2,3-Butanediol from the leachates of pine needles induces the resistance of Panax notoginseng to the leaf pathogen Alternaria panax

Compared with the use of monocultures in the field,cultivation of medicinal herbs in forests is an effective strategy to alleviate disease.Chemical interactions between herbs and trees play an important role in disease suppression in forests.We evaluated the ability of leachates from needles of Pinus armandii to induce resistance in Panax notoginseng leaves,identified the components via gas chromatography-mass spectrometry(GC-MS),and then deciphered the mechanism of 2,3-Butanediol as the main component in the leachates responsible for resistance induction via RNA sequencing(RNA-seq).Prespraying leachates and 2,3-Butanediol onto leaves could induce the resistance of P.notoginseng to Alternaria panax.The RNA-seq results showed that prespraying 2,3-Butanediol onto leaves with or without A.panax infection upregulated the expression of large number of genes,many of which are involved in transcription factor activity and the mitogen-activated protein kinase(MAPK) signaling pathway.Specifically,2,3-Butanediol spraying resulted in jasmonic acid(JA)-mediated induced systemic resistance(ISR) by activating MYC2 and ERF1.Moreover,2,3-Butanediol induced systemic acquired resistance(SAR) by upregulating pattern-triggered immunity(PTI)-and effector-triggered immunity(ETI)-related genes and activated camalexin biosynthesis through activation of WRKY33.Overall,2,3-Butanediol from the leachates of pine needles could activate the resistance of P.notoginseng to leaf disease infection through ISR,SAR and camalexin biosynthesis.Thus,2,3-Butanediol is worth developing as a chemical inducer for agricultural production.

Interfacial water molecules are the most important participants in the hydrogen evolution reaction(HER).Hence,understanding the behavior and role that interfacial water plays will ultimately reveal the HER mechanism.Unfortunately,investigating interfacial water is extremely challenging owing to the interference caused by bulk water molecules and complexity of the interfacial environment.Here,the behaviors of interfacial water in different cationic electrolytes on Pd surfaces were investigated by the electrochemistry,in situ core-shell nanostructure enhanced Raman spectroscopy and theoretical simulation techniques.Direct spectral evidence reveals a red shift in the frequency and a decrease in the intensity of interfacial water as the potential is shifted in the positively direction.When comparing the different cation electrolyte systems at a given potential,the frequency of the interfacial water peak increases in the specified order:li+

Molecular Characterization and Antibacterial Activity of a G-Type Lysozyme in Yellow Drum(Nibea albiflora)

Lysozyme(EC3.***.1.17)plays an important role in the immune response;as a nonspecific immune factor,it can resist causative agents.Lysozyme can be divided into c-type and g-type in fish.In a previous study,through genome-wide association analysis,the g-type lysozyme gene,which is named NaLyg in yellow drum(Nibea albiflora),was found to be a key candidate gene for disease resistance in response to Vibrio harveyi infection.The cDNA of NaLyg was 1025 bp,including four exons and three introns,and its open reading frame(ORF)had a full-length of 582 bp,encoding 193 amino acids.NaLyg was found to be conserved during evolution through bioinformatic analyses.The NaLyg protein possessed a sugar binding domain and three catalytic sites,including Glu71,Asp84 and Asp101.Quantitative qRT-PCR results confirmed that NaLyg gene mRNA was visibly increased after V.harveyi infection.The NaLyg protein purified by prokaryotic expression killed some gram-negative bacterial pathogens by inducing cell wall destruction,including V.harveyi,Aeromonas hydrophila and Edwardsiella tarda.Moreover,the NaLyg protein killed two gram-positive bacteria,Bacillus subtilis and Staphylococcus aureus.Taken together,the experimental results suggested that the NaLyg protein of N.albiflora played an important role in fighting bacterial infections.

SDH-FCOS:An Efficient Neural Network for Defect Detection in Urban Underground Pipelines

Urban underground pipelines are an important infrastructure in cities,and timely investigation of problems in underground pipelines can help ensure the normal operation of cities.Owing to the growing demand for defect detection in urban underground pipelines,this study developed an improved defect detection method for urban underground pipelines based on fully convolutional one-stage object detector(FCOS),called spatial pyramid pooling-fast(SPPF)feature fusion and dual detection heads based on FCOS(SDH-FCOS)model.This study improved the feature fusion component of the model network based on FCOS,introduced an SPPF network structure behind the last output feature layer of the backbone network,fused the local and global features,added a top-down path to accelerate the circulation of shallowinformation,and enriched the semantic information acquired by shallow features.The ability of the model to detect objects with multiple morphologies was strengthened by introducing dual detection heads.The experimental results using an open dataset of underground pipes show that the proposed SDH-FCOS model can recognize underground pipe defects more accurately;the average accuracy was improved by 2.7% compared with the original FCOS model,reducing the leakage rate to a large extent and achieving real-time detection.Also,our model achieved a good trade-off between accuracy and speed compared with other mainstream methods.This proved the effectiveness of the proposed model.

Huangqin Decoction Delays Progress of Colitis-Associated Carcinogenesis by Regulating Nrf2/HO-1 Antioxidant Signal Pathwayin Mice

Objective:To investigate the effect of Huangqin Decoction(HQD)on nuclear factor erythroid 2 related-factor 2(Nrf2)/heme oxygenase(HO-1)signaling pathway by inducing the colitis-associated carcinogenesis(CAC)model mice with azoxymethane(AOM)/dextran sodium sulfate(DSS).Methods:The chemical components of HQD were analyzed by liquid chromatography-quadrupole-time-of-flight mass spectrometry(LC-Q-TOF-MS/MS)to determine the molecular constituents of HQD.Totally 48 C57BL/6J mice were randomly divided into 6 groups by a random number table,including control,model(AOM/DSS),mesalazine(MS),low-,medium-,and high-dose HQD(HQD-L,HQD-M,and HQD-H)groups,8 mice in each group.Except for the control group,the mice in the other groups were intraperitoneally injected with AOM(10 mg/kg)and administrated with 2.5%DSS orally for 1 week every two weeks(totally 3 rounds of DSS)to construct a colitis-associated carcinogenesis mouse model.The mice in the HQD-L,HQD-M and HQD-H groups were given HQD by gavage at doses of 2.925,5.85,and 11.7 g/kg,respectively;the mice in the MS group was given a suspension of MS at a dose of 0.043 g/kg(totally 11 weeks).The serum levels of malondialdehyde(MDA)and superoxide dismutase(SOD)were measured by enzyme-linked immunosorbent assay.The mRNA and protein expression levels of Nrf2,HO-1,and inhibitory KELCH like ECH-related protein 1(Keap1)in colon tissue were detected by quantitative real-time PCR,immunohistochemistry,and Westem blot,respectively.Results:LC-Q-TOF-MS/MS analysis revealed that the chemical constituents of HQD include baicalin,paeoniflorin,and glycyrrhizic acid.Compared to the control group,significantly higher MDA levels and lower SOD levels were observed in the model group(P<0.05),whereas the expressions of Nrf2 and HO-1 were significantly decreased,and the expression of Keap1 increased(P<0.01).Compared with the model group,serum MDA level was decreased and SOD level was increased in the HQD-M,HQD-H and MS groups(P<0.05).Higher expressions of Nrf2 and HO-1 were observed in the HQD groups.Co

A novel topology with controllable wideband baseband impedance for power amplifiers

This paper presents a novel topology to control the baseband impedance of a power amplifier(PA)to avoid performance deterioration in concurrent dual-band mode.This topology can avoid pure resonance of capacitors and inductors LC,which leads to a high impedance at some frequency points.Consequently,it can be applied to transmitters that are excited by broadband signals.In particular,by adjusting the circuit parameters and increasing stages,the impedance of the key frequency bands can be flexibly controlled.A PA is designed to support this design idea.Its saturated output power is around 46.7 dBm,and the drain efficiency is>68.2%(1.8-2.3 GHz).Under concurrent two-tone excitation,the drain efficiency reaches around 40%even under 5.5 dB back-off power with the tone spacing from 10 MHz to 500 MHz.These results demonstrate that the proposed topology is capable of controlling wideband baseband impedance.

Co-pyrolysis of Sewage Sludge with Paint Sludge: Kinetics and Thermodynamic Analysis via Iso-conversional Methods

This study explored the synergistic interaction of sewage sludge(SS)and automotive paint sludge(PS)during co-pyrolysis for the optimized treatment of sewage sludge in cement kiln systems,utilizing thermogravimetric analysis(TGA)and thermogravimetric-mass spectrometry(TGA-MS).The result reveals the coexisting synergistic and antagonistic effects in the co-pyrolysis of SS/PS.The synergistic effect arises from hydrogen free radicals in SS and catalytic components in PS,while the main source of the antagonistic effect is that,during the mechanical mixing process,the SS/PS is converted from the particulate form into a dough-like rubbery which contributes to the film-forming effect,hindering the volatilization of volatile components.SS/PS co-pyrolysis reduces the yielding of tar production while increasing coke and gas.This study will provide some in-depth insights into the co-pyrolysis of SS/PS,and offer theoretical support for the subsequent research on the collaborative disposal processes in cement kilns.

Rhododendron is famous for its high ornamental value.However,the genus is taxonomically difficult and the relationships within Rhododendron remain unresolved.In addition,the origin of key morphological characters with high horticulture value need to be explored.Both problems largely hinder utilization of germplasm resources.Most studies attempted to disentangle the phylogeny of Rhododendron,but only used a few genomic markers and lacked large-scale sampling,resulting in low clade support and contradictory phylogenetic signals.Here,we used restriction-site associated DNA sequencing(RAD-seq)data and morphological traits for 144 species of Rhododendron,representing all subgenera and most sections and subsections of this species-rich genus,to decipher its intricate evolutionary history and reconstruct ancestral state.Our results revealed high resolutions at subgenera and section levels of Rhododendron based on RAD-seq data.Both optimal phylogenetic tree and split tree recovered five lineages among Rhododendron.Subg.Therorhodion(cladeⅠ)formed the basal lineage.Subg.Tsutsusi and Azaleastrum formed cladeⅡand had sister relationships.CladeⅢincluded all scaly rhododendron species.Subg.Pentanthera(cladeⅣ)formed a sister group to Subg.Hymenanthes(cladeⅤ).The results of ancestral state reconstruction showed that Rhododendron ancestor was a deciduous woody plant with terminal inflorescence,ten stamens,leaf blade without scales and broadly funnelform corolla with pink or purple color.This study shows significant distinguishability to resolve the evolutionary history of Rhododendron based on high clade support of phylogenetic tree constructed by RAD-seq data.It also provides an example to resolve discordant signals in phylogenetic trees and demonstrates the application feasibility of RAD-seq with large amounts of missing data in deciphering intricate evolutionary relationships.Additionally,the reconstructed ancestral state of six important characters provides insights into the innovation of key characters in Rhododendron.

Celastrol regulates the oligomeric state and chaperone activity of 𝛼B-crystallin linked with protein homeostasis in the lens

Protein misfolding and aggregation are crucial pathogenic factors for cataracts,which are the leading cause of visual impairment worldwide.α-crystallin,as a small molecular chaperone,is involved in preventing protein misfolding and maintaining lens transparency.The chaperone activity of α-crystallin depends on its oligomeric state.Our previous work identified a natural compound,celastrol,which could regulate the oligomeric state of αB-crystallin.In this work,based on the UNcle and SEC analysis,we found that celastrol induced𝛼αB-crystallin to form large oligomers.Large oligomer formation enhanced the chaperone activity of αB-crystallin and prevented aggregation of the cataract-causing mutant αA3-G91del.The interactions between𝛼αB-crystallin and celastrol were detected by the FRET(Fluorescence Resonance Energy Transfer)technique,and verified by molecular docking.At least 9 binding patterns were recognized,and some binding sites covered the groove structure of αB-crystallin.Interestingly,αB-R120G,a cataract-causing mutation located at the groove structure,and celastrol can decrease the aggregates of αB-R120G.Overall,our results suggested celastrol not only promoted the formation of large αB-crystallin oligomers,which enhanced its chaperone activity,but also bound to the groove structure of its α-crystallin domain to maintain its structural stability.Celastrol might serve as a chemical and pharmacological chaperone for cataract treatment.

Low-permittivity liLn(PO3)4(Ln=La,Sm,Eu)dielectric ceramics for microwave/millimeter-wave communication

The development of dielectric materials with low permittivity and low loss is a great challenge in wireless communication.In this study,liLn(PO_(3))_(4)(Ln=La,Sm,Eu)ceramic systems were successfully prepared using the traditional solid-state method.X-ray diffraction analysis indicated that the liLn(PO_(3))_(4)ceramics crystallized in a monoclinic structure when sintered at 850–940℃.The characteristic peak shifted to higher angles with variations in the Ln element,which was ascribed to a reduction in the cell volume.Further analysis by structure refinement revealed that the reduction in the cell volume resulted from the decrease in chemical bond lengths and the compression of[liO4]and[PO4]tetrahedra.Remarkably,the liLn(PO_(3))_(4)ceramic system displayed exceptional performance at low sintering temperatures(910–925℃),including a high quality factor(Q·f)of 41,607–75,968 GHz,low temperature coefficient of resonant frequency(τf)ranging from−19.64 to−47.49 ppm/℃,low permittivity(εr)between 5.04 and 5.26,and low density(3.04–3.26 g/cm^(3)).The application of Phillips–van Vechten–Levine(P–V–L)theory revealed that the increased Q·f value of the liLn(PO_(3))_(4)systems can be attributed to the enhanced packing fraction,bond covalency,and lattice energy,and the stability ofτf was associated with the increase in the bond energy.Furthermore,a prototype microstrip patch antenna using liEu(PO3)4 ceramics was fabricated.The measurement results demonstrated excellent antenna performance with a bandwidth of 360 MHz and a peak gain of 5.11 dB at a central frequency of 5.08 GHz.Therefore,low-εr liLn(PO_(3))_(4)ceramic systems are promising candidates for microwave/millimeter-wave communication.
