Multimodal nonlinear endomicroscopic imaging probe using a double-core double-clad fiber and focus-combining micro-optical concept

Multimodal non-linear microscopy combining coherent anti-Stokes Raman scattering,second harmonic generation,and two-photon excited fluorescence has proved to be a versatile and powerful tool enabling the label-free investigation of tissue structure,molecular composition,and correlation with function and disease *** a routine medical application,the implementation of this approach into an in vivo imaging endoscope is ***,this is a difficult task due to the requirements of a multicolour ultrashort laser delivery from a compact and robust laser source through a fiber with low losses and temporal synchronization,the efficient signal collection in epi-direction,the need for small-diameter but highly corrected endomicroobjectives of high numerical aperture and compact ***,we introduce an ultra-compact fiber-scanning endoscope platform for multimodal non-linear endomicroscopy in combination with a compact four-wave mixing based fiber *** heart of this fiber-scanning endoscope is an in-house custom-designed,single mode,double clad,double core pure silica fiber in combination with a 2.4 mm diameter NIR-dual-waveband corrected endomicroscopic objective of 0.55 numerical aperture and 180µm field of view for non-linear imaging,allowing a background free,low-loss,high peak power laser delivery,and an efficient signal collection in backward direction.A linear diffractive optical grating overlays pump and Stokes laser foci across the full field of view,such that diffraction-limited performance is demonstrated for tissue imaging at one frame per second with sub-micron spatial resolution and at a high transmission of 65%from the laser to the specimen using a distal resonant fiber scanner.

Cardiovascular diseases in general and atherothrombosis as the most common of its individualdisease entities is the leading cause of death in the developed ***,visualizationand characterization of inner arterial plaque composition is of vital diagnostic interest,especilly for the early recognition of vulnerable *** clinical techniques provide valuablemorphological information but cannot deliver information about the chemicai composition ofindividual ***,spectroscopic imaging techniques have recently drawn consider-able *** on the spectroscopic properties of the individual plaque components,as forinstance diferent types of lipids,the composition of atherosclerotic plaques can be analyzedqualitatively as well as ***,we compare the feasibility of multimodal nonlinearimaging combining,two-photon fluorescence(TPF),coherent anti-Stokes Raman scatering(CARS)and second-hamonic generation(SHG)microscopy to contrast composition and mor-phology of lipid deposits against the surrounding matrix of connective tissue with diffractionlimited spatial *** this contribution,the spatial distribution of major constituents of the arterial wall and atherosclerotic plaques like elastin,collagen,triglycerides and cholesterol can be simultaneously visualized by a combination of nonlinear imaging methods,providing a powerful label-free complement to standard histopathological methods with great potential forin vivo application.

Quantum Mechanics of a Quasi-Euclidean Space with Planck Length, Rotational Symmetry and Translational Symmetry

This work is focused on a quasi-Euclidean space with UV cutoff, IR cutoff and symmetries. Mathematical analysis reveals that the UV cutoff results in the minimum structures of space. Dominated by rotational symmetry, the structure should be a local one in situ or on a sphere. Investigations show that a 10D minimum structure is a non-local one with transformability between in-situ state and spherical state due to its special topology. Based on the quantum behaviors of the 10D structure controlled by translational symmetry, IR cutoff determines two long-range interactions with dimensionless constants of ~1/137.036 and ~1/1.628E+38, respectively.

We present a step-by-step approach for constructing a framework for knowledge process analysis （KPA）. We intend to apply this framework to the analysis of own research projects in an exploratory way and elaborate it through the accumulation of case studies. This study is based on a methodology consisting of knowledge process modeling, primitives synthesis, and reflective verification. We describe details of the methodology and present the results of case studies： a novel methodology, a practical work guide, and a tool for KPA; insights for improving future research projects and education; and the integration of existing knowledge creation theories.

The Famennian is a period of increased taxonomic diversity for the first representatives of the major plant groups,especially the spermatophytes (*** seed plants) that,today,represent the most abundant and diversified plant group on Earth [1,2].Early spermatophytes evolved a unique ovule-centered reproductive syndrome that allowed them to germinate and grow rapidly in disturbed habitats and,secondarily,to colonize dry habitats unfavorable to the reproduction of their free-sporing predecessors [3,4](Fig.1).In these ovulate organisms,the female gametophyte grows and produces gametes inside the indehiscent megasporangium (nucellus) that differentiates a pollen chamber retaining the pollen grains in its apical *** integument encloses the *** the Runcaria protovule that preceded them by 20 million years,Famennian ovules were all surrounded by a cupule,except perhaps Spermolithus and Warsteinia [5-7].

Effect of thrombectomy on oedema progression and clinical outcome in patients with a poor collateral profile

Background and purpose The impact of the cerebral collateral circulation on lesion progression and clinical outcome in ischaemic stroke is well ***,collateral status modifies the effect of endovascular treatment and was therefore used to select patients for therapy in prior *** purpose of this study was to quantify the effect of vessel recanalisation on lesion pathophysiology and clinical outcome in patients with a poor collateral *** and methods 129 patients who had an ischaemic stroke with large vessel occlusion in the anterior circulation and a collateral score(CS)of 0-2 were *** profile was defined using an established 5-point scoring system in CT *** progression was determined using quantitative lesion water uptake measurements on admission and follow-up CT(FCT),and clinical outcome was assessed using modified Rankin Scale(mRS)scores after 90 *** Oedema formation in FCT was significantly lower in patients with vessel recanalisation compared with patients with persistent vessel occlusion(mean 19.5%,95%CI:17%to 22%vs mean 27%,95%CI:25%to 29%;p<0.0001).In a multivariable linear regression analysis,vessel recanalisation was significantly associated with oedema formation in FCT(ß=−7.31,SD=0.015,p<0.0001),adjusted for CS,age and Alberta Stroke Program Early CT Score(ASPECTS).Functional outcome was significantly better in patients following successful recanalisation(mRS at day 90:4.5,IQR:2-6 vs 5,IQR:5-6,p<0.001).Conclusion Although poor collaterals are known to be associated with poor outcome,endovascular recanalisation was still associated with significant oedema reduction and comparably better outcome in this patient *** with poor collaterals should not generally be excluded from thrombectomy.

Oligodendrocyte precursor cells(OPCs)and microglia are two very fascinating cell types with a multitude of important but different *** a first glance,they appear not to share many cellular properties,nor are directly related to one another or derived from a common *** all differences,emerging data show that both cell types express the protein nerve/glial antigen 2(NG2)after pathological insults(Figure 1).For years,it remained controversial whether microglia really could express NG2 upon injury,with contradictory results reported among different disease *** this question,we could recently show by using triple transgenic knock-in mice and either an acute injury model(stab wound injury)or the middle cerebral artery occlusion combined with immunohistochemistry that a subset of microglia activates the cspg4 gene in a disease dependent manner leading to a bonafide microglia-specific NG2 protein expression besides OPCs and *** data show that the cspg4 gene not only gets transcribed in microglia based on reporter expression after recombination,but also the protein itself is expressed(Huang et al.,2020).
