Tumor suppressor p53-mediated cell cycle arrest and DNA damage repair may exert cytoprotective effects against cancer therapies, including WEE1 inhibition. Considering that p53 activation can also lead to multiple types of cell death, the role of this tumor suppressor in WEE1 inhibitor-based therapies remains disputed. In this study, we reported that nucleolar stress-mediated p53 activation enhanced the WEE1 inhibitor AZD1775-induced ferroptosis to suppress lung cancer growth. Our findings showed that AZD1775 promoted ferroptosis by blocking cystine uptake, an action similar to that of Erastin. Meanwhile, inhibition of WEE1 by the WEE1 inhibitors or si RNAs induced compensatory upregulation of SLC7A11, which conferred resistance to ferroptosis. Mechanistically, AZD1775 prevented the enrichment of H3K9me3, a histone marker of transcriptional repression, on the SLC7A11 promoter by repressing the expression of the histone methyltransferase SETDB1, thereby enhancing NRF2-mediated SLC7A11 transcription. This finding was also validated using the H3K9me3 inhibitor BRD4770. Remarkably, we found that the nucleolar stress-inducing agent Actinomycin D(Act. D) inhibited SLC7A11 expression by activating p53, thus augmenting AZD1775-induced ferroptosis. Moreover, the combination of AZD1775 and Act. D synergistically suppressed wild-type p53-harboring lung cancer cell growth both in vitro and in vivo. Altogether, our study demonstrates that AZD1775 promotes ferroptosis by targeting cystine uptake but also mediates the adaptive activation of SLC7A11 through the WEE1-SETDB1 cascade and NRF2-induced transcription, and inhibition of SLC7A11 by Act. D boosts the anti-tumor efficacy of AZD1775 by enhancing ferroptosis in cancers with wild-type p53.

Solar flare prediction is an important subject in the field of space *** learning technology has greatly promoted the development of this *** this study,we propose a novel solar flare forecasting model integrating Deep Residual Network(ResNet)and Support Vector Machine(SVM)for both≥C-class(C,M,and X classes)and≥M-class(M and X classes)*** collected samples of magnetograms from May 1,2010 to September 13,2018 from Space-weather Helioseismic and Magnetic Imager(HMI)Active Region Patches and then used a cross-validation method to obtain seven independent data *** then utilized five metrics to evaluate our fusion model,based on intermediate-output extracted by ResNet and SVM using the Gaussian kernel *** results show that the primary metric true skill statistics(TSS)achieves a value of 0.708±0.027 for≥C-class prediction,and of 0.758±0.042 for≥M-class prediction;these values indicate that our approach performs significantly better than those of previous *** metrics of our fusion model’s performance on the seven datasets indicate that the model is quite stable and robust,suggesting that fusion models that integrate an excellent baseline network with SVM can achieve improved performance in solar flare ***,we also discuss the performance impact of architectural innovation in our fusion model.

In the field of perovskite solar cells(PSCs), the research on defects in the buried interface has been relatively limited due to its non-exposure; however, this interface significantly impacts the performance enhancement of inverted PSCs. This study employs phenylethylammonium chloride(PEACl) molecules as a buffer layer to modify the buried interface of p-i-n structured PSCs,aiming to enhance the uniformity of self-assembled monolayers(SAMs) and facilitate the uniform nucleation and growth of perovskite films on the substrate. Furthermore, the introduction of the PEACl buffer layer effectively passivates defects at the bottom of the perovskite layer and notably enhances the crystal quality of the perovskite film by mitigating residual stress,thereby reducing nonradiative recombination loss. Following these optimizations, the MA-free PSCs treated with PEACl achieve a power conversion efficiency(PCE) of 24.11%, with significant improvements in storage, thermal stability, and operational stability. Particularly noteworthy is the device's performance in an unencapsulated state, whereas after 1,500 hours of continuous light operation stability testing, it retains 97% of its original efficiency. This study not only enriches the systematic understanding of the characteristics of the buried interface in PSCs but also contributes significantly to advancing the commercial production of perovskite photovoltaic technology.

The complete decomposition of formaldehyde(HCHO) at ambient temperature is the most potential strategy for HCHO elimination from indoor ***,extra low content of Pt nanoparticles(0.025 wt%)supported on water-solubility carbon nitride/ceria(Pt/gC_(3)N_(4)@CeO_(2)) was prepared for gaseous HCHO removal at ambient temperature in a simulated indoor *** light(8 W) illumination could visibly boost the complete decomposition of HCHO into CO_(2) over Pt/gC_(3)N_(4)@CeO_(2).The cooperative effect in the distinct heterostructure and plenty of surface reactive oxygen species contribute primarily to the enhanced catalytic performance of Pt/g-C_(3)N_(4)@CeO_(2).Moreover,the possible mechanism of HCHO oxidation over Pt/g-C_(3)N_(4)@CeO_(2) assisted by the fluorescent light irradiation was proposed based on the physicochemical and optical characterization as well as the result of in situ diffuse reflectance infrared Fourier transform *** work might shed some light on the potential application of the versatile catalysts for ambient-temperature catalytic decomposition of HCHO by making full use of the indoor energies.

Enhanced performances of polypropylene membranes by molecular layer deposition of polyimide

Molecular layer deposition （MLD） for the deposition of polyimide （PI） at low temperature of 110 ℃ has been firstly introduced into the field of membrane separation. With the optimized MLD deposition parameters, such low de- position temperature has successfully expanded the application of MLD for the surface modification of polymeric materials. Globular PI particulates grow on both the free surfaces as well as the pore walls of the polypropylene （PP） membranes as isolated islands during progressive precursor exposures. The PI-deposited PP membranes ex- hibit synergistically improved performances in various aspects. Evidently improved surface hydrophilicity and per- meation performance （30%） have been achieved v/a the MLD deposition of polyimide films. The overall separation efficiency maintained higher than 85% even after 250 cycles of MLD deposition. More importantly, the thermal sta- bility has been improved and the integrity of the porous structure for PI-deposited PP membranes has been well preserved even after harsh treatment, which ensures its potential application in industries.

Body misperception plays an important role in the development of weight and dietary disorders among children and adolescents.A school-based health promotion program(2014-2015)was conducted to promote the school health education and improve the teenagers'physical health among Chinese children and *** on this program,we intended to examine weight status and weight misperception among Chinese children and adolescents and to explore the relationship between weight misperception and lifestyle behaviors.A total of 10708 Chinese children and adolescents in 3rd and 7th grade from Shandong and Qinghai province participated in the *** participants,dietary and activity patterns were clustered by latent class analysis(LCA).Logistic regression analysis was undertaken to explore the relationship between weight perception and demographic factors or dietary and activity *** the gender-specific difference of children and adolescents,analyses were separately conducted among boys and *** total prevalence of weight misperception was 44.50%.Boys,especially those in higher grade and living in wealthier district,were more likely to misperceive body *** were more likely to overestimate their weight(26.10%)while boys tended to underestimate the weight(28.32%).Three latent dietary and activity patterns including obesogenic pattern,malnourished pattern and healthy pattern were *** participants who had weight misperception were more likely to choose unhealthy dietary and exercise *** high prevalence of weight misperception was closely related to the unhealthy weight pattern and unhealthy dietary or exercise *** research found that most children and adolescents failed to perceive their weight correctly and boys tended to underestimate their weight while girls were subjected to ***,comprehensive intervention programs should focus on improving self-weight awareness,and appropriate guidance should be made to

Photosynthesis is involved in the essential process of transforming light energy into chemical *** the interaction between photosynthesis and the circadian clock has been confirmed,the mechanism of how light intensity affects photosynthesis through the circadian clock remains ***,we propose a first computational model for circadian-clock-controlled photosynthesis,which consists of the light-sensitive protein P,the core oscillator,photosynthetic genes,and parameters involved in the process of *** model parameters were determined by minimizing the cost function(δ=8.56),which is defined by the errors of expression levels,periods,and phases of the clock genes(CCA1,PRR9,TOC1,ELF4,GI,and RVE8).The model recapitulates the expression pattern of the core oscillator under moderate light intensity(100μmol m^(-2) s^(-1)).Further simulation validated the dynamic behaviors of the circadian clock and photosynthetic outputs under low(62.5μmol m^(-2) s^(-1))and normal(187.5μmol m^(-2) s^(-1))*** exposed to low light intensity,the peak times of clock and photosynthetic genes were shifted backward by 1–2 hours,the period was elongated by approximately the same length,and the photosynthetic parameters attained low values and showed delayed peak times,which confirmed our model *** study reveals a potential mechanism underlying the circadian regulation of photosynthesis by the clock under different light intensities in tomato.
