Objective:To analyze the effects of feeding Helianthus tuberosus(HT) tubers on glucose tolerance and lipid profile in rats fed a high-fat diet(HFD). Methods:A normal HFD or HFD including 10 w/w% HT tubers(HFD + HT) was fed to F334/Jcl rats. After 10 weeks,organ weights,glucose tolerance,and lipid profile were analyzed. Results:The body weight,liver weight,and epidermal fat content in the HFD group were higher than those of the normal group,and similar to those of the HFD + HT group. The oral glucose tolerance test at 10 weeks revealed that the blood glucose level 30 minutes after beginning the test in the HFD + HT group was significantly lower than that in the HFD group. Liver triglyceride and total cholesterol levels in the HFD + HT group were significantly lower than those in the HFD group. Fecal triglyceride and total cholesterol levels in the HFD + HT group were higher than those in the HFD group. Histological analyses revealed that fat and glycogen accumulation increased in the HFD group,but decreased in the HFD + HT group. Conclusions:These results indicate that HT tubers have anti-fatty liver effects based on improvements in glucose tolerance and the hepatic lipid profile.

CYP1A1 , GSTM1 , GSTT1 and NQO1 polymorphisms and colorectal adenomas in Japanese men

AIM: To investigate the role of functional genetic poly-morphisms of metabolic enzymes of tobacco carcinogens in the development of colorectal adenomas. METHODS: The study subjects were 455 patients with colorectal adenomas and 1052 controls with no polyps who underwent total colonoscopy in a preretirement health examination at two Self Defense Forces hospitals. The genetic polymorphisms studied wereCYP1A1*2A (rs 4646903), CYP1A1*2C (rs 1048943), GSTM1 (null or non-null genotype), GSTT1 (null or non-null genotype) and NQO1 C609T (rs 1800566). Genotypes were determined by the polymerase chain reaction (PCR)-restriction fragment length polymorphism or PCR method using genomic DNA extracted from the buffy coat. Cigarette smoking and other life-style factors were ascertained by a self-administered questionnaire. The associations of the polymorphisms with colorectal adenomas were examined by means of OR and 95%CI, which were derived from logistic regression analysis. Statistical adjustment was made for smoking, alcohol use, body mass index and other factors. The gene-gene interaction and effect modification of smoking were evaluated by the likelihood ratio test. RESULTS: None of the five polymorphisms showed a significant association with colorectal adenomas, nor was the combination of GSTM1 and GSTT1 . A borderline significant interaction was observed for the combination of CYP1A1*2C and NQO1 (P = 0.051). The OR associated with CYP1A1*2C was significantly lower than unity among individuals with the NQO1 609CC genotype. The adjusted OR for the combination of the CYP1A1*2C allele and NQO1 609CC genotype was 0.61 (95%CI: 0.42-0.91). Although the interaction was not statistically significant (P = 0.24), the OR for individuals carrying the CYP1A1*2C allele and GSTT1 null genotype decreased significantly compared with those who had neither CYP1A1*2C allele nor GSTT1 null genotype (adjusted OR: 0.69, 95%CI: 0.49-0.97). The carbon nanotube (CNT) growth of iron oxide-deposited trench-patterns and the locally-ordered CNT arrays on silicon substrate were achieved by simple thermal chemical vapor deposition(STCVD) of ethanol vapor. The CNTs were uniformly synthesized with good selectivity on trench-patterned silicon substrates. This fabrication process is compatible with currently used semiconductor-processing technologies, and the carbon-nanotube fabrication process can be widely applied for the development of electronic devices using carbon-nanotube field emitters as cold cathodes and can revolutionize the area of field-emitting electronic devices. The site-selective growth of CNT from an iron oxide nanoparticle catalyst patterned were also achieved by drying-mediated self-assembly technique. The present method offers a simple and cost-effective method to grow carbon nanotubes with self-assembled patterns.

Fungal aromatic compounds comprise an important and structurally diverse group of secondary *** genome sequencing projects revealed many putative biosynthetic gene clusters of fungal aromatic compounds,but many of these genes seem to be silent under typical laboratory culture *** gain access to this untapped reservoir of natural products,we utilized chemical epigenetic modifiers to induce the expression of dormant biosynthetic *** a result,the concomitant supplementation of the histone deacetylase inhibitors suberoylanilide hydroxamic acid(500mM)and nicotinamide(50mM)to the culture medium of a fungal pathogen,Stagonospora nodorum,resulted in the isolation of three aromatic compounds(1-3),including a novel natural butyrophenone,(+)-4'-methoxy-(2S)-methylbutyrophenone(1),and two known polyketides,alternariol(2)and(-)-(3R)-mellein methyl ether(3).

The characterization of electrical property of multi-walled carbon nanotubes (MWCNTs) on a nanometer scale is essential for their potential application in nano-electronic devices. The MWCNTs were synthesized on Fe2O3/SiO2/Si substrate and Pt plate substrate by simple thermal chemical vapor deposition (STCVD) technique and the electrical measurements of individual MWCNT grown on silicon substrate and Pt plate substrate were performed by home-made 'nano-manipulator', respectively. According to current-voltage curves obtained in the experiments the current density that the MWCNTs can carry is calculated to be about 107 A/cm2, which is much larger than that of normal metals.

A 58-year-old man was admitted to our hospital. Laboratory data showed severe liver injury and that the patient was positive for immunoglobulin M anti-hepatitis A virus(HAV) antibodies. He was also complicated with severe renal dysfunction and had an extremely high level of serum hepatocyte growth factor(HGF). Therefore, he was diagnosed with severe acute liver failure with acute renal failure(ARF) caused by HAV infection. Prognosis was expected to be poor because of complications by ARF and high serum HGF. However, liver and renal functions both improved rapidly without intensive treatment, and he was subsequently discharged from our hospital on the 21 st hospital day. Although complication with ARF and high levels of serum HGF are both important factors predicting poor prognosis in acute liver failure patients, the present case achieved a favorable outcome. Endogenous HGF might play an important role as a regenerative effector in injured livers and kidneys.

Fusidic acid is the only fusidane-type antibiotic that has been clinically used. However,biosynthesis of this important molecule in fungi is poorly understood. We have recently elucidated the biosynthesis of fusidane-type antibiotic helvolic acid, which provides us with clues to identify a possible gene cluster for fusidic acid(fus cluster). This gene cluster consists of eight genes, among which six are conserved in the helvolic acid gene cluster except fusC1 and fusB1. Introduction of the two genes into the Aspergillus oryzae NSAR1 expressing the conserved six genes led to the production of fusidic acid. A stepwise introduction of fusC1 and fusB1 revealed that the two genes worked independently without a strict reaction order. Notably, we identified two short-chain dehydrogenase/reductase genes fusC1 and fusC2 in the fus cluster, which showed converse stereoselectivity in 3-ketoreduction. This is the first report on the biosynthesis and heterologous expression of fusidic acid.

Silicon Deposition in Leaf Trichomes of Cucurbitaceae Horticultural Plants: A Short Report

Silicon deposition in leaf trichome of six horticultural Cucurbitaceae species, cucumber (Cucumis sativus), pumpkin (Cucurbita maxima), melon (Cucumis melo), watermelon (Citrullus lanatus), sponge gourd (Luffa cylindrica) and bottle gourd (Lagenaria siceraria var. hispida) was observed by an X-ray microanalyzer coupled with an environmental scanning electron microscope. The elements that presented in the surface of three or four leaves of the individual species were detected and mapped by the X-ray microanalyzer. In leaves of cucumber, pumpkin, and melon, high accumulation of silicon was detected in cells surrounding the bases of the trichome hair and the hair itself deposited calcium. On the other hand, in sponge gourd and bottle gourd, high accumulation of silicon was detected only in the hair. In watermelon leaves, silicon deposited both in the hair and in cells surrounding the bases of the hair. Thus, horticultural Cucurbitaceae plants have interspecific variation in the pattern of silicon deposition in leaf trichomes.
