Clinical applications of 5-aminolevulinic acid-mediated fluorescence for gastric cancer

5-aminolevulinic acid(ALA) is a naturally occurring amino acid that is a protoporphyrin IX(Pp IX) precursor and a next-generation photosensitive substance. After exogenous administration of ALA, Pp IX specifically accumulates in cancer cells owing to the impaired metabolism of ALA to Pp IX in mitochondria, which results in a red fluorescence following irradiation with blue light and the formation of singlet oxygen. Fluorescence navigation by photodynamic diagnosis(PDD) using ALA provides good visualization and detection of gastric cancer lesions and is a potentially valuable diagnostic tool for gastric cancer for evaluating both the surgical resection margins and extension of the lesion. Furthermore, PDD using ALA might be used to detect peritoneal metastases during preoperative staging laparoscopy, where it could provide useful information for the selection of a therapeutic approach. Another promising application for this modality is in the evaluation of lymph node metastases. Photodynamic therapy(PDT) using ALA to cause selective damage based on the accumulation of a photosensitizer in malignant tissue is expected to be a non-invasive endoscopic treatment for superficial early gastric cancer. ALA has the potential to be used not only as a diagnostic agent but also as a therapeutic drug, resulting in a new strategy for cancer diagnosis and therapy. Here, we review the current use of PDD and PDT in gastric cancer and evaluate its future potential beyond conventional modalities combined with a light energy upconverter, a light-emitting diode and nearinfrared rays as light sources.

Blue-light-induced rapid chloroplast de-anchoring in Vallisneria epidermal cells

In the outer periclinal cytoplasm of leaf epidermal cells of an aquatic angiosperm Vallisneria, blue light induces ＂chloroplast de-anchoring＂, a rapid decline in the resistance of chloroplasts against centrifugal force. Chloroplast de- anchoring is known induced within lmin of irradiation with high-fluence-rate blue light specifically, preceding the com- mencement of chloroplasts migration toward the anticlinal cytoplasm. However, its regulatory mechanism has remained elusive, although pharmacological analysis suggested that a calcium release from intracellular calcium stores is necessary for the response, in search of the responsible photoreceptors, immunoblotting analysis using antibodies against phototropins demonstrated that cross-reactive polypeptides of 120-kDa exist in the plasma-membrane fraction prepared from the leaves. In vitro phosphorylation analysis revealed that 120-kDa polypeptides were phosphorylated by exposure to blue light in a fluence-dependent manner. The blue-light-induced phosphorylation activity was sensitive to a Ser/Thr kinase inhibitor, staurosporine, and unusually was retained at a high level for a long time in darkness. Furthermore, phototropin gene homologs （Vallisneria PHOTOTROPINI and PHOTOTROPIN2） expressed in leaves were isolated. We propose that calcium- regulated chloroplast de-anchoring, possibly mediated by phototropins, is an initial process of the blue-light-induced avoidance response of chloroplasts in Vallisneria.

High circulating tumor cell concentrations in a specific subtype of gastric cancer with diffuse bone metastasis at diagnosis

AIM: To clarify the biological feature contributing to gastric cancer with diffuse bone metastases at ***: The participants visited the Department of Clinical Oncology, Akita University Hospital, from January 2014 to August 2015. The selection criterion for gastric cancer with diffuse bone metastases at diagnosis includes over 29 hot spots of bone scintigraphy. Circulating tumor cell were collected from 20 m L of peripheral venous blood drawn using a Cell Search kit and a Cell Tracks Auto Prep system by SRL, a clinical laboratory. The endpoints of this study were correlations between circulating tumor cells(CTC) count and therapeutic outcomes. RESULTS: Among 39 patients with gastric cancer, 5 patients met the criterion. The incidence of this subtype was 12.8%. CTC counts ranged from 235 to 6440 cells/7.5 m L of peripheral blood(median of 1724). These values were much higher than common gastric cancers(2 cells). In chemo-sensitive cases, CTC counts decreased within 14 d(median) from 275, 235 and 1724 to 2, 7 and 66, respectively. On the other hand, CTC counts increased after treatment failure or insensitive case from 2, 7 and 6440 to 787, 513 and 7885, respectively. The correlation between CTC count and survival time showed a trend, but did not reach significance(Y = 234.6- 0.03 X, P = 0.085).CONCLUSION: High CTC count is a biological hallmark of this subtype, and can be used as a direct and definitive indicator of therapeutic outcome.

Study on Preparation and Property of Poly-Aminosilicone-Rare Earth Composite

The poly-aminosilicone-rare earth composite was prepared by poly-aminosilicone cross-linked with rare earth and active silanol. The thermal stability of the composites was studied by thermogravimetric analysis (TG). Force condition of the composites in electric field was analyzed and relative polarizability was derived. It is found that the composites containing different rare earth ions have different relative polarizability. The experiment results reveal that organosilicon materials with different electrical performance can be obtained by this way. Meanwhile, the absorption and flourescene spectrum of composites were also investigated. Compared to rare earth chloride, the spectrum properties of the composite are changed obviously. The possible reasons for these phenomena were discussed.

Fatigue Resistance of Filled NR with PMMA-Wrapped and Rare Earth-Doped Alumina-Siloxane Gel

Poly (methyl methacrylate) (PMMA) was used to wrap alumina-siloxane sol which was produced by water glass, aluminum nitrate and α-methacrylic acid, and as a result, alumina-siloxane gel wrapped by PMMA was obtained. Meanwhile, rare earth ions were employed to dope in the course of reaction, and the formed rare earth doped PMMA microcapsule powder was filled into natural rubber (NR). It is found through the analysis of mechanical properties that Young′s modulus universally improves and a remarkable resistance to fatigue is displayed. Retention rate of tensile strength is twice that of the controlled sample after ten thousand times of extension fatigue.

Characterization of the genes involved in nitrogen cycling in wastewater treatment plants using DNA microarray and most probable number-PCR

To improve nitrogen removal performance of wastewater treatment plants （WWTPs）, it is essential to understand the behavior of nitrogen cycling communities, which comprise various microorganisms. This study characterized the quantity and diversity of nitrogen cycling genes in various processes of municipal WWTPs by employing two molecular-based methods：most probable number-polymerase chain reaction （MPN-PCR） and DNA microarray. MPN-PCR analysis revealed that gene quantities were not statistically different among processes, suggesting that conventional actwated sludge processes （CAS） are similar to nitrogen removal processes in their ability to retain an adequate population of nitrogen cycling microorganisms. Furthermore, most processes in the WWTPs that were researched shared a pattern：the nitS and the bacterial amoA genes were more abundant than the nirK and archaeal amoA genes, respectivelv. DNA microarray analysis revealed that several kinds of nitrification and denitrification genes were detected in both CAS and anaerobic-oxic processes （AO）, whereas limited genes were detected in nitrogen removal processes. Results of this study suggest that CAS maintains a diverse community of nitrogen cycling microorganisms; moreover, the microbial communities in nitrogen removal processes may be specific.

Beh et's disease complicated by multiple aseptic abscesses of the liver and spleen

Aseptic abscesses are an emergent entity and have been described in inflammatory bowel disease,especially in Crohn's disease,and in other ***,aseptic abscesses associated with Beh et's disease are extremely *** report a Japanese male diagnosed with an incomplete type of Beh et's disease who developed multiple aseptic abscesses of the spleen and *** 2002,the spleen abscesses were accompanied by paroxysmal oral aphthous ulcers and erythema *** the patient's response to antibiotic treatment was inadequate,a splenectomy was *** inflammatory cell infiltration,largely of polymorphonuclear neutrophils,was observed without evidence of bacterial or fungal *** the patient had no history of ocular symptoms or genital ulcers,a diagnosis of incomplete Beh et's disease was made according to the Japanese diagnostic criteria because of the presence of paroxysmal arthritis and epididymitis since *** 2005,multiple liver abscesses developed with right hypochondrial pain and seemed to be attributed to Beh et's disease because the abscesses yielded negative results during a microbiologic investigation and failed to go into remission under antibiotic *** prednisone(15 mg/d) was started in May 2006,and the abscesses dramatically disappeared 4 wk after *** the patient had a relapse of the liver abscesses in association with the tapering of prednisone,the augmentation of prednisone dosage yielded a *** abscesses of the liver and spleen were strongly suggested to be attributed to Beh et's *** should be aware of the existence of aseptic abscesses as uncommon manifestations of Beh et's disease.

Investigation of air pollution concentration in Kathmandu valley during winter season

The monthly concentrations of NO2, NOx, SO2 and O3 measured by a passive sampler from February 2003 to January 2004 showed that the air pollution during the winter season in Kathmandu valley was higher than the summer season. The O3 level was found the highest during April, May and June due to strong radiation. The hourly concentrations of NO2, NOx, O3 and suspended particulate matter（SPM） were also measured by automatic instruments on December 2003. Temperature at the height of 60 m and 400 m at Raniban Mountain in the northwest of Kathmandu valley was measured on February 2001 in the winter season and the average potential temperature gradient was estimated from observed temperature. Wind speed was also measured at the department of hydrology, airport section, from 18 February to 6 March 2001. It was found that the stable layer and the calm condition in the atmosphere strongly affected the appearance of the maximum concentrations of NO2 and SPM in the morning, and that the unstable layer and the windy condition in the atmosphere was considerably relevant to the decrease of air pollution concentrations at daytime. The emission amounts of NOx, HCs and total suspended particle（TSP） from transport sector in 2003 were estimated from the increasing rate of vehicles on the basis of the emission amounts in 1993 to be 3751 t/a, 30570 t/a and 1317 t/a, respectively. The diurnal concentrations in 2003 calculated by the two-layers box model reproduced the characteristics of air pollution in Kathmandu valley such as the maximum value of O3 and its time, the maximum value of NO in the morning, and the decrease of NO and NO2 at daytime. The comparison with the concentrations in 1993 calculated suggested that the main cause of air pollution was the emission from transport sector.

Impacts of converting from leaded to unleaded gasoline on ambient lead concentrations in Jakarta metropolitan area

Total suspended particulate mater （TSP） concentrations were monitored for one year from July 2000 and for one year from April 2003 in Jakarta City. Thirteen elemental TSP components, aluminum （Al）, sodium （Na）, iron （Fe）, lead （Pb）, potassium （K）, zinc （Zn）, titanium （Ti）, manganese （Mn）, bromine （Br）, copper （Cu）, chromium （Cr）, nickel （Ni）, and vanadium （V） were analyzed by a sequential X-ray fluorescence spectrometer. Al, Na, Fe, K, and Pb were major components at most of the sampling locations in 2000. However, only Pb in 2003 dramatically decreased to one tenth. The phase-out of leaded gasoline began on July 1, 2001 in Jakarta City and lead content in gasoline decreased to one tenth, too. The decrease in Pb concentration was a result of the phase-out of leaded gasoline, as lead emissions mainly are exhaust gas from vehicles.
