Atorvastatin, etanercept and the nephrogenic cardiac sympathetic remodeling in chronic renal failure rats

BACKGROUND Chronic renal failure(CRF) patients are predisposed to arrhythmias, while the detailed mechanisms are unclear. We hypothesized the chronic inflammatory state of CRF patients may lead to cardiac sympathetic remodeling, increasing the incidence of ventricular arrhythmia(VA) and sudden cardiac death. And explored the role of atorvastatin and etanercept in this *** A total of 48 rats were randomly divided into sham operation group(Sham group), CRF group, CRF + atorvastatin group(CRF + statin group), and CRF + etanercept group(CRF + rhTNFR-Fcgroup). Sympathetic nerve remodeling was assessed by immunofluorescence of growth-associated protein 43(GAP-43) and tyrosine hydroxylase positive area fraction. Electrophysiological testing was performed to assess the incidence of VA by assessing the ventricular effective refractory period and ventricular fibrillation threshold. The levels of tumor necrosis factor-alpha(TNF-α) and interleukin-1beta were determined by Western blotting and enzyme-linked immunosorbent *** Echocardiogram showed that compared with the Sham group, left ventricular end-systolic diameter and ventricular weight/body weight ratio were significantly higher in the CRF group. Hematoxylin-eosin and Masson staining indicated that myocardial fibers were broken, disordered, and fibrotic in the CRF group. Western blotting, enzyme-linked immunosorbent assay,immunofluorescence and electrophysiological examination suggested that compared with the Sham group, GAP-43 and TNF-α proteins were significantly upregulated, GAP-43 and tyrosine hydroxylase positive nerve fiber area was increased, and ventricular fibrillation threshold was significantly decreased in the CRF group. Clean air actions （CAAs） in China have been linked to considerable benefits in public ***,whether the beneficial effects of CAAs are equally distributed geographically is *** high-resolution maps of the distributions of major air pollutants (fine particulate matter[PM2.5]and ozone[O3]) and population,we aimed to track spatiotemporal changes in health impacts from,and geographic inequality embedded in,the reduced exposures to PM2.5and O3from 2013 to *** used a method established by the Global Burden of Diseases *** analyzing the changes in loss of life expectancy（LLE） attributable to PM2.5and O3,we calculated the gain of life expectancy （GLE） to quantify the health benefits of the air-quality ***,we assessed the geographic inequality embedded in the GLE using the Gini index （GI）.Based on risk assessments of PM2.5and O3,during the first stage of CAAs（2013 to 2017）,the mean GLE was 1.87 *** of the sum of the GLE was disproportionally distributed in about one quarter of the population exposed （GI 0.44）.During the second stage of CAAs（2017 to 2020）,the mean GLE increased to 3.94 months and geographic inequality decreased （GI 0.18）.According to our assessments,CAAs were enhanced,from the first to second stages,in terms of not only preventing premature mortality but also ameliorating health *** enhancements were related to increased sensitivity to the health effects of air pollution and synergic control of PM2.5and *** findings will contribute to optimizing future CAAs.

Benefitting from narrow beam divergence,photonic crystal surface-emitting lasers are expected to play an essential role in the ever-growing fields of optical communication and light detection and *** operating with 1.55μm wavelengths have attracted particular attention due to their minimum fiber loss and high eye-safe ***,high interband absorption significantly decreases their performance at this 1.55μm ***,stronger optical feedback is needed to reduce their threshold and thus improve the output *** this goal,photonic-crystal resonators with deep holes and high dielectric contrast are often ***,the relevant techniques for high-contrast photonic crystals inevitably complicate fabrication and reduce the final *** this paper,we demonstrate the first continuous-wave operation of 1.55μm photonic-crystal surface-emitting lasers by using a‘triple-lattice photonic-crystal resonator’,which superimposes three lattice point groups to increase the strength of in-plane optical *** this geometry,the in-plane 180°coupling can be enhanced threefold compared to the normal single-lattice *** theoretical and experimental investigations demonstrate the much lower threshold current density of this structure compared to‘single-lattice’and‘double-lattice’photonic-crystal resonators,verifying our design *** findings provide a new strategy for photonic crystal laser miniaturization,which is crucial for realizing their use in future high-speed applications.

A multi-omics technology to the study of lactic acid bacteria responses to environmental stress: the past， current and future trends

Lactic acid bacteria and the fermentation environment interact to form an intertwined system. Lactic acid bacteria are constantly evolving to adapt to different fermentation environments， causing changes in their physiological processes. To achieve a targeted improvement of their adaptability to various environments， a detail analysis of their evolutionary physiological processes is required. While several studies have been carried out in the past by using single-omics techniques to investigate their response to environmental stress， most researchers are now using a multi-omics approach to explore more detail in the biological regulatory networks and molecular mechanisms of lactic acid bacteria in response to environmental stress， thereby overcoming the limitations of single- omics analysis. Scope and approach: In this review， we describe the various single-omics approaches that have been used to study environmental stress in lactic acid bacteria， present the advantages of various multi-omics combined analysis approaches， and discuss the potential and practicality of applying emerging single-cell transcriptomics and single-cell metabolomics techniques to the molecular mechanism study of microbes response to environmental stress. Key ?ndings and conclusions: Multi-omics approaches enable the accurate identification of complex microbial physiological processes in different environments， allow people to comprehensively reveal the molecular mechanisms of microbes response to stress from different perspectives. Single-cell omics techniques， analyze the targeted regulation of microbial functions in a multi-dimensional space， provides a new perspective on understanding microbes responses environment stress.

Lipid and sugar homeostasis is critical for insect development and *** this study,we characterized an acetyl coenzyme A carboxylase gene in Blattella germanica(BgACC)that is involved in both lipogenesis and sugar *** found that BgACC was dominantly expressed in the fat body and integument,and was significantly upregulated after *** of BgACC in 5th-instar nymphs did not affect their normal molting to the next nymphal stage,but it caused a lethal phenotype during adult ***-RNA interference(RNAi)significantly downregulated total free fatty acid(FFA)and triacylglycerol(TAG)levels,and also caused a significant decrease of cuticular hydrocarbons(CHCs).Repression of BgACC in adult females affected the development of oocytes and resulted in sterile females,but BgACC-RNAi did not affect the reproductive ability of ***,knockdown of BgACC also changed the expression of insulin-like peptide genes(BglLPs),which mimicked a physiological state of high sugar *** addition,BgACC was upregulated when *** were fed on a high sucrose diet,and repression of BgACC upregulated the expression of the glycogen synthase gene(BgGlyS).Moreover,BgACC-RNAi increased the circulating sugar levels and glycogen storage,and a longevity assay suggested that BgACC was important for the survival of *** under conditions of high sucrose *** results confirm that BgACC is involved in multiple lipid biogenesis and sugar homeostasis processes,which further modulates insect reproduction and sugar *** study benefits our understanding of the crosstalk between lipid and sugar metabolism.

Regulating atomic Fe-Rh site distance for efficient oxygen reduction reaction

Exploring the atomic interaction mechanisms of dense single-atom catalysts(SACs)is of great significance for their application in oxygen reduction reaction(ORR).However,the intrinsic mechanism of the site-distance effect on the catalytic performance has been largely ***,we demonstrate the site-distance effect of Fe-Rh_(x)@NC catalysts in ORR theoretically and *** charge analysis reveals that the strong interaction between Fe and Rh atoms at a certain atomic distance(dFe-Rh)alters the catalytic electronic structure,facilitating the optimization of catalyst adsorption *** by the theoretical calculations,we designed and synthesized the Fe-Rhx@NC catalysts through a spatial confinement *** characterization results prove that the Fe-Rh_(2)@NC has the optimal d_(Fe-Rh),which improves its intrinsic ORR activity,providing a half wave potential of 0.91 V,higher than that of the commercial Pt/C(0.86 V).This study emphasizes the importance of determining the basic mechanism of the site-distance effect in dissimilar metal atoms catalysts,which is conducive to the design of efficient catalyst systems for practical applications.

Alternative relay regulates the adenosine triphosphatase activity of Locusta migratoria striated muscle myosin

Locust(Locusta migratoria)has a single striated muscle myosin heavy chain(Mhc)gene,which contains 5 clusters of alternative exclusive exons and 1 differently included penultimate *** alternative exons of Mhc gene encode 4 distinct regions in the myosin motor domain,that is,the N-terminal SH3-like domain,one lip of the nucleotide-binding pocket,the relay,and the ***,we investigated the role of the alternative regions on the motor function of locust muscle *** Sf9-baculovirus protein expression system,we expressed and purified 5 isoforms of the locust muscle myosin heavy meromyosin(HMM),including the major isoform in the thorax dorsal longitudinal flight muscle(FL1)and 4 isoforms expressed in the abdominal intersegmental muscle(AB1 to AB4).Among these 5 HMMs,FL1-HMM displayed the highest level of actin-activated adenosine triphosphatase(ATPase)activity(hereafter referred as ATPase activity).To identify the alternative region(s)responsible for the elevated ATPase activity of FL1-HMM,we produced a number of chimeras of FL1-HMM and *** with the relay of AB4-HMM(encoded by exon-14c)substantially decreased the ATPase activity of FL1-HMM,and conversely,the relay of FL1-HMM(encoded by exon-14a)enhanced the ATPase activity of *** showed that the exon-14a-encoded residues Gly474 and Asn509 are responsible for the elevated ATPase activity of *** results indicate that the alternative relay encoded by exon-14a/c play a key role in regulating the ATPase activity of FL1-HMM and AB4-HMM.

Dynamic response and failure process of horizontal-layered fractured structure rock slope under strong earthquake

Rock slope with horizontal-layered fractured structure(HLFS)has high stability in its natural ***,a strong earthquake can induce rock fissure expansion,ultimately leading to slope *** this study,the dynamic response,failure mode,and spectral characteristics of rock slope with HLFS under strong earthquake conditions were investigated based on the large-scale shaking table model *** this basis,multiple sets of numerical calculation models were further established by UDEC discrete element *** influencing factors were considered in the parametric study of numerical simulations,including slope height,slope angle,bedding-plane spacing and secondary joint spacing as well as bedrock dip *** results showed that the failure process of rock slope with HLFS under earthquake action is mainly divided into four phases,i.e.,the tensile crack of the slope shoulder joints and shear dislocation at the top bedding plane,the extension of vertical joint cracks and increase of shear displacement,the formation of step-through sliding surfaces and the instability,and finally collapse of fractured rock *** acceleration response of slopes exhibits elevation amplification effect and surface *** simulations indicate that the seismic stability of slopes with HLFS exhibits a negative correlation with slope height and angle,but a positive correlation with bedding-plane spacing,joint spacing,and bedrock dip *** results of this study can provide a reference for seismic stability evaluation of weathered rock slopes.
