced biliary interventions, like endoscopic sphincterotomy(ES), endoscopic papillary balloon dilatation(EPBD),and cholecystectomy, are inconsistent in the previous *** To clarify the risks of cholangiocarcinoma after ES/EPBD, cholecystectomy or no intervention for cholelithiasis using the National Health Insurance Research Database(NHIRD).METHODS From data of NHIRD 2004-2011 in Taiwan, we selected 7938 cholelithiasis cases as well as 23814 control group cases(matched by sex and age in a 1:3 ratio). We compared the previous risk factors of cholangiocarcinoma and cholangiocarcinoma rate in the cholelithiasis and control groups. The incidences of total and subsequent cholangiocarcinoma were calculated in ES/EPBD patients, cholecystectomy patients, cholelithiasis patients without intervention,and groups from the normal *** In total, 537 cases underwent ES/EPBD, 1743 cases underwent cholecystectomy,and 5658 cholelithiasis cases had no intervention. Eleven(2.05%), 37(0.65%), and7(0.40%) subsequent cholangiocarcinoma cases were diagnosed in the ES/EPBD,no intervention, and cholecystectomy groups, respectively, and the odds ratio for subsequent cholangiocarcinoma was 3.13 in the ES/EPBD group and 0.61 in the cholecystectomy group when compared with the no intervention *** In conclusion, symptomatic cholelithiasis patients who undergo cholecystectomy can reduce the incidence of subsequent cholangiocarcinoma, while cholelithiasis patients who undergo ES/EPBD are at a great risk of subsequent cholangiocarcinoma according to our findings.

Background:The few previous studies that focused on the effects of compression garments(CG)on distance running performance have simultaneously measured electromyogram,physiological,and perceptual ***,this study investigated the effects of CG on muscle activation and median frequency during and after distance running,as well as blood-lactate concentration and rating of perceived exertion(RPE)during distance ***:Eight healthy male recreational runners were recruited to randomly perform two 40 min treadmill running trials,one with CG,and the other with control garment made of normal *** RPE and the surface electromyography(EMG)of 5 lower extremity muscles including gluteus maximus(GM),rectus femoris(RF),semitendinosus(ST),tibialis anterior(TA),and gastrocnemius(GAS)were measured during the running *** blood-lactate levels before and after the running trial were ***:Wearing CG led to significant lower muscle activation(p0.05).Conclusion:Wearing CG was associated with significantly lower muscle activation and higher median frequency in the running-related key muscles during distance *** finding suggested that wearing CG may improve muscle function,which might enhance running performance and prevent muscle fatigue.

Nanoseparation-lnspired Manipulation of the Synthesis of CdS Nanorods

CdS nanorods have been sorted by length using a density gradient ultracentrifuge rate separation method. The fractions containing longer rods showed relatively stronger oxygen-related surface trap emission, while the shorter ones had dominant band-edge emission. These results suggest that the final length distribution of CdS nanorods is not a result of random nucleation and growth, but is related to the local synthesis conditions. Inspired by these findings, different synthesis environments (N2, air, and O2) have been employed in order to tailor the length distribution. In addition to the rod length, the photoluminescence properties of CdS nanorods can also be manipulated. Increasing the oxygen partial pressure significantly changed the growth behavior of CdS nanorods by improving the anisotropic growth.

Platelet-rich plasma(PRP)can stimulate the proliferation of stem cells and have a positive effect on tissue *** many commercialized PRP preparation kits are already on the market,first-line clinical workers are still not satisfied with most of the PRP *** work of commercial PRP kits is based on the density of blood ***,the blood elements are very close in density which makes the separation ***,the mentioned commercialized kits are generally contaminated by leucocytes and *** this study,a home-designed PRP device was developed to use a separation membrane with adequate cut-off pore size of 5μm,3μm and 2μm for the groups of H5M,H3M,and H2M,respectively,to be placed in the middle of the centrifuge *** home-designed H2M showed a very promising results regardless of the final volume(1.82±0.09 ml),platelet yield(8.39±0.44%),Red Blood Cells(0%),White Blood Cells(0%),and Relative Concentration of Platelet Increment value(225.09%).Further,it showed a good result in cell viability and cytotoxicity and confirmed a good multilineage *** concentration in PRP prepared by group H2M was relatively stable and far above *** the fibrin fibers were linked together as bridging strands or strings and turned into an inter-connected porous structure for nutrients transportation and regenerative cell *** believe that the home-designed group H2M should have a great potential to develop into the final product to meet the requirements of first-line clinical workers.

三大主力红军会师后,国民党“围剿”红军态势不断演进,红军面临形势极其危险。在此背景下,胡宗南所部第七十八师孤军深入,被中共和红军设伏于山城堡等地。山城堡战役是国民党“围剿”红军进入关键时期的一次重要战役,此战中国民党各路军队未能协同配合是失败的主要原因,战后国共对抗不断升级,从这个意义上讲,对山城堡战役评价过高是不适当的。山城堡战役后,蒋介石、胡宗南对山城堡战役进行了深刻反思并调整了战略部署。因西安事变爆发,国共才结束了对抗,实现了国内和平。

Low-power laser irradiation inhibits arecoline-induced fibrosis:an in vitro study

Oral submucous fibrosis （OSF） is a potentially malignant disorder that is characterized by a progressive fibrosis in the oral submucosa. Arecoline, an alkaloid compound of the areca nut, is reported to be a major aetiological factor in the development of OSF. Low-power laser irradiation （LPLI） has been reported to be beneficial in fibrosis prevention in different damaged organs. The aim of this study was to investigate the potential therapeutic effects of LPLI on arecoline-induced fibrosis. Arecoline- stimulated human gingival fibroblasts （HGFs） were treated with or without LPLI. The expression levels of the fibrotic marker genes alpha-smooth muscle actin （a-SMA） and connective tissue growth factor （CTGF/CCN2） were analysed by quantitative real- time reverse transcription polymerase chain reaction （RT-PCR） and western blots. In addition, the transcriptional activity of CCN2 was further determined by a reporter assay. The results indicated that arecoline increased the messenger RNA and protein expression of CCN2 and a-SMA in HGF. Interestingly, both LPLI and forskolin, an adenylyl cyclase activator, reduced the expression of arecoline-mediated fibrotic marker genes and inhibited the transcriptional activity of CCN2. Moreover, pretreatment with SQ22536, an adenylyl cyclase inhibitor, blocked LPLI＇s inhibition of the expression of arecoline-mediated fibrotic marker genes. Our data suggest that LPLI may inhibit the expression of arecoline-mediated fibrotic marker genes via the cAMP signalling pathway.

BACKGROUND Cholangiocarcinoma is a disease with a high mortality *** previous study revealed that cholelithiasis patients who undergo endoscopic sphincterotomy(ES)/endoscopic papillary balloon dilatation are at a higher risk for subsequent cholangiocarcinoma than cholelithiasis patients who undergo *** To clarify the relationship between recurrent biliary events and subsequent cholangiocarcinoma risk in choledocholithiasis *** From one million random cases in the Taiwan National Health Insurance Research Database 2004–2011,we selected symptomatic choledocholithiasis patients older than 18 years who were admitted from January 2005 to December 2009(study group).Cases for a control group were defined as individuals who had never been diagnosed with cholelithiasis,matched by sex and age in a 1:3 *** study group was further divided into ES/endoscopic papillary balloon dilatation,both ES/endoscopic papillary balloon dilatation and cholecystectomy,and no intervention *** We included 2096 choledocholithiasis patients without previous intervention or cholangiocarcinoma.A total of 12(2.35%),11(0.74%),and 1(1.00%)subsequent cholangiocarcinoma cases were diagnosed among 511 ES/endoscopic papillary balloon dilatation patients,1485 patients with no intervention,and 100 ES/endoscopic papillary balloon dilatation and cholecystectomy patients,*** incidence rates of recurrent biliary event were 527.79/1000 person-years and 286.69/1000 person-years in the subsequent cholangiocarcinoma and no cholangiocarcinoma group,showing a high correlation between subsequent cholangiocarcinoma risk and recurrent biliary *** Choledocholithiasis patients who undergo further cholecystectomy after ES/endoscopic papillary balloon dilatation have decreased subsequent cholangiocarcinoma risk due to reduced recurrent biliary events.

Health disparities are associated with gastric cancer mortality-to-incidence ratios in 57 countries

AIM To evaluate the association between mortality-to-incidence ratios(MIRs) and health *** In this study, we used the GLOBOCAN 2012 database to obtain the cancer incidence and mortality data for 57 countries, and combined this information with the World Health Organization(WHO) rankings and total expenditures on health/gross domestic product(e/GDP). The associations between variables and MIRs were analyzed by linear regression analyses and the 57 countries were selected according to their data quality. RESULTS The more developed regions showed high gastric cancer incidence and mortality crude rates, but lower MIR values than the less developed regions(0.64 vs 0.80, respectively). Among six continents, Oceania had the lowest(0.60) and Africa had the highest(0.91) MIR. A good WHO ranking and a high e/GDP were significantly associated with low MIRs(P = 0.001 and P = 0.001, respectively).CONCLUSION The MIR variation for gastric cancer would predict regional health disparities.
