Sensitivity of Different Cytotoxic Responses of Vero Cells Exposed to Organic Chemical Pollutants and their Reliability in the Bio-toxicity Test of Trace Chemical Pollutants

Objective To find a sensitive cytotoxic response to reflect the bio-toxicity of trace organic pollutants, the sensitivity and reliability of morphological change and proliferation inhibition of Vero cells exposed to 2, 4, 6-trichlorophenol （TCP） and the leachate from products related to drinking water （PRDW） were compared, and the mechanism of the morphological change in Vero cells exposed to chemical pollutants was studied. Methods Vero cells were treated by different concentration of TCP and the leachate from PRDW. Methylthiazol-2-yl-2, 5-diphenyl tetrazolium bromide （MTT） assay was carried out for proliferation inhibition. Bioluminescence method was carried out as another method to test the toxicity of TCP. Flow Cytometry assay was used to test cell Apoptosis and damage of cell-membrane. Results 0.25 mg/L TCP had an effect on cell morphology, and the proportion of morphologically changed cells increased with increasing TCP concentration. At low TCP concentrations, inhibition of cell proliferation did not seem to correlate to TCP concentration, and was negative when TCP concentration was 〈1.0 mg/L. After exposure to leachate from PRDW extracted at different temperatures, the percentage of morphologically changed cells increased with extracting temperature, but the inhibition of cell proliferation failed to reflect the correlation between extracting temperature and proliferation inhibition of Vero cells. Although the Sensitivity of bioluminescence method seems to be similar to morphological change in Veto cells, the bacterial in this method is not homologous enough with human body cells to reflect the toxicity to human body. These imply cell morphological change is a more sensitive and reliable method to reflect bio-toxicity of organic pollutants than proliferation inhibition. An Association between Immunosenescence and CD4^+CD25^+ Regulatory T Cells: A Systematic Review

Objective Age-related increment of the prevalence of CD4^＋CD25^＋ regulatory T （Treg） cells were described controversially, and whether such changes explain immune dysfunction in the elderly is still unclear. The aim of this systematic review is to evaluate the role of the Tregs in immunosenescence. Methods Medline and manual searches were performed to identify all published epidemiological and animal studies investigating the efficacy of the association between immunosenescence and Treg cells. Results It was founded that the frequency, phenotypic characteristics, and number/function of Tregs were altered significantly with aging. Medical conditions in individuals with advanced ageas well as apoptosis intensity of Treg cells had an impact on the accumulation of Tregs which in turn could deteriorate cytotoxic activity of CD8＋ T and NK cells and production of IL-2. The range of immune cells that could be suppressed by Treg cells was quite wide and covered CD4^＋CD25^＋ T cells, NK cells, dendritic cells and even monocytes. These changes were observed both in humans and experimental animals. Besides, it was believed that frequency of Tregs increased with age and was accompanied by intensified suppressive activity for Tregs in patients, for example, with Alzheimer disease （AD） and Parkinson disease （PD）. The impaired condition of CD4＋ T cells, so-called immunosenescence, rendered transplant recipients less responsive to an allogeneic kidney graft, an effect that was limited to transplant recipients who were aged over 60 years. Conclusions Treg cells are associated with immunosenescence. All these changes contribute to the aging-related decline of immune responses and lead to the higher risk of immune-mediated diseases, cancer or infections in aged individuals.

Temporal changes of symbiont density and host fitness after rifampicin treatment in a whitefly of the Bemisia tabaci species complex

Microbial symbionts are essential or important partners to phloem-feeding insects. Antibiotics have been used to selectively eliminate symbionts from their host insects and establish host lines with or without certain symbionts for investigating functions of the symbionts. In this study, using the antibiotic rifampicin we attempted to selectively eliminate certain symbionts from a population of the Middle East-Asia Minor 1 whitefly of the Bemisia tabaci species complex, which harbors the primary symbiont ＂Candidatus Portiera aleyrodidarum＂ and two secondary symbionts ＂Candidatus Hamiltonella defensa＂ and Rickettsia. Neither the primary nor the secondary symbionts were completely depleted in the adults （F0） that fed for 48 h on a diet treated with rifampicin at concentrations of 1 100/zg/mL. However, both the primary and secondary symbionts were nearly completely depleted in the offspring （F 1） of the rifampicin-treated adults. Although the F1 adults produced some eggs （F2）, most of the eggs failed to hatch and none of them reached the second instar, and consequently the rifampicin-treated whitefly colony vanished at the F2 generation. Interestingly, quantitative polymerase chain reaction assays showed that in the rifampicin-treated whiteflies, the density of the primary symbiont was reduced at an obviously slower pace than the secondary symbionts. Mating experiments between rifampicin-treated and untreated adults demonstrated that the negative effects ofrifampicin on host fitness were expressed when the females were treated by the antibiotic, and whether males were treated or not by the antibiotic had little contribution to the negative effects. These observations indicate that with this whitefly population it is not feasible to selectively eliminate the secondary symbionts using rifampicin without affecting the primary symbiont and establish host lines for experimental studies. High-pressure-induced phase transition in cinchomeronic acid polycrystalline form-I

Diamond anvil cells combined with Raman spectroscopy and angle-dispersive x-ray diffraction(ADXRD) were used to investigate the compression behavior of cinchomeronic acid(C_7H_5NO_4, CA), a hydrogen-bonded polymorphs. The compression of form-I at approximately 6.5 GPa caused an irreversible phase transition that produced the new polymorph form-III. Lattice and internal modes in the Raman spectra were analyzed to determine the modifications in the local environment of CA form-I molecules. The form-III was indexed and refined to a low-symmetry triclinic structure with space group P1. The mechanism for the phase transition involved the reconstructions in the hydrogen-bonded networks in CA form-I.

Adult idiopathic thrombocytopenic purpura （ITP） is a .chronic acquired organ-specific autoimmune hemorrhagic disease characterized by the production of auto-antibodies against antigens on the membranes of platelet, resulting in enhanced Fc-mediated destruction of the platelets by macrophages in the reticuloendothelial 1-3 system. Splenectomy remains the best treatment of chronic ITP, but approximately 30% of patients do not respond or relapse after surgery;4-6 these patients are defined as patients with chronic refractory ITE There is no consensus on an appropriate sequence of treatments for patients with chronic refractory ITE79 Management of these patients remains a dilemma. We have treated 31 patients with chronic refractory ITP using low dose combination chemotherapy between 1994 and 2005, and the results are presented as follows.
