Interplay of neuropilin-1 and semaphorin 3A after partial hepatectomy in rats

AIM： To elucidate the role of neuropilin-1 （Nrp-1） and semaphorin 3A （Sema3A） in sinusoidal remodeling dur- ing liver regeneration in rats. METHODS： Male Wistar/ST rats at 7 wk of age, weigh- ing about 200 g, were used for all animal experiments. In vivo, at 24, 48, 72, 96, 144 and 192 h after two- thirds partial hepatectomy （PHx）, the remnant livers were removed. Liver tissues were immunohistochemi- cally stained for Nrp-1, Sema3A and SE-1, a liver sinu- soidal endothelial cell （SEC） marker. Total RNA of the liver tissue was extracted and reversely transcribed into cDNA. The mRNA expression of Sema3A was ana- lyzed by quantitative real-time polymerase chain reac- tion and normalized to that of ribosomal protein $18. In vitro, SECs were isolated from rat liver and cultured in endothelial growth medium containing 20 ng/mL vascular endothelial cell growth factor. Migration of SECs in primary culture was assessed by cell transwell assay with or without recombinant Sema3A. Apoptotic cells were determined by a terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end labeling method. RESULTS： In vitro, immunohistochemistry study re- vealed that Sema3A and Nrp-1 were constitutively ex- pressed in hepatocytes and SECs, respectively, in normal rat liver tissues. Nrp-1 expression in SECs was quantified by the percentage of immunostained area with anti- Nrp-1 antibody in relation to the area stained with SE-1. Between 24 h and 96 h following resection of liver, Nrp-1 expression in SECs was transiently increased. Compared with the baseline （5.2% ± 0.1%）, Nrp-1 expression in SECs significantly increased at 24 h （17.3% ± 0.7%, P 〈 0.05）, 48 h （39.1% ± 0.6%, P 〈 0.01）, 72 h （46.9% ± 4.5%, P 〈 0.01） and 96 h （29.9% ±3.8%, P 〈 0.01） after PHx, then returned to the basal level at termination of liver regeneration. Site effects by generalized inversion technique using strong motion recordings of the 2008 Wenchuan earthquake

The generalized inversion of S-wave amplitude spectra from the free-field strong motion recordings of the China National Strong Motion Observation Network System （NSMONS） are used to evaluate the site effects in the Wenchuan area. In this regard, a total of 602 recordings from 96 aftershocks of the Wenchuan earthquake with magnitudes of M3.7-M6.5 were selected as a dataset. These recordings were obtained from 28 stations at a hypocenter distance ranging from 30 km to 150 km. The inversion results have been verified as reliable by comparing the site response at station 62WUD using the Generalized Inversion Technique （GIT） and the Standard Spectral Ratio method （SSR）. For all 28 stations, the site predominant frequency F and the average site amplification in different frequency bands of 1.0-5.0 Hz, 5.0-10.0 Hz and 1.0-10.0 Hz have been calculated based on the inversion results. Compared with the results from the horizontal-to-vertical spectral ratio （HVSR） method, it shows that the HVSR method can reasonably estimate the site predominant frequency but underestimates the site amplification. The linear fitting between the average site amplification for each frequency band and the V20 （the average uppermost-20 m shear wave velocity） shows good correlation. A distance measurement called the asperity distance DAspt is proposed to reasonably characterize the source-to-site distance for large earthquakes. Finally, the inversed site response is used to identify the soil nonlinearity in the main shock and aftershocks of Wenchuan earthquake. In ten of the 28 stations analyzed in the main shock, the soil behaved nonlinearly, where the ground motion level is apparently beyond a threshold ofPGA 〉 300 cm/s^2 or PGV 〉 20 cm/s, and only one station coded 51SFB has evidence of soil nonlinear behavior in the aftershocks.

Prediction of GPCR-G Protein Coupling Specificity Using Features of Sequences and Biological Functions

Understanding the coupling specificity between G protein-coupled receptors （GPCRs） and specific classes of G proteins is important for further elucidation of receptor functions within a cell. Increasing information on GPCR sequences and the G protein family would facilitate prediction of the coupling properties of GPCRs. In this study, we describe a novel approach for predicting the coupling specificity between GPCRs and G proteins. This method uses not only GPCR sequences but also the functional knowledge generated by natural language processing, and can achieve 92.2% prediction accuracy by using the C4.5 algorithm. Furthermore, rules related to GPCR-G protein coupling are generated. The combination of sequence analysis and text mining improves the prediction accuracy for GPCR-G protein coupling specificity, and also provides clues for understanding GPCR signaling.

A High-Precision US-Guided Robot-Assisted HIFU Treatment System for Breast Cancer

Breast cancer is the most commonly diagnosed cancer in women. A strong treatment candidate is high- intensity focused ultrasound （HIFU）, a non-invasive therapeutic method that has already demonstrated its promise. To improve the precision and lower the cost of HIFU treatment, our group has developed an ultrasound （US）-guided, five-degree-of-freedom （DOF）, robot-assisted HIFU system. We constructed a fully functional prototype enabling easy three-dimensional （3D） US image reconstruction, target seg-mentation, treatment path generation, and automatic HIFU irradiation. The position was calibrated using a wire phantom and the coagulated area was assessed on heterogeneous tissue phantoms. Under the US guidance, the centroids of the HIFU-ahlated area deviated by less than 2 mm from the planned treatment region. The overshoot around the planned region was well below the tolerance of clinical usage. Our system is considered to he sufficiently accurate for breast cancer treatment.

Structural phase transition, antiferromagnetism and two superconducting domes in LaFeAsO_(1-x)F_x(0

We report^(75) As nuclear magnetic resonance(NMR)/nuclear quadrupole resonance(NQR) and transmission electron microscopy(TEM) studies on LaFeAsO_(1-x)F_x. There are two superconducting domes in this material. The first one appears at 0.03 ≤ x ≤0.2 with T_c^(max) = 27 K, and the second one at 0.25 ≤x≤0.75 with T_c^(max) = 30 K. By NMR and TEM, we demonstrate that a C4-to-C2 structural phase transition(SPT) takes place above both domes, with the transition temperature T_s varying strongly with x. In the first dome, the SPT is followed by an antiferromagnetic(AF) transition, but neither AF order nor low-energy spin fluctuations are found in the second dome. By ^(75) As nuclear spin-lattice relaxation rate(1/T_1) measurements, we find that AF order and superconductivity coexist microscopically in LaFeAsO_(0.97) F_(0.03). In the coexisting region, 1/T_1 decreases at T_c but becomes proportional to T below 0.6 T_c, indicating gapless excitations. Therefore, in contrast to the early reports, the obtained phase diagram for x ≤ 0.2 is quite similar to the doped BaFe_2As_2 system. The electrical resistivity p in the second dome can be fitted by ρ = ρ0 + AT^n with n = 1 and a maximal coefficient A at around xopt = 0.5-0.55 at which T_s extrapolates to zero and Tc is the maximal, which suggests the importance of quantum critical fluctuations associated with the SPT. We have constructed a complete phase diagram of LaFeAsO_(1-x)F_x, which provides insight into the relationship between SPT, antiferromagnetism and superconductivity.
