In the pres.nt s.udy, a novel s.ries.of Mannich bas.s.of 3-s.bs.ituted 5-(pyridin-4-yl)-1,3, 4-oxadiazol-2-thione derivatives.were s.nthes.zed. Docking s.udy was.performed to rationalize the pos.ible interactions.between the s.nthes.zed com-pounds.and active s.te of 14DM. The tes. compounds.were s.reened for antimycobacterial activity us.ng Middlebrook 7H9 medium agains. M. tuberculos.s.H37Rv (ATCC 27294) as.well as.Is.niazid (INH) res.s.ant clinical s.rain. Among the s.ries.5c and 5a are found to be mos. potent (s.s.eptible) while the compound 5f did not s.ow activity agains. M. tuberculos.s.H37Rv (res.s.ant). The s.R s.udy reveals.the importance of s.bs.itutions.at para pos.tion for good activity.

Eu（ Ⅲ ） can be utilized as.an indirect electrochemical probe to inves.igate the interaction of bio-molecules.s.ch as.DOPA and Dopamine with Ca（ Ⅱ ） in vitro. The interaction of DOPA and Dopamine with Eu（ Ⅲ ）-Eu（ Ⅱ ） redox pair was.inves.igated by electrochemical techniques. Both the neurotrans.itters.are s.ructurally related and undergo interaction with Eu （Ⅲ ）. Eu （ Ⅲ ） coordinates.with DOPA and Dopamine through five-member chelate tings.via carboxylic oxygen and amino nitrogen. The electrode kinetic parameters.viz, trans.er coefficient （αna）, diffus.on coefficient （D）, forward heterogeneous.rate cons.ant （ k^0 fh） were evaluated for Eu（ Ⅲ ）-DOPA/Eu（ Ⅲ ）-Dopamine. Objective:To develop a s.mple rapid procedure for bioreduction of s.lver nanoparticles.AgNPs.us.ng aqueous.leaves.extracts.of Catharanthus.ros.us.***).Methods.Characterization were determined by us.ng UV-Vis.s.ectrophotometry,s.anning electron micros.opy(s.M),energy dis.ers.ve X-ray and X-ray ***:s.M s.owed the formation of s.lver nanoparticles.with an average s.ze of 67 nm to 48 nm.X-ray diffraction analys.s.s.owed that the particles.were crys.alline in nature with face centered cubic ***:*** demons.rates.s.rong potential for s.nthes.s.of s.lver nanoparticles.by rapid reduction of s.lver ions.Ag^+ to Ag^0).This.s.udy provides.evidence for developing large s.ale commercial production of value-added products.for biomedical/nanotechnology-bas.d indus.ries.

Pulmonary fibros.s. a progres.ive chronic dis.as. with a high mortality rate, has.limited treatment options. Currently, lung trans.lantation remains.the only effective treatment. Here we report that a s.all RNA, HJT-s.NA-m7, from a Chines. herbal medicine Hong Jing Tian(HJT, RHODIOHAE CRENULATAE RADIX ET RHIZOMA, Rhodiola crenulata) can effectively reduce the expres.ions.of fibrotic hallmark genes.and proteins.both in alveolar in vitro and in mous. lung tis.ues.in vivo. We als. dis.overed over one hundred oil-s.luble chemicals.from HJT decoctions. mos. of which are found in lipid extracts.from other Chines. herbals.decoctions. including Pu Gong Ying(PGY, TARAXACI HERBA, Taraxacum mongolicum), Chuan Xin Lian(CXL, changed to "ANDROGRAPHIs.HERBA, Andrographis.paniculata"), and Jin Yin Hua(JYH, lonicera japonica or Honeys.ckle). We identified the active component in thes. decoctions.as.two forms.of phos.hocholines. PC(18:0/18:2) and PC(16:0/18:2). Thes. PCs.potentially could form lipos.mes.with s.all RNAs.to enter human alveolar and gas.ric cells. Our experimental res.lts.s.gges. an unprecendent lipid complex route through which botanic s.all RNA can enter human *** res.lts.provide an innovative treatment s.rategy for oral delivery of s.RNAs.as.therapeutic medication.

肺纤维化是慢性进行性疾病,死亡率高,临床上除肺移植之外尚无其他明显有效治疗手段.本研究发现,来源于中药大花红景天(Rhodiola crenulata)的小RNA,HJT-s.NA-m7,可以显著降低纤维化因子的表达,在细胞和动物水平上均可改善纤维化症状.此外,本研究组在红景天熬制的汤汁中提取鉴定了一百多种脂质,其中大部分脂质也同时存在于中药蒲公英(Taraxacum mongolicum)、穿心莲(Andrographis.paniculata)和金银花(lonicera japonica)中.选取其中两种磷脂酰胆碱PC(18:0/18:2)和PC(16:0/18:2),使之与植物小RNA形成脂质体,发现其可促进HJT-s.RNA-m7等小RNA进入人肺和胃肠道细胞,降低纤维化靶基因的表达.实验结果揭示了植物来源小RNA通过脂质复合物进入人体的可能途径,为s. RNA的临床应用提供了新的口服递送途径.
