BACKGROUND:Th.s study aimed to evaluate th. discriminatory performance of 11 vital sign-based early warning scores(EWSs)and th.ee sh.ck indices in early sepsis prediction in th. emergency department(ED).METh.DS:We performed a retrospective study on consecutive adult patients with.an infection over 3 month. in a public ED in h.ng *** primary outcome was sepsis(Sepsis-3 definition)with.n 48 h.of ED *** c-statistics and th. Transient and stable expression of transgenes is central to many investigations in plant biology research. Ch.mical regulation of expression can circumvent problems of plant leth.lity caused by constitutive overexpression or allow inducible knock （out/down） approach.s. Several ch.mically inducible or repressible systems h.ve been described and successfully applied. h.wever, cloning and application-specific modification of most available inducible expression systems h.ve been limited and remained complicated due to restricted cloning options. h.re we describe a new set of 57 vectors th.t enable transgene expression in transiently or stably transformed cells. All vectors h.rbor a synth.tically optimized XVE expression cassette, allowing I^-estradiol mediated protein expression. Plasmids are equipped with.th. reporter genes GUS, GFP, mCh.rry, or with.h. and Strepll epitope tags and h.rbor an optimized multiple cloning site for flexible and simple clon- ing strategies. Moreover, th. vector design allows simple substitution of th. driving promoter to ach.eve tissue-specificity or to modulate expression ranges of inducible transgene expression. We report details of th. kinetics and dose-dependence of expression induction in Arabidopsis leaf mesoph.ll protoplasts, transiently transformed Nicotiana benth.miana leaves, and stably transformed Arabidopsis plants. Using th.se vectors, we investigated th. influence of CBL （Calcineurin B-like） protein expression on th. subcellular localization of CIPKs （Calcineurin B-like interacting protein kinases）. Th.se analyses uncovered th.t induced co-expression of CBL3 is fully sufficient for dynamic translocation of CIPK5 from th. cytoplasm to th. tonoplast. Th.s, th. vector system presented h.re facilitates a broad range of research.applications.

Maidanetske study area,close to Uman City of Ukraine,were analysed to evaluate th. effect of soil organic carbon(SOC)on th. parameters related to soil microstructural stability derived from *** organic carbon was removed with.h.(2)O_(2)to determine th. soil texture-dependent values of soil rh.ological properties,wh.ch.were compared to th. values determined for samples with.intact water-stable *** sh.ar resistance-related parameters increased for aggregated soil samples compared to SOC-free soil samples,indicating an increase in soil stability due to *** contrast,th. values of th. overall viscoelasticity and th. sh.ar strain were reduced for aggregated soil samples,indicating decreased soil *** functions were applied to predict th. sh.ar strain-dependent loss and storage moduli and sh.ar stress values as a function of SOC *** particles(630–2000μm)and volumetric water content improved th. *** conclude th.t increased SOC content,th.ough.th. gluing and cementing effects of SOC and altered aggregate sh.pes compared to SOC-free soil materials,contributes to h.gh.r microstructural ***,th. increased water content in th. SOC-containing soil samples reversed soil strength.ning *** was due to th. fact th.t a more rapid increase in positive water pressure under sh.ar stress weakened th. samples and th. sph.rical aggregates began to rotate more easily,th.s loosing energy,wh.n compared to platy particles of th. SOC-free soil materials.

In th.s paper, we sh.w th.t supercritical fluids h.ve a greater significance in th. generation of pegmatites,and for ore-forming processes related to granites th.n is usually assumed. We sh.w th.t th. supercritical melt or fluid is a silicate ph.se in wh.ch.volatiles; principally h.2O are completely miscible in all proportions at magmatic temperatures and pressures. Th.s ph.se evolves from felsic melts and ch.nges into h.droth.rmal fluids, and its unique properties are particularly important in sequestering and concentrating low abundance elements, such.as metals. In our past research. we h.ve focused on processes observed at upper crustal levels, h.wever extensive work by us and oth.r research.rs h.ve demonstrated th.t supercritical melt/fluids sh.uld be abundant in melting zones at deep-crustal levels too. We propose th.t th.se fluids may provide a connecting link between lower and upper crustal magmas,and a h.gh.y efficient transport mech.nism for usually melt incompatible elements. In th.s paper, we explore th. unique features of th.s fluid wh.ch.allow th. partitioning of variouselements and compounds, potentially up to extreme levels,and may explain various features both.of mineralization and th. magmas th.t produced th.m.

Induction and mobilization of transposable elements （TEs） following DNA damage or oth.r stresses h.s been reported in prokaryotes and eukaryotes. Recently it was discovered th.t eukaryotic TEs are frequently associated with.long non-coding RNAs （IncRNAs）, many of wh.ch.are also upregulated by stress. Yet, it is unknown wh.th.r DNA damage-induced transcriptional activation of TEs and IncRNAs occurs sporadically or is a synch.onized, genome-wide response. h.re we investigated th. transcriptome of Arabidopsis wild- type （WT） and ataxia telangiectasia mutated （atm） mutant plants 3 h.after induction of DNA damage. In WT, expression of 5.2% of th. protein-coding genes is 〉 2-fold ch.nged, wh.reas in atm plants, only 2.6% of th.se genes are regulated, and th. response of genes associated with.DNA repair, replication, and cell cy- cle is largely lost. In contrast, only less th.n 0.6% of TEs and IncRNAs respond to DNA damage in WT plants, and th. regulation of 〉95% of th.m is ATM-dependent. Th. ATM-downstream factors BRCA1, DRM1, JMJ30, AGO2, and th. ATM-independent AGO4 participate in th. regulation of individual TEs and IncRNAs. Remarkably, protein-coding genes located adjacent to DNA damage-responsive TEs and IncRNAs are frequently coexpressed, wh.ch.is consistent with.th. h.poth.sis th.t TEs and IncRNAs located close to genes commonly function as controlling elements.

Grain quality is an important agricultural trait th.t is mainly determined by grain size and ***, we ch.racterize th. role of th. rice transcription factor （TF） SALT-RESPONSIVE ERF1 （SERF1） during grain develop-ment. Th.ough.genome-wide expression profiling and ch.omatin immunoprecipitation, we found th.t SERF1 directlyregulates RICE PROLAMIN-BOX BINDING FACTOR （RPBF）, a TF th.t functions as a positive regulator of grain filling. Lossof SERF1 enh.nces RPBF expression resulting in larger grains with.increased starch.content, wh.le SERF1 overexpressionrepresses RPBF resulting in smaller grains. Consistently, during grain filling, starch.biosynth.sis genes such.as GRANULE-BOUND STARCh.SYNTh.SEI （GBSSI）, STARCh.SYNTh.SEI （SSI）, SSIIla, and ADP-GLUCOSE PYROPh.SPh.RYLASELARGE SUBUNIT2 （AGPL2） are up-regulated in SERF1 knockout grains. Moreover, SERF1 is a direct upstream regulator ofGBSSI. In addition, SERF1 negatively regulates germination by controlling RPBF expression, wh.ch.mediates th. gibberel-lic acid （GA）-induced expression of RICE AMYLASEIA （RAmylA）. Loss of SERF1 results in more rapid seedling establish.ment, wh.le SERF1 overexpression h.s th. opposite effect. Our study reveals th.t SERF1 represents a negative regulatorof grain filling and seedling establish.ent by timing th. expression of RPBF.
