Atomic force microscopy analysis of orientation and bending of oligodeoxynucleotides in polypod-like structured DNA

We previously demonstrated that polypod-like structured DNA, or polypodna, constructed with three or more oligodeoxynucleotides （ODNs）, is efficiently taken up by immune cells such as dendritic cells and macrophages, depending on its structural complexity. The ODNs comprising the polypodna should bend to form the polypod-like structure, and may do so by adopting either a bend- type conformation or a cross-type conformation. Here, we tried to elucidate the orientation and bending of ODNs in polypodnas using atomic force microscopy （AFM）. We designed two types of pentapodnas （i.e., a polypodna with five pods） using 60- to 88-base ODNs, which were then immobilized on DNA origami frames. AFM imaging showed that the ODNs in the pentapodna adopted bend-type conformations. Tetrapodna and hexapodna also adopted bend-type conformations when they were immobilized on frames under unconstrained conditions. These findings provide useful information toward the coherent design of, and the structure-activity relationships for, a variety of DNA nanostructures.

All engineering students need to develop their important skills of leadership in project management. Many students have never been leaders in their social and school lives. A leading role is unimaginable to them and hence they cannot imagine how to achieve it. The purpose of this paper is to report a result of a new leadership education program which links a variety of simulated experiences with real actions of students in project based learning （PBL） to develop their leadership ability. The first step is for graduate students to gain knowledge in the leadership arena. Then, they utilize simulation to experience leadership actions many times. Simulation provides a safe environment in which they can try out many different approaches in taking leadership in various situations. In the next step, students as a team utilize PBL, so that the above simulated experiences can help them to actually take leadership. Students can apply trained leadership to actual projects. It is highly effective to apply conscious leadership to a project aimed at a specific goal in limited circumstances. This education repeats both of the steps above, raising leadership abilities in an upward spiral. In terms of students＇ evaluation of leadership education in project management, 360-degree assessments were carried out by teachers, senior, and junior students before and after the course, and authors compare their assessments thoroughly. As a result, authors are assured that students not only gained knowledge but also raised leadership abilities in their actions after this education. Six months after the time of leadership education employing simulated experiences and PBL, follow-up interviews were conducted on its effects. Authors recognized the cyclic period that students apply simulated experiences to PBL and that they seek different approaches in simulation for solving problems found in reality. Development of Preparative Chromatography for Proteomic Approach of Mycorrhizal Symbiosis

Although mechanism of symbiosis between arbuscular mycorrhizal fungi (AMF) and host plants has been investigated by genetic analysis, very little knowledge has been obtained because genome analysis of AMF is not perfect yet. Thus, we tried to develop mass purification of proteins using preparative chromatography in order to accelerate roteomic analysis of proteins related to mycorrhizal symbiosis, such as 24 and 53 kDa proteins. In particular, our data showed that 53 kDa proteins would be restrictively expressed when mycorrhizal fungi and host plants were stressed. However, 24 kDa proteins, which appear to be a usable indicator for the existence of various my-corrhizal fungi, were habitually detected in not only AMF but also other mycorrhizal fungi such as ectomycorrhizal fungi (EF). Moreover, we discovered new preparative chromatographical techniques for isolation and mass purification of those proteins. We are convinced that this chromato-graphical technique will greatly contribute to proteomic approach of mycorrhizal symbiosis.

Effect of Intention on Outcome Bias in Decision Making—Implications for Safety Management

The evidence of outcome bias was explored in a two-player (Player 1: allocator and Player 2: evaluator) economic game experiment where the reward allocation was made between two players. The experimental factors were the intention of an allocator (Player 1), the type of chosen dice (selfish, fair, and generous), and the outcome (selfish, fair, and generous). The outcome bias occurred when the type of dice chosen by the allocator (Player 1) was not only a selfish one but also a generous one. The comparison between the two conditions (intentional and no-intentional conditions) definitely showed that Player 2 punished Player 1 to a larger extent when the outcome was disadvantageous for Player 2 (selfish outcome) and Player 2 rewarded Player 1 when the outcome was advantageous (generous outcome) irrespective of whether the die was chosen out of the three types intentionally or not. Moreover, the outcome bias was not observed when the outcome was fair. Thus, we could verify the hypothesis that we are readily got trapped in the outcome bias. Some implications were given for safety management that put more emphasis on the process than on the outcome.

A threshold scheme, which is introduced by Shamir in 1979, is very famous as a secret sharing scheme. We can consider that this scheme is based on Lagrange＇s interpolation formula. A secret sharing scheme has one key. On the other hand, a multi-secret sharing scheme has more than one key, that is, a multi-secret sharing scheme has p （〉_ 2） keys. Dealer distribute shares of keys among n participants. Gathering t （〈 n） participants, keys can be reconstructed. Yang et al. （2004） gave a scheme of a （t, n） multi-secret sharing based on Lagrange＇s interpolation. Zhao et al. （2007） gave a scheme of a （t, n） verifiable multi-secret sharing based on Lagrange＇s interpolation. Recently, Adachi and Okazaki give a scheme of a （t, n） multi-secret sharing based on Hermite interpolation, in the case ofp 〈 t. In this paper, we give a scheme ofa （t, n） verifiable multi-secret sharing based on Hermite interpolation.

On the other hand, a multi-secret sharing scheme has more than one keys;that is, a multi-secret sharing scheme has p (≥2) keys. Dealers distribute shares of keys among n participants. Gathering t (≤n) participants, keys can be reconstructed. In this paper, we give a scheme of a (t,n) multi-secret sharing based on Hermite interpolation, in the case of p≤t.

A Survey of Learning Needs about Oral Care for Intubated Patients among New Nursing Staff

Our aim was to understand the actual conditions of oral care for intubated patients by new nurses. This study was carried out from December 2017 to March 2018. We applied the theme of the dissipation of the dissociation between nursing skills learned in basic nursing education and nursing skills required in practice. A questionnaire survey was conducted at four facilities among new nursing staff who had acquired their nursing license in the past year. The questionnaire was composed of questions about their experiences of oral care for intubated patients, when they learned these skills, and what they wanted to learn. Answers were obtained from 118 new nurses. Learning about oral care was important for approximately 20% of the nurses, although it was not considered mandatory by all of the nurses. As infection control measures have revealed the acute needs of care for intubated patients, it is important for nursing students to learn about oral care and attitudes towards intubated patients, along with methods to make intubated patients more comfortable.

A Feasibility Study of Power Generation from Sewage Using a Hollowed Pico-Hydraulic Turbine

This study is concerned with the feasibility of power generation using a pico-hydraulic turbine from sewage flowing in pipes. First, the sewage flow rate at two connection points to the Toyogawa River-Basin Sewerage, Japan, was explored for over a year to elucidate the hydraulic energy potential of the sewage. Second, the performance of the pico-hydraulic turbine was investigated via laboratory experiments that supposed the turbine to be installed in the sewage pipe at the connection points. This study indicates that the connection points have hydraulic potential that can be used for power generation throughout the year. It also demonstrates that the pico-hydraulic turbine can be usefully employed for power generation from sewage flowing in the pipe at the connection points.

Primary biliary cirrhosis (PBC) is an autoimmune disease characterized by anti-mitochondrial antibodies and destruction of intra-hepatic bile ducts. Though little is known about the etiology of PBC, some reports suggest that xenobiotics and viral infections may induce PBC. We describe a case of PBC after the aortoiliac reconstruction surgery using a Y-graft.
