Intracellular redox potential-driven live-cell synthesis of CdSe quantum dots in Staphylococcus aureus

Biosynthesized semiconductor quantum dots(QDs) have bright fluorescence,adjustable particle sizes,and environmental friendliness,endowing them with convenience and potential for biological *** to the unclear mechanism of the cellular environment on the live-cell synthesis of QDs,it is still difficult to regulate their optical ***,the critical role of the intracellular redox environment in regulating the fluorescence properties of biosynthesized CdSe QDs in Staphylococcus aureus(***) has been *** glutathione peroxidase(GPx) activity directly affects the intracellular H_(2)O_(2) produced in the SeO_(3)^(2-) reduction,which further manipulates the glutathione redox cycle to determine the content of low-valence *** a result,the fluorescence intensity of the synthesized CdSe QDs increases by 60% in the GPx overexpressed *** cellular redox potential that is controlled by the GSH redox cycle provides the driving force for the reduction of SeO_(3)^(2-),facilitating the synthesis of CdSe QDs in *** *** proposed mechanism of the cellular redox state provides a new perspective for regulating the synthesis of semiconductor nanomaterials in live cells.

AIM:To investigate the effect of electroacupuncture(EA)on the mitochondria-dependent apoptotic signaling pathway in the ciliary muscle of guinea pigs with negative lensinduced myopia(liM).METHODS:Guinea pigs were randomly divided into normal control(NC)group,liM group,liM+SHAM acupoint(liM+SHAM)group,and liM+EA *** in the NC group received no intervention,while those in other three groups were covered with-6.0 diopter(D)lenses on right ***,animals in the liM+EA group received EA at Hegu(li4)combined with Taiyang(EX-HN5)acupoints,while those in the liM+SHAM group were treated at sham *** treatments for 1,2,and 4wk,morphological changes in ciliary muscles were observed with hematoxylin and eosin(H&E)staining and nick end labeling(tuNEL),and the expression of the mitochondrial apoptotic signaling pathway-related molecules in ciliary muscles was measured by real-time quantitative polymerase chain reaction(qPCR)and Western ***,the adenosine triphosphate(ATP)contents were also determined in ciliary ***:Axial length increased significantly in the liM and liM+SHAM groups and decreased in the liM+EA *** ciliary muscle fibers were broken and destroyed in both liM and liM+SHAM groups,whereas those in the liM+EA group improved *** assay showed the number of apoptotic cells increased in the liM and liM+SHAM groups,whereas reduced in the liM+EA *** contents showed a significant decrease in the liM and liM+SHAM groups,whereas increased after EA *** with the NC group,the dynamin-related protein 1(DRP1),Caspase3,and apoptotic protease activator 1(APAF1)levels were significantly increased in the liM group and decreased in the liM+EA ***:The results provide evidence of EA inhibiting the development of myopia by regulating the mitochondrial apoptotic signaling pathway.

Biological structural materials not only exhibit remarkable mechanical properties but also often embody dynamic characteristics such as environmental responsiveness,autonomy,and self-healing,which are difficult to achieve in conventional engineering *** merging materials science,synthetic biology,and other disciplines,engineered living materials(ELMs)provide a promising solution to combine living organisms with abiotic components,thus facilitating the construction of functional“living”*** natural materials,ELMs possess vitality and hold immense application potential in areas such as medicine,electronics,and construction,captivating increasing research attention *** an emerging branch of ELMs,structural ELMs aim to mimic living biological structural materials by achieving desired mechanical performance while maintaining important“living”*** we summarize the recent progress and provide our perspectives for this emerging research *** first summarize the superiority of structural ELMs by reviewing biological structural materials and biomimetic material design ***,we provide a systematic discussion on the definition and classifications of structural ELMs,their mechanical performance,and physiological ***,we summarize some critical challenges faced by structural ELMs and highlight directions of future *** hope this review article can provide a timely summary of the state of the art and relevant perspectives for future development of structural ELMs.

NIR-IIb-triggered photodynamic therapy combined with chemotherapy platform based on rare-earth-doped nanoparticles

Photodynamic therapy（PDT）,with high spatiotemporal selectivity,minimal side effects and less drug resistance,has emerged as a promising approach for the treatment of ***,inadequate penetration depth and poor efficacy seriously restrict its practical *** address these challenges,we developed a combined therapy platform（RENP@C/D-FA） based on rare-earth-doped nanoparticles（RENPs） with a structure of NaErF4:Tm（0.5%）@NaYF4,on which photosensitizer Ce6（chlorin e6） and chemo-therapeutic drug（DOX）(doxorubicin) were loaded by encapsulating with an amphiphilic polymer distearoyl phosphatidylethanolamine-polyethylene glycol-folic acid（DSPE-PEG-FA）.RENPs with NaErF4as the matrix possess multiple excitation/emission peaks,enabling activation of PDT under near-infrared-Ⅱb（NIR-Ⅱb,1500-1700 nm） excitation and luminescence imaging in NIR-Ⅱb region by absorbing 808 nm near-infrared ***,DOX can be selectively released in the acidic microenvironment of *** from the tumor targeting capability of folic acid,this platform can successfully ablate tumor cells and inhibit tumor growth by combining PDT and *** innovative approach holds great promise as a powerful tool for cancer treatment,showing the potential to revolutionize PDT.

Identification and characterization of camptothecin tailoring enzymes in Nothapodytes tomentosa

Camptothecin is a complex monoterpenoid indole alkaloid with remarkable antitumor *** that two C-10 modified camptothecin derivatives,topotecan and irinotecan,have been approved as potent anticancer agents,there is a critical need for methods to access other aromatic ringfunctionalized congeners(e.g.,C-9,C-10,etc.).However,contemporary methods for chemical oxidation are generally harsh and low-yielding when applied to the camptothecin scaffold,thereby limiting the development of modified *** herein,we have identified four tailoring enzymes responsible for C-9 modifications of camptothecin from Nothapodytes tomentosa,via metabolomic and transcriptomic *** consist of a cytochrome P450(Nt CPT9H)which catalyzes the regioselective oxidation of camptothecin to 9-hydroxycamptothecin,as well as two methyltransferases(Nt OMT1/2,converting 9-hydroxycamptothecin to 9-methoxycamptothecin),and a uridine diphosphate-glycosyltransferase(Nt UGT5,decorating 9-hydroxycamptothecin to9-β-D-glucosyloxycamptothecin).Importantly,the critical residues that contribute to the specific catalytic activity of Nt CPT9H have been elucidated through molecular docking and mutagenesis *** work provides a genetic basis for producing camptothecin derivatives through metabolic *** will hasten the discovery of novel C-9 modified camptothecin derivatives,with profound implications for pharmaceutical manufacture.

Converting low-grade thermal energy into electricity through ther-mogalvanic cells offers a promising approach to addressing environ-mental challenges[1].However,thermogalvanic cells face obstacles,due to limited thermopower,connected to differences in solvent-dependent entropy(ΔS)among redox anions and varying concentration ratios(ΔC)between hot and cold sides[2,3].A typical strategy to enhance the thermopower involves the improvement ofΔS of the redox ***-fortunately,ΔC tends to approach zero spontaneously due to the ther-modynamic instability and redox ion ***,the challenge is to maintain significantΔC for both redox ions and unravel the intricate modulation mechanisms[4].

SWG: an architecture for sparse weight gradient computation

On-device training for deep neural networks(DNN) has become a trend due to various user preferences and scenarios. The DNN training process consists of three phases, feedforward(FF), backpropagation(BP), and weight gradient(WG) update. WG takes about one-third of the computation in the whole training process. Current training accelerators usually ignore the special computation property of WG and process it in a way similar to FF/BP. Besides, the extensive data sparsity existing in WG, which brings opportunities to save computation, is not well explored. Nevertheless, exploiting the optimization opportunities would meet three underutilization problems, which are caused by(1) the mismatch between WG data dimensions and hardware parallelism,(2) the full sparsity, i.e., the sparsity of feature map(Fmap),error map(Emap), and gradient, and(3) the workload imbalance resulting from irregular sparsity. In this paper, we propose a specific architecture for sparse weight gradient(SWG) computation. The architecture is designed based on hierarchical unrolling and sparsity-aware(HUSA) dataflow to exploit the optimization opportunities of the special computation property and full data sparsity. In HUSA dataflow, the data dimensions are unrolled hierarchically on the hardware architecture. A valid-data trace(VDT) mechanism is embedded in the dataflow to avoid the underutilization caused by the two-sided input sparsity. The gradient is unrolled in PE to alleviate the underutilization induced by output sparsity while maintaining the data reuse opportunities. Besides, we design an intra-and inter-column balancer(IIBLC) to dynamically tackle the workload imbalance problem resulting from the irregular sparsity. Experimental results show that with HUSA dataflow exploiting the full sparsity, SWG achieves a speedup of 12.23× over state-of-the-art gradient computation architecture, Train Ware. Numerical study on the effective thermal conductivity and thermal tortuosity of porous media with different morphologies

Effective thermal conductivity and thermal tortuosity are crucial parameters for evaluating the effectiveness of heat conduction within porous *** direct pore-scale numerical simulation method is applied to investigate the heat conduction processes inside porous structures with different *** thermal conduction performances of idealized porous structures are directly compared with real foams across a wide range of *** foam structures are reconstructed using X-ray computed tomography and image processing techniques,while Kelvin and Weaire-Phelan structures are generated through periodic unit cell *** detailed temperature fields inside the porous structures are determined by solving the heat conduction equation at the pore *** results present that the equivalent thermal conductivity of Kelvin and Weaire-Phelan structures is similar to and greater than that of the real foam structure with the same strut *** thermal tortuosity of real foam structure is relatively larger and the heat conduction path becomes straighter by adopting the anisotropic *** thermal tortuosity of the fluid channels for Kelvin,Weaire-Phelan,and real foam structures is close to *** thermal conductivity of porous structures with heat transfer fluid increases as the thermal conductivity ratio of fluid to solid becomes larger.A small porosity of porous media leads to a larger equivalent thermal conductivity due to the dominant contribution of porous skeleton in the heat conduction *** derived from parallel and series models,as well as the Maxwell-Eucken models,provide decent predictions of effective thermal conductivity,with an average error of less than 8%in the entire range of thermal conductivity ratio.

Compared with electrical neural networks,optical neural networks(ONNs)have the potentials to break the limit of the bandwidth and reduce the consumption of energy,and therefore draw much attention in recent *** far,several types of ONNs have been ***,the current ONNs cannot realize the acceleration as powerful as that indicated by the models like quantum neural *** to construct and realize an ONN with the quantum speedup is a huge ***,we propose theoretically and demonstrate experimentally a new type of optical convolutional neural network by introducing the optical *** is called the correlated optical convolutional neural network(COCNN).We show that the COCNN can exhibit“quantum speedup”in the training *** character is verified from the two *** is the direct illustration of the faster convergence by comparing the loss function curves of the COCNN with that of the traditional convolutional neural network(CNN).Such a result is compatible with the training performance of the recently proposed quantum convolutional neural network(QCNN).The other is the demonstration of the COCNN’s capability to perform the QCNN phase recognition circuit,validating the connection between the COCNN and the ***,we take the COCNN analog to the 3-qubit QCNN phase recognition circuit as an example and perform an experiment to show the soundness and the feasibility of *** results perfectly match the theoretical *** proposal opens up a new avenue for realizing the ONNs with the quantum speedup,which will benefit the information processing in the era of big data.
