海洋生态系统的动力问题,如可观测的化学—生物量的时空变化是由于生物和物理过程决定的。预报海洋系统的未来发展需要有一个理论框架,如模型,而这个模型是根据对相关的各个不同过程的研究和了解而发展的。从理论上描述海洋系统的自然方法似乎是把化学生物模型与环境模型结合在一起。但是,环境模型相对比较先进,而化学—生物过程的定量理论描述比较落后。本文讨论了一些关于发展相容理论的方法和问题,指出了海洋生物学模型和海洋学模型耦合的潜在益处。

Aims: The value of ECG in the perioperative risk stratification under modern treatment options is uncertain. The main objective of the present analysis was to determine the usefulness of a pre-operative ECG derivation for evaluating the risk of perioperative morbidity and mortality. Methods: We performed a secondary analysis of the prospective, international, multicenter, observational “No-Risk” Study (N-terminal B-type natriuretic peptide [NT-proBNP] for the assessment of the perioperative cardiac risk after major noncardiac surgery) to determine the prognostic value of 12-lead ECG. Inclusion criteria were age >55 years and at least one of the following cardiovascular risk factors: arterial hypertension, diabetes mellitus, dyslipidemia, active smoking, and family history positive for coronary heart disease. The combined primary endpoint included total mortality, acute myocardial infarction (NSTEMI and STEMI), cardiopulmonary resuscitation, heart failure, and asystole or ventricular fibrillation during hospitalization. ECGs from 616 patients enrolled from 2006 to 2009 prior to noncardiac surgery in the No-Risk Study were analyzed. Results: The mean age was 67.6 (±8.1) years;300 (48.7%) patients were male. Fourteen (2.3%) patients suffered from the combined primary endpoint while in the hospital. In Kaplan-Meier analyses, a pathologic Q wave and QTc > 500 ms were significantly related to the incidence of the primary endpoint (p p = 0.042, respectively), whereas other ECG parameters such as LBBB, RBBB, PQ interval, QRS interval, and others were not related to worse in-hospital outcome. Conclusion: The 12-lead ECG is still an important diagnostic tool for perioperative risk assessment of cardiovascular events in noncardiac surgery in patients at risk.

The two LIM domain-containing proteins from plants （LIMs） typically exhibit a dual cytoplasmic-nuclear dis-tribution, suggesting that, in addition to their previously described roles in actin cytoskeleton organization, they partici-pate in nuclear processes. Using a south-western blot-based screen aimed at identifying factors that bind to plant histone gene promoters, we isolated a positive clone containing the tobacco LIM protein WLIM2 （NtWLIM2） cDNA. Using both green fluorescent protein （GFP） fusion-and immunology-based strategies, we provide clear evidence that NtWLIM2 local-izes to the actin cytoskeleton, the nucleus, and the nucleolus. Interestingly, the disruption of the actin cytoskeleton by latrunculin B significantly increases NtWLIM2 nuclear fraction, pinpointing a possible novel cytoskeletal-nuclear crosstalk. Biochemical and electron microscopy experiments reveal the ability of NtWLIM2 to directly bind to actin filaments and to crosslink the latter into thick actin bundles. Electrophoretic mobility shift assays show that NtWLIM2 specifically binds to the conserved octameric cis-elements （Oct） of the Arabidopsis histone H4A748 gene promoter and that this binding largely relies on both LIM domains. Importantly, reporter-based experiments conducted in Arabidopsis and tobacco proto-plasts confirm the ability of NtWLIM2 to bind to and activate the H4A748 gene promoter in live cells. Expression studies indicate the constitutive presence of NtWLIM2 mRNA and NtWLIM2 protein during tobacco BY-2 cell proliferation and cell cycle progression, suggesting a role of NtWLIM2 in the activation of basal histone gene expression. Interestingly, both live cell and in vitro data support NtWLIM2 di/oligomerization. We propose that NtWLIM2 functions as an actin-stabilizing protein, which, upon cytoskeleton remodeling, shuttles to the nucleus in order to modify gene expression.

Using Host-Specificity of Cryptosporidium to Understand Contaminant Sources, Seasonality, and Human Health Risk in Three Watersheds of Differing Land-Use

Three tributaries of the Grand River watershed (Ontario, Canada), each representing different watershed types (urban, agricultural/rural, and mixed land-use) were examined to understand the spatial, temporal, and host-source distribution of the waterborne pathogen, Cryptosporidium. Cryptosporidium was frequently found throughout the study (73%, 65/89) with occurrence and concentrations observed to be similar among the varying watershed types. However, applying advanced genotyping techniques, marked differences in dominant host sources could be observed in each watershed. The agricultural/rural and mixed land-use watersheds were dominated by genotypes typically associated with cattle (i.e., C. andersoni), while the urban watershed had the highest diversity of Cryptosporidium genotypes with a variety of wildlife as the common source of contamination (e.g., muskrat and cervine genotypes). A similar seasonal trend observed in the urban, agricultural, and mixed land-use watershed suggests that factors beyond specific land use activities (e.g. autumn manure spreading) may influence the timing and concentration of Cryptosporidium in these streams. Corresponding genotyping results provided additional insight into source inputs during these seasonal peaks, indicating that wildlife may be important seasonal contributors to Cryptosporidium contamination in these streams. Despite the abundance of Cryptosporidium in these watersheds, most of the genotypes observed were of limited human health importance. This study provides evidence regarding the significance of including genotyping results into studies examining waterborne Cryptosporidium. Using this technique can provide a greater understanding of the risk to the population using water sources, as well as provide insight into the probable sources and timing of contamination. This ancillary information can contribute to implementation of targeted management strategies to further protect sources of drinking water and recreation

Smoking behavior of HIV-infected patients

Recent reports describe an increased rate of cardiovascular events in smoking HIV-infected subjects. However, a lot is still unknown about smoking in this patient population. The purpose of the study was to analyze smoking behavior in HIV-infected subjects as a risk factor of coro-nary atherosclerosis and determine its effect on the probability of coronary events. We analyzed the cardiovascular risk factors of 294 HIV-infected adults (age: 42.1 ? 10.1 years;77% males). An elevated tobacco abuse was observed in 63.6% of the HIV-infected patients. Tobacco use was much more common in HIV-infected males than in females (67.8% vs. 49.2%;p 0.2). Analysing the way of infection and the status of smoking, patients with HIV-infection acquired by heterosexual contact exhibited sig-nificantly lower rates of smoking compared with patients with HIV-infection acquired by MSM (man having sex with man) or by intravenous drug abuse (52.7% vs. 67.4%/82.1%, p < 0.01). The effect of smoking on the 10yrs. probability of coronary events determined by Framingham- equation was superior compared with all other classic cardiovascular risk factors. HIV-infected patients exhibited an increased tobacco use. Knowledge about smoking behavior in this pa-tient population is essential to evaluate the risk of cardiovascular events and to implicate prevention strategies for HIV-infected subjects.
