AIM: To investigate whether accelerated catabolism of ganglioside and decreased ganglioside content contribute to the etiology of pro-inflammatory intestinal disease. METHODS: Intestinal mucosa from terminal ileum or colon was obtained from patients with ulcerative colitis or inflammatory Crohn's disease(n = 11) undergoing bowel resection and compared to control samples of normal intestine from patients with benign colon polyps(n = 6) and colorectal cancer(n = 12) in this observational case-control study. Gangliosides and phospholipids of intestinal mucosa were characterized by class and ceramide or fatty acid composition using liquid chromatography triple-quad mass spectrometry. Content and composition of ganglioside classes GM1, GM3, GD3, GD1 a, GT1 and GT3 were compared among subject groups. Content and composition of phospholipid classes phosphatidylcholine(PC) and phosphatidylethanolamine were compared among subject groups. Unsaturation index of individual ganglioside and phospholipid classes was computed and compared among subject groups. Ganglioside catabolism enzymes beta-hexosaminidase A(HEXA) and sialidase-3(NEU3) were measured in intestinal mucosa using western blot and compared among subject groups. RESULTS: Relative GM3 ganglioside content was 2-fold higher(P < 0.05) in intestine from patients with inflammatory bowel disease(IBD) compared to control intestine. The quantity of GM3 and ratio of GM3/GD3 was also higher in IBD intestine than control tissue(P < 0.05). Control intestine exhibited 3-fold higher(P < 0.01) relative GD1 a ganglioside content than IBD intestine. GD3 and GD1 a species of ganglioside containing three unsaturated bonds were present in control intestine, but were not detected in IBD intestine. The relative content of PC containing more than two unsaturated bonds was 30% lower in IBD intestine than control intestine(P < 0.05). The relative content of HEXA in IBD intestine was increased 1.7-fold(P < 0.05) and NEU3 was increased 8.3-fold

Purpose The aims of the present study were two-fold:(i)to investigate the relationship between physical characteristics and the game statistics associated with ball-carrying capability amongst sub-elite rugby union players,and(ii)to predict the level of change in these physical characteristics required to improve the associated game statistic via regression *** Thirty-eight senior professional players(forwards,n=22;backs,n=16)were assessed for body mass(BM),back squat(BS)single-repetition maximum(1RM)normalised to BM(1RM/BM),10 m sprint velocity(S10),10 m sprint momentum(SM10),and the game statistics from 22 games within the 2019/20 RFU Championship *** relationship between these measures and the predicted level of change in a physical measure required to improve the total number of the associated game statistic by one were assessed by Pearson’s correlation coefficient and simple regression *** In forwards,an~11.5%reduction in BM,an~11.8%improvement in BS 1RM/BM,or an~11.5%increase in S10 was required to improve the game statistics associated with ball-carrying *** backs,a~19.3%increase in BM or a~15.6%improvement in SM10 was *** These findings demonstrate that improvements in lower-body relative strength,acceleration performance,and position-specific alterations in body mass are required to maximise the ball-carrying capability and therefore match outcome of sub-elite rugby union players.

Morphological transformation of surface structures is widely manifested in nature and highly preferred for many applications such as wetting interaction;however,in situ tuning of artificial morphologies independent of smart responsive materials remains ***,with the aid of microfluidics,we develop a pneumatic programmable superrepellent surface by tailoring conventional wetting materials(e.g.,polydimethylsiloxane)with embedded flexible chambers connecting a microfluidic system,thus realizing a morphological transformation for enhanced liquid repellency based on a nature‐inspired rigid‐flexible hybrid principle(i.e.,triggering symmetry breaking and oscillator coupling mechanisms).The enhancement degree can be in situ tuned within around 300 ms owing to pneumatically controllable chamber *** also demonstrate that the surface can be freely programmed to achieve elaborated morphological pathways and gradients for preferred droplet manipulation such as directional rolling and *** study highlights the potential of an in situ morphological transformation to realize tunable wettability and provides a programmable level of droplet control by intellectualizing conventional wetting materials.

The genus Sinocyclocheilus (golden-line barbel) includes 25species of cave-dwelling blind fish (cavefish)and more than 30 surface-dwelling species with normal *** environments are dark and generally nutrient-poor with few *** of several genera evolved convergent morphological adaptations in visual,pigmentation,brain,olfactory,and digestive systems. AIM To investigate viability assessment of segmental small bowel ischemia/reperfusion in a porcine *** In 15 pigs, five or six 30-cm segments of jejunum were simultaneously made ischemic by clamping the mesenteric arteries and veins for 1 to 16 h. Reperfusion was initiated after different intervals of ischemia(1-8 h) and subsequently monitored for 5-15 h. The intestinal segments were regularly photographed and assessed visually and by palpation. Intraluminal lactate and glycerol concentrations were measured by microdialysis, and samples were collected for light microscopy and transmission electron microscopy. The histological changes were described and *** Using light microscopy, the jejunum was considered as viable until 6 h of ischemia, while with transmission electron microscopy the ischemic muscularis propria was considered viable until 5 h of ischemia. However, following ≥ 1 h of reperfusion, only segments that had been ischemic for ≤ 3 h appeared viable, suggesting a possible upper limit for viability in the porcine mesenteric occlusion model. Although intraluminal microdialysis allowed us to closely monitor the onset and duration of ischemia and the onset of reperfusion, we were unable to find sufficient level of association between tissue viability and metabolic markers to conclude that microdialysis is clinically relevant for viability assessment. Evaluation of color and motility appears to be poor indicators of intestinal *** Three hours of total ischemia of the small bowel followed by reperfusion appears to be the upper limit for viability in this porcine mesenteric ischemia model.
