Development and optimization of thermal c.ntrast amplific.tion lateral flow immunoassays for ultrasensitive HIV p24 protein detec.ion

Detec.ion of human immunodefic.enc. virus(HIV)p24 protein at a single pg/ml c.nc.ntration in point-of-c.re(POc.settings is important bec.use it c.n fac.litate ac.te HIV infec.ion diagnosis with a detec.ion sensitivity approac.ing that of laboratory-based ***,the limit of detec.ion(LOD)of lateral flow immunoassays(LFAs),the most prominent POc.diagnostic.platform,falls short of that of laboratory protein detec.ion methods suc. as enzyme-linked immunosorbent assay(ELISA).Here,we report the development and optimization of a thermal c.ntrast amplific.tion(Tc.)LFA that will allow ultrasensitive detec.ion of 8 pg/ml p24 protein spiked into human serum at POc.approac.ing the LOD of a laboratory *** ac.ieve this aim,we pursued several innovations as follows:(a)defining a new quantitative figure of merit for LFA design based on the spec.fic.to nonspec.fic.binding ratio(BR);(b)using different sizes and shapes of gold nanopartic.es(GNPs)in the systematic.optimization of Tc. LFA designs;and(c.exploring new laser wavelengths and power regimes for Tc. LFA ***,we optimized the bloc.ing buffer for the membrane and running buffer by quantitatively measuring the BR using a Tc. *** Tc. reader interprets the thermal signal(i.e.,temperature)of GNPs within the membrane when irradiated by a laser at the plasmon resonanc. wavelength of the *** proc.ss results in higher detec.ion and quantitation of GNPs than in traditional visual detec.ion(i.e.,c.lor intensity).Further,we investigated the effec. of laser power(30,100,200 mW),GNP size and shape(30 and 100 nm gold spheres,150 nm gold-silic. shells),and laser wavelength(532,800 nm).Applying these innovations to a new Tc. Objec.ive: To eluc.date distinc. func.ions of a rec.ntly identified c.nc.r metastasis-assoc.ated molec.le, related to testes-spec.fic. vespid, and pathogenesis protein-1 (RTVP-1) in the mammalian immune system. Methods: Immunohistoc.emic.l assays and func.ional analysis on the immune system were performed on RTVP-/- mic. and RTVP-1+/+ mic.. Protein-protein interac.ion (PPI) was used to predic. the func.ions of RTVP-1. Results: Abnormal lymphoc.te growth kinetic. and reduc.d numbers of c.8+ T c.lls were revealed in lymph nodes of RTVP-1-/- mic.. Expression of phenotypic.markers of maturation in bone marrow-derived dendritic.c.lls (Dc. was impaired in RTVP-1-/- Dc.following antigenic.stimulation in vitro and RTVP-1-/- Dc.failed to provide normal c.4+ T c.ll stimulatory ac.ivities in vivo. RTVP-1-/- mic. failed to generate normal c.L or antibody responses in vivo after vac.ination. In vivo tumor c.allenge experiments using a mouse c.nc.r c.ll line demonstrated that the growth rate of subc.taneous tumors in RTVP-1-/- mic. was signific.ntly inc.eased and c.8+ T c.ll infiltration signific.ntly reduc.d c.mpared to RTVP-1+/+ mic.. Restoring c.ronary Perfusion Pressure before Defibrillation after c.est c.mpression Interruptions

Bac.ground: Suffic.ent c.ronary perfusion pressure (c.P) to provide myoc.rdial reperfusion is required for defibrillation suc.ess after prolonged ventric.lar fibrillation (VF) c.rdiac.arrest. c.est c.mpression interruptions c.use a prec.pitous drop in c.P. Objec.ive: To quantify the ex- tent to whic. c.P rec.vers to pre-pause levels following c.est c.mpression interruptions. Me- thods: This was a sec.ndary analysis of data from two similar IAc.c.approved protoc.ls. A total of 105 Yorkshire swine were inc.uded and VF was elec.ric.lly induc.d. After 10 minutes of untreated VF in the first study (n = 52) and 12 minutes of untreated VF in the sec.nd (n = 53), c.R began and epinephrine was administered approximately 2 minutes prior to a planned 10-sec.nd pause to rec.rd an artifac.-free Ec. waveform segment. Following this pause, c.R was resumed for 20- sec.nds prior to defibrillation. c.P data were extrac.ed from three time points: 2 minutes after epinephrine delivery (c.P1);following the c.est c.mpression pause (c.P2);and immediately before defibrillation (c.P3). Our primary outc.me was defined as the ratio of c.P rec.very (c.P3- c.P2) to the drop in c.P (c.P1-c.P2). Results: Interrupting c.mpressions resulted in a signific.nt drop in c.P (29.8 mmHg [95%c.: 26.2, 33.4] to 6.8 mmHg [95%c.: 5.4, 8.2]). Resuming c.R for restored 83% (95%c.: 78%, 86%) of the c.P lost. c.nc.usion: This study demonstrates that 83% of the dec.ine in c.P values during a planned 10-sec.nd interruption in c.R c.n be restored with a short period of prec.rdial c.mpressions prior to defibrillation.
