Bioactive profile of borno brown Vigna unguiculata grains as influenced by pre-harvest synthetic chemicals

This study investigated the impact of pre-harvest synthetic chemical treatments on the post-harvest bioactive contents,pasting,colour,and thermal properties of borno brown ***(L.)Walp *** of borno brown ***(L.)Walp was planted on an already ploughed,harrowed and ridged plot of land with a portion cultivated as organic and another *** 90 days,mature grains were harvested and analyzed using standard *** showed that a total of 44.33 g bioactive compounds/100g grain was present in the organic borno brown *** grains whereas,20.56 g bioactive compounds/100g grain was present in the non-organic(control).The identified bioactive compounds in the organic grains included[heneicosane(4.50g/100g grain)];[12-Methyl-E,E-2,13-octadecadien-1-ol(4.09/100g grain)];[3-(2-Hydroxy-6-methylphenyl)-4(3H)-quinazolinone(7.40 g/100g grain)];[Methaqualone(9.08 g/100g grain)].The non-organic contained[heneicosane(4.07g/100g grain)];[3-(2-Hydroxy-6-methylphenyl)-4(3H)-quinazolinone(5.09 g/100g grain)];[Methaqualone(10.61 g/100g grain)].Flavonoids,alkaloids,total phenolic,2,2-diphenyl-1-picryl-hydrazl(DPPH)and ferric ion reducing antioxidant power(FRAP)contents of the organic grains were 19 mgRE/100g,3.40 mg/100g,1550 mgGAE/100g,2840μmolTEAC/100g,and 20.08 mmol Fe^(2+)/100g,*** non-organic grains had flavonoids(11.31 mgRE/100g),alkaloids(1.86 mg/100g),total phenolic(1110 mgGAE/100g),DPPH(1779μmolTEAC/100g)and FRAP(12.24 mmol Fe^(2+)/100g).The peak viscosity of 111.52 RVU,final viscosity(141.25 RVU)and pasting temperature(82.01℃)were obtained for the organic grains while non-organic had peak viscosity of 93.10RVU,final viscosity(95.20 RVU)and pasting temperature(80.10℃).The colour parameters L^(*),a^(*),b^(*)and ΔE were 73.10,0.68,14.47 and 12.98 for the organic and 84.25,0.30,21.10,and 0.00,respectively for non-organic *** thermal property showed enthalpy(ΔH)and peak temperature(T_(p))of 3.12 J/g and 78.25℃ f

Opening of liver mitochondrial permeability transition pore in streptozotocin-induced diabetic rats and its inhibition by methanol fraction of Ficus mucoso(Welw) root bark

Objective: Several pathologies arise from the inappropriate opening of the mitochondrial permeability transition(mPT) pore. In this regard, inhibition of mPT pore represents a cytoprotective approach to preserve mitochondrial function for treatment of diseases characterized by excessive tissue wastage such as diabetes mellitus. The aim of this study, therefore, was to study the effects of fractions of Ficus mucoso, a medicinal plant used in the traditional treatment of diabetes, on mPT pore in normal and streptozotocin(STZ)-induced diabetic rat ***: Different solvent fractions of the crude methanol extract of F. mucoso were obtained by vacuum liquid chromatography and were tested on the mPT pore. Of all the fractions tested, methanol fraction of F. mucoso(MFFM) was the most potent and was used for in vivo studies. Diabetes mellitus was induced by a single intraperitoneal injection of 60 mg/kg STZ, while treatment lasted for 14 d. In vivo, 30 male Wistar rats were divided into five groups: A, normo-glycemic control(distilled water);B, STZ(65 mg/kg;diabetic control);C, STZ + MFFM(25 mg/kg);D, STZ + MFFM(50 mg/kg);E, STZ + glibenclamide(5 mg/kg). The mPT, mitochondrial ATPase activity, lipid peroxidation and cytochrome c release were assessed spectrophotometrically while blood glucose levels were monitored using ***: In vitro, the solvent fractions of F. mucoso, at all concentrations tested, had no effect on the mPT pore, in the absence of calcium, with no significant release of cytochrome c. Interestingly, calciumdependent pore opening was inhibited by all solvent fractions of F. mucoso, with the MFFM having the highest inhibitory effect of 83% at 3 mg/mL. Nutritional assessment,glycemic indices and anti-diabetic potentials of dough meal generated from optimized blends of matured plantain,soya cake and wheat bran flours

Management of blood glucose levels associated with type 2 diabetes using nutrients derived from food without having a harmful effect on one’s health is gradually gaining research *** dough meal blends were developed using optimal mixture design of response surface methodology(RSM).The optimum blends based on the protein and crude fibre contents were as follows:PSW1:mature plantain 70.0%+soya cake 25.0%+wheat bran 5.0%;PSW2:mature plantain 68.0%+soya cake 23.0%+wheat bran 9.00%;PSW3:mature plantain 69.0%+soya cake 24.0%+wheat bran 7.0%,while 100% mature plantain flour(PLNT)and cerolina were used as *** nutritional value,amino acid profile,pasting properties,α-amylase,α-glucosidase inhibition potential,in-vitro glycemic index and consumer acceptability were determined using standard *** showed that the inclusion of soya cake and wheat bran significantly increased(P<0.05)protein 14.08 g/100 g and fibre 3.02 g/100 g as *** higher(P<0.05)branched chain and aromatic amino acids were observed in the composite dough meal compared to *** peak viscosity(Pv),trough viscosity(Tv)and final viscosity(Fv)are lower in dough meal blends with increase in incorporation of soya cake and wheat ***3 showed the highest essential amino acid index,predicted biological value,α-glucosidase,α-amylase inhibition and lowest estimated glycaemic index at(P≤0.05).Hence,the study established that PSW3 showed the best potentials to modulate blood sugar level.

Hypoglycemic efficacy of Ocimum gratissimum and Vernonia amygdalina compared with insulin and glibenclamide in type Ⅰ and type Ⅱ diabetic rat models

Objective: To compare the efficacy of Ocimum gratissimum (O. gratissimum) and Vernonia amygdalina (V. amygdalina) with those of insulin and ***: Type Ⅰ and Ⅱ diabetes mellitus (DM) were induced by a single intraperitoneal injection of 65 mg/kg of streptozotocin and intraperitoneal administration of nicotinamide (100 mg/kg) along with streptozotocin, respectively. The state of diabetes was confirmed weekly by testing blood glucose level using a ***: The weekly blood glucose levels were higher in type I DM than in type Ⅱ DM. Type Ⅰ DM plus O. gratissimum showed a weekly progressive significant reduction in blood glucose compared to type Ⅰ DM control. Type Ⅰ DM control showed a duration dependent significant higher blood glucose concentration compared to normal control. Type I DM plus V. amygdalina also showed a time dependent significant lower glucose level compared to normal control and type Ⅰ DM control. Combination treatment of type Ⅰ DM (O. gratissimum plus V. amygdalina) showed a significantly elevated glucose concentration compared to normal control which was similar to type I DM control. Insulin treatment in type I DM showed a weekly progressive significant reduction of glucose concentration compared to normal control and type I DM control. Type Ⅱ DM control showed a fairly constant blood glucose concentration throughout the duration of treatment that was significantly higher than that of the normal control. Type Ⅱ DM plus O. gratissimum showed a fairly steady significant reduction of glucose concentration compared to type Ⅱ DM control and normal control. Type Ⅱ DM plus V. amygdalina also showed a fairly constant significant reduction of glucose concentration compared to type Ⅱ DM control and normal control. Integrated Geophysical and Geochemical Methods for Environmental Assessment of Municipal Dumpsite System

Leachate originating from open refuse dumpsite systems can be delineated through an integration of qualitative and quantitative methods. This study was designed to examine extent of leachate and pollution from one of the numerous open refuse dumpsites in Lagos metropolis. Qualitative assessment was determined using electrical resistivity tomography (ERT), vertical electrical sounding (VES) and induced polarization geophysical methods. Both ERT and VES methods revealed persistent low resistivity (1 - 20 Ωm) of leachate to the depth above 35 m. The two methods were projected to produce 3-D view of the site which shows a NW-SE flow pattern of the leachate and possibly, the groundwater. IP values observed over the polluted zone was -2.9 - 8 ms, indicating a sandy layer. Quantitative assessment was achieved by analysis of geochemical substances in the water samples taken from wells and boreholes in the precinct of the dumpsite. Here, we examine the macroelements, salts (sulphates, nitrates and chlorides), heavy metals, radioactive metals contents and physical parameters of the water samples. The analyses reveal the presence of these substances in the water and their strong correlations justified the provenance as the same. As part of the quantitative evaluation, physical parameters (pH, TDS, DO, salinity, total hardness, turbidity, electrical conductivity EC and temperature) of the water samples were also determined. The samples pH plotted in the acidic domain unsuitable for human consumption. Leachate flow direction was generated from the decreasing concentration of measured parameters (geochemical elements and physicals properties) in NW-SE direction which agrees with similar flow pattern deduced from ERT results.

Nutrient and Anti-Nutrient Content of Soy-Enriched Tapioca

Tapioca is a cassava-based food product made in the form of irregular lumps of partly gelatinized starch grits. Tapioca was enriched with varying proportions of soybean flour (0, 85% - 15%, 75% - 25%, 50% - 50%) to produce Soy-tapioca (ST). Nutrient and anti-nutrient composition of ST were determined by standard methods, while sensory evaluation was also carried out. Results showed significant increase in protein, fat and mineral contents of tapioca as the level of soy-substitution increased. There was a decrease in the cyanogenic potential and an increase in the level of trypsin inhibitor as soy-substitution increased. Tannin, phytic acid and oxalate contents of the soy-tapioca samples were below minimum levels of safety. ST (50% - 50%) was more nutrient dense than the other products, but ST (85% - 15%) was more accepted in terms of colour, taste and texture. ST suggests a safe, nutritious and acceptable food product that can enhance food and nutrition security among cassava consuming populations.
