Rapid Detection of Haemophilus influenzae and Haemophilus parainfluenzae in Nasopharyngeal Swabs by Multiplex PCR

Objective To establish multiplex PCR-based assays for detecting *** and ***. And the PCR-based assays were applied to detect the carriage rates of *** and *** in nasopharyngeal swab specimens which were collected from healthy children. Methods Multiplex primers for species-specific PCR were designed by using DNAstar soft based on the sequences of 165 rRNA genes from genus Haemophilus to detect *** and ***. Results The sensitivity of the 165 rRNA PCR assay for detecting *** and *** was 97.53% and 100% respectively, and the specificity was 95.89% and 96.63% respectively. Youden＇s Index on the ability to detect *** and *** was 0.9342 and 0.9663 respectively. 666 nasopharyngeal swab specimens were collected from healthy children. The detection rates of *** and *** were 14.11% and 16.07% respectively by using isolation and culture methods. The detection rates of *** and *** were 43.54% and 57.96% respectively by 165 rRNA PCR assays. The carriage rates of serotypes a, b, c, d, e, f and non-typeable isolates were 0% （0/666）, 0.15% （1/666）, 1.20% （8/666）, 0.15% （1/666）, 1.20% （8/666）, 1.80% （12/666）, 95.50% （636/666） respectively. Conclusion The multiplex PCR assays were very rapid, reliable and feasible methods for detection of *** and *** in pharyngeal swab specimens which were compared to conventional isolation and culture methods. 95.5% of *** strains in healthy children were nontypeable. The encapsulated or typable strains were mainly three serotypes which was c, e, and f serotype.

A large-area bionic skin for high-temperature energy harvesting applications

For the large amount of waste heat wasted in daily life and industrial production,we propose a new type of flexible thermoelectric generators(F-TEGs)which can be used as a large area bionic skin to achieve energy harvesting of thermal *** reference to biological structures such as pinecone,succulent,and feathers,we have designed and fabricated a biomimetic flexible TEG that can be applied in a wide temperature range which has the highest temperature energy harvesting capability *** laminated free structure of the bionic F-TEG dramatically increases the efficiency and density of energy *** F-TEGs(single TEG only 101.2 mg in weight),without an additional heat sink,demonstrates the highest output voltage density of 286.1 mV/cm^(2)and the maximum power density is 66.5 mW/m^(2) at a temperature difference of nearly 1000℃.The flexible characteristics of F-TEGs make it possible to collect the diffused thermal energy by flexible attachment to the outer walls of high-temperature pipes and vessels of different diameters and *** work shows a new design and application concept for flexible thermal energy collectors,which fills the gap of flexible energy harvesting in high-temperature environment.

Fracture mode identification of low alloy steels and cast irons by electron back-scattered diffraction misorientation analysis

The fracture modes of low alloy steels and cast irons under tensile and fatigue conditions were identified by electron back-scattered diffraction（EBSD） misorientation analysis in this research. The curves of grain reference orientation deviation（GROD） distribution perpendicular to the fracture surface were obtained by EBSD observation, and the characteristics of each fracture mode were identified. The GROD value of the specimen fractured in tension decreases to a constant related to the elongation of corresponding specimen in the far field（farther than 5 mm away from the fracture surface）. The peak exhibits in GROD curves of two smooth specimens and a notched specimen near the fracture surface（within 5 mm away from the fracture surface）, and the formation mechanisms were discussed in detail based on the influences of specimen geometries（smooth or notched） and material toughness. The GROD value of fatigue fractured specimen is close to that at undeformed condition in the whole field, except the small area near the crack path. The loading conditions（constant stress amplitude loading or constant stress intensity factor range K loading） and the EBSD striation formation during fatigue crack propagation were also studied by EBSD observation parallel to the crack path.

Enhance liquid nitrogen fracturing performance on hot dry rock by cyclic injection

Producing complex fracture networks in a safe way plays a critical role in the hot dry rock (HDR) geothermal energy exploitation. However, conventional hydraulic fracturing (HF) generally produces high breakdown pressure and results only in single main fracture morphology. Furthermore, HF has also other problems such as the increased risk of seismic events and consuption of large amount of water. In this work, a new stimulation method based on cyclic soft stimulation (CSS) and liquid nitrogen (LN2) fracturing, known as cyclic LN2 fracturing is explored, which we believe has the potential to solve the above issues related to HF. The fracturing performances including breakdown pressure and fracture morphology on granites under true-triaxial stresses are investigated and compared with cyclic water fracturing. Cryo-scanning electron microscopy (Cryo-SEM) tests and X-ray computed tomography (CT) scanning tests were used for quantitative characterization of fracture parameters and to evaluate the cyclic LN2 fracturing performances. The results demonstrate that the cyclic LN2 fracturing results in reduced breakdown pressure, with between 21% and 67% lower pressure compared with using cyclic water fracturing. Cyclic LN2 fracturing tends to produce more complex and branched fractures, whereas cyclic water fracturing usually produces a single main fracture under a low number of cycles and pressure levels. Thermally-induced fractures mostly occur around the interfaces of different particles. This study shows the potential benefits of cyclic LN2 fracturing on HDR. It is expected to provide theoretical guidance for the cyclic LN2 fracturing application in HDR reservoirs.

Aerobic Cr(VI) Reduction by an Indigenous Soil Isolate Bacillus thuringiensis BRC-ZYR2

Chromium(Cr) may cause losses in the yield of field plant, which is one of the favorite habitats of Bacillus thuringiensis(Bt). The purposes of our study were to assess the Cr(VI)-resistance and Cr(VI)-reducing abilities of an indigenous soil isolate of Bt and to determine the factors governing Cr(VI) reduction. Towards this end a novel dichromate-reducing Bt BRC-ZYR2, characterized with insecticidal crystal proteins(ICPs), was isolated from a uranium deposit. Minimum inhibitory concentrations(MICs) of Cr(VI) were determined by broth dilution method and the concentrations of Cr(VI) and total Cr in the supernatant were quantified colorimetrically using 1,5-diphenylcarbazide(DPC) reagent and a mixture of sulfuric-nitric acids, respectively. The isolate contained five ICP genes(cry1Ba, cry1 Bb, cry1Be/cry1 Bf, cry9 Ca and cry9Da) and exhibited a high level of Cr(VI) resistance with MICs of 150 mg L-1at pH 7.0 and 30?C, and 500 mg L-1under optimal conditions(pH 9.0 and 40?C). The total Cr concentration was similar to initial concentration of Cr(VI) under the optimal condition, suggesting that the essential removal of the Cr(VI) was dependent on Bt reduction. Under optimal conditions, the initial Cr(VI) concentrations from 25 to 75 mg L-1significantly decreased in 24 h after incubation. Addition of Mn2+, Co2+, Mo2+and Cu2+activated Bt-mediated Cr(VI) reduction, while Zn2+, Ni2+and glucose were found to inhibit the reduction. Our results indicated that this isolate could be a promising biopesticide with the potential for both insect biocontrol and Cr bioremediation in the field.

To meet the expectation set by Moore’s law on transistors,the search for thickness-scalable high dielectric constant(k)gate layers has become an emergent research *** investigations have failed to solve the“polarizability–scalability–insulation robustness”*** this work,we show that this trilemma can be solved by using a gate layer of a high k ferroelectric oxide in its superparaelectric(SPE)*** the SPE,its polar order becomes local and is dispersed in an amorphous matrix with a crystalline size down to a few nanometers,leading to an excellent dimensional scalability and a good field-stability of the k *** an example,a stable high k value(37±3)is shown in ultrathin SPE films of(Ba_(0.95),Sr_(0.05))(Zr_(0.2),Ti_(0.8))O_(3)deposited on LaNiO_(3)-buffered Pt/Ti/SiO_(2)/(100)Si down to a 4 nm thickness,leading to a small equivalent oxide thickness of~0.46 *** aforementioned characteristic microstructure endows the SPE film a high breakdown strength(~10.5 MV·cm^(−1)for the 4 nm film),and hence ensures a low leakage current for the operation of the complementary metal oxide semiconductor(CMOS)***,a high electrical fatigue resistance is displayed by the SPE *** results reveal a great potential of superparaelectric materials as gate dielectrics in the next-generation microelectronics.

Design of depth-control planting unit with single-side gauge wheel for no-till maize precision planter

No-till planters are very popular for maize seeding in fields covered with residue in annual wheat-maize double cropping system in North China ***,there is no suitable depth control mechanism for existing no-till maize planters,and as a result,it is hard to obtain consistent planting depth,uniform emergence,and good passing ability at the same *** the above reasons,a proper planting unit with a new type of depth-control mechanism was developed in this *** mechanism consists of a single-side gauge wheel,a parallel four-bar linkage,a pair of double-disc opener,a V-shape press wheel and a depth regulator,which can adjust the planting depth from 30 mm to 90 mm to satisfy the agronomic requirement under different field *** on analysis and calculation,the width of gauge wheel was set to 50 mm while the length of parallel four-bar linkage was set to 350 *** experiment was conducted and the results indicated that the newly designed planting unit with single-side gauge wheel performed well with regard to planting depth uniformity and anti-blocking *** planting depth uniformity and seed spacing quality were 95.45%and 91.90%,respectively,when the average height of stubble was 22.5 cm and residue amount was 0.64 kg/m^(2)in the *** can satisfy the requirement of no-till maize planting on the cropland with residue and stubble in North China Plain.

Clinical features of coronary artery ectasia in the elderly

Objective To investigate the incidence, imaging and clinical characteristics in elderly patients with coronary artery ectasia （CAE）. Methods A retrospective analysis was conducted on patients with CAE who underwent coronary angiography between January 2006 and December 2012. According to age, the enrolled patients were divided into two groups （elderly group, age≥ 65 years; non-elderly group, age 〈 65 years）. The clinical feature, imaging characteristics and the 5-year survival rate of the two groups were *** The preva-lence of CAE in elderly patients was 0.33%. Patients in elderly group were found to have significantly higher proportion of female （30.1%vs. 10.1%,P〈 0.001）, three-vessel disease （60.5%vs. 45.2%,P = 0.003） and localized ectasia （55.0%vs. 40.2%,P = 0.003）. In addition, body mass index （20.90 ± 2.71 kg/m2vs. 22.31 ± 2.98 kg/m2,P 〈 0.001） and percentage of current smokers （45.0%vs. 64.6%,P 〈 0.001） were significantly lower in elderly group. Cumulative survival curves demonstrated reduced 5-year cumulative survival at the follow-up in the elderly group compared with the non-elderly group （88.0%vs. 96.0%,P = 0.002）. But the 5-year event free survival rate failed to show a significant difference between the two groups （31.0%vs. 35.0%,P= 0.311）.ConclusionThe prevalence of CAE in elderly patients was 0.33%, which was about 1/3 of the entire numbers of CAE patients. There were significant differences between the elderly and the non-elderly patients with CAE in terms of coronary artery disease risk factors and coronary artery ectatic characteristics. CAE might be asso-ciated with increased mortality risk in the elderly.
