potato is the thirdmost important food crop in *** pathogens threaten sustainable crop production but can be controlled,in many cases,through the deployment of disease resistance genes belonging to the family of nucleotide-binding,leucine-rich-repeat(NLR)*** identify effective disease resistance genes in established varieties,we have successfully established SMRT-AgRenSeq in tetraploid potatoes and have further enhanced the methodology by including dRenSeq in an approach that we term *** inclusion of dRenSeq enables the filtering of candidates after the association analysis by establishing a presence/absence matrix across resistant and susceptible varieties that is translated into an F1 *** a SMRT-RenSeq-based sequence representation of the NLRome from the cultivar Innovator,SMRT-AgRenSeq-d analyses reliably identified the late blight resistance benchmark genes Rpi-R1,Rpi-R2-like,Rpi-R3a,and Rpi-R3b in a panel of 117 varieties with variable phenotype *** benchmark genes were identified with an F1 score of 1,which indicates absolute linkage in the *** method also identified nine strong candidates for Gpa5 that controls the potato cyst nematode(pCN)species Globodera pallida(pathotypes pa2/3).Assuming that NLRs are involved in controlling many types of resistances,SMRT-AgRenSeq-d can readily be applied to diverse crops and pathogen systems.

Scale-up of biomass conversion using 1-ethyl-3-methylimidazolium acetate as the solvent

This scale-up study demonstrated the feasibility of an ionic liquid(IL)pretreatment process at 40 kg scale,using the IL 1-ethyl-3-methylimidazolium acetate([C2C1Im][OAc])as the *** pretreatment was followed by enzymatic hydrolysis through which the process efficiency for biomass conversion to monomeric sugars was *** results show that 43 wt%of switchgrass was dissolved in IL after 2 h of pretreatment at 160℃ with 15 wt%solid loading.A 120 h enzymatic hydrolysis of the pretreated switchgrass results in 96%glucan and 98%xylan conversion.[C2C1Im][OAc]pretreatment has been successfully scaled up to 40 kg with improved sugar titers and yields relative to bench scale(6 kg).The mass flow of the overall process was established and the major scale-up challenges of the process were identified.

An Exploratory Analysis of the Market perspective on Reclaiming Chromated Copper Arsenate (CCA) from Decommissioned preservative-Treated Wood Utility poles

In the area of recycling of spent chromated copper arsenate (CCA)-treated wood, most studies to date have focused on methods of removing/extracting the residual preservative from the wood matrix. It is well recognized that exposure of CCA-treated wood to an acid solution can reverse the CCA fixation process thereby converting the CCA elements into their water-soluble form. The economic viability of the process is enhanced because it can be integrated with other technologies and products (e.g., “green” spray foam insulation, etc.). The market for the “green” CCA is the same as for traditional CCA-the wood treating industry, principally utility poles and pilings. A market research study was conducted to determine the suitability of spent CCA-treated wood as a source for recycled, “green” CCA for manufacturing “green” spray-foam insulation. Specifically, we wanted to discern the attitudes and overall perspectives of buyers/sellers (i.e., utilities and wood treating companies) of CCA preservatives and treated wood products, disposal methods and costs for decommissioned CCA-treated wood, and understand perceptions of and willingness-to-pay for “green” CCA preservatives extracted from the technologies used in this research. Results show that 60% of wood preservative treating respondents and 60% of electric utility company respondents are somewhat or greatly interested in using out-of-service utility poles as feedstock for “green insulation” as part of a new potential business venture.

Acute kidney injury(AKI)leads to unacceptably high mortality due to difficulties in timely intervention and less efficient renal delivery of therapeutic ***,a series of polyvinylpyrrolidone(pVp)-curcumin nanoparticles(pCurNp)are designed to meet the renal excretion threshold(~45 kDa),presenting a controllable delivery nanosystem for kidney *** accumulation of the relatively small nanoparticles,^(89)Zr-pCurNp M10 with the diameter between 5 and 8 nm,is found to be 1.7 times and 1.8 times higher than the accumulation of^(89)Zr-pCurNp M29(20-50 nm)and M40(20-50 nm)as revealed by pET ***,serum creatinine analysis,kidney tissues histology,and tubular injury scores revealed that pCurNp M10 efficiently treated cisplatin-induced ***,pCurNp offers a novel and simple strategy for precise pET image-guided drug delivery of renal protective materials.

Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is a novel beta-coronavirus that has emerged and caused a global pandemic of coronavirus disease 2019(COVID-19).1,2 Clinical manifestations of COVID-19 are age dependent,with children appearing to be less vulnerable than adults to severe disease.3,4 It has been reported that a subset of children and adults without previous infection had higher levels of humoral and cellular immunity to SARS-CoV-2,which may influence disease severity.5,6,7,8 Another study found that children who had never been exposed to SARS-CoV-2 had higher levels of cross-reactive antibodies to the spike glycoprotein subunit S2 of SARS-CoV-2 than adults.9 These findings raise the hypothesis that children may have a higher degree of preexisting cross-reactive immunity that provides some level of protection from severe *** this study,we determined antibody responses to SARS-CoV-2 in adults after infection and compared these responses in both children and adults with no prior exposure to ***,we mapped antibody epitopes elicited after infection,and cross-reactive antibody epitopes without prior SARS-CoV-2 infection,at high resolution using SARS-CoV-2 spike protein peptide arrays.

Evolution of pE35 and ppE68 Gene Families in Mycobacterium: Roles of Horizontal Gene Transfer and Evolutionary Constraints

Mycobacterium is a genus of bacteria with over a hundred non-pathogenic and pathogenic species, best recognized for certain members known to cause diseases such as tuberculosis and leprosy. Two novel protein families important in the pathogenesis of Mycobacterium species are the pE and ppE families. These two protein families affect the antigenic profiles, disturbing host immunity. To better understand the origin and evolution of these gene families and the differences in their composition between pathogenic and non-pathogenic strains, several bioinformatic analyses were conducted both among Mycobacterium and closely related species that contain pE35 and ppE68 gene homologs. The methods included protein homology searches (BLASTp), horizontal gene transfer analysis (IslandViewer), phylogenetic analysis, gene cluster analysis and structural and functional constraints. Results revealed that pE and ppE gene homologs were not only limited to Mycobacterium, but also existed in three other non-mycobacterial genera, Rhodococcus, Tsukamurella and Segniliparus, and were possibly initially acquired from non-mycobacterial microorganisms by multiple horizontal gene transfers. Results also demonstrated that pE and ppE genes were more diverse and more rapidly evolving in pathogenic Mycobacterium as compared with non-pathogenic Mycobacterium and other non-mycobacterial species. These findings possibly shed light on the diverse functions and origins of the pE/ppE proteins among these organisms.
