This work evaluated the effect of dissolved oxygen and the initial inoculum concentration on xylose reductase (XR) production by Candida guilliermondii from sugarcane bagasse hemicellulosic hydrolysate. Both the parameters were studied under an experimental design 22 with triplicate at central point. The statistical analysis of the results indicated a significant negative effect on XR production from the variable inoculum. The variable dissolved oxygen also showed a negative effect on XR production. We found the maximum enzyme activity (2.5 U?mg?1) when both the factors were applied at their lowest levels. The yeast showed to be potentially capable for xylose reductase production when sugarcane bagasse hemicellulosic hydrolysate was used as carbon source. Also, the results presented important information for further optimization of xylose reductase attainment.

Objective To evaluate the chemical composition and effects of Artemisia vulgaris(AV)hydroalcoholic extract(HEAV)on breast cancer cells(MCF-7 and SKBR-3),chronic myeloid leukemia(K562)and NIH/3T3 *** Phytochemical analysis of HEAV was done by high-performance liquid chromatography-mass(HPLC)*** and cell death studies were performed using trypan blue and Annexin/FITC-7AAD,***-1(Fer-1)and necrostatin-1(Nec-1)were used to assess the mode of HEAV-induced cell death and acetoxymethylester(BAPTA-AM)was used to verify the involvement of cytosolic calcium in this *** calcium measurements were made using *** HEAV decreased the viability of MCF-7,SKBR-3 and K562 cells(P<0.05).The viability of HEAV-treated K562 cells was reduced compared to HEAV-exposed fibroblasts(P<0.05).Treatment of K562 cells with HEAV induced cell death primarily by late apoptosis and necrosis in assays using annexin V-FITC/7-AAD(P<0.05).The use of Nec-1 and Fer-1 increased the viability of K562 cells treated with HEAV relative to cells exposed to HEAV alone(P<0.01).HEAV-induced Ca^(2+)release mainly from lysosomes in K562 cells(P<0.01).Furthermore,BAPTA-AM,an intracellular Ca^(2+)chelator,decreased the number of non-viable cells treated with HEAV(P<0.05).Conclusions HEAV is cytotoxic and activates several modalities of cell death,which are partially dependent on lysosomal release of Ca^(2+).These effects may be related to artemisinin and caffeoylquinic acids,the main compounds identified in HEAV.

Background:Hemodialysis(HD) per se is a risk factor for *** the growing body of evidence on blood-flow restriction(BFR) exercise in HD patients,identification of possible risk factors related to the prothrombotic agent D-dimer is required for the safety and feasibility of this training *** aim of the present study was to identify risk factors associated with higher D-dimer levels and to determine the acute effect of resistance exercise(RE) with BFR on this ***:Two hundred and six HD patients volunteered for this study(all with a glomerular filtration rate of 70 years old),and HD ***,RE+BFR significantly increases D-dimer after 4 *** with borderline baseline D-dimer levels(400-490 ng/mL) displayed increased risk of elevating D-dimer over the normal range(≥500 ng/mL).Conclusion:These results identified factors associated with a heightened prothrombotic state and may assist in the screening process for HD patients who wish to undergo RE+BFR.D-dimer and/or other fibrinolysis factors should be assessed at baseline and throughout the protocol as a precautionary measure to maximize safety during RE+BFR.

With the increase in antimicrobial resistance,it has become necessary to explore alternative approaches for combating and preventing ***-cinnamaldehyde(CNM)and Benzyl4-amino(B4AM)are bioactive compounds derived from chalcones but with restricted solubility in aqueous *** can enhance the solubility of compounds and can be a promising alternative in the development of novel antimicrobials,with reduced side effects and prolonged *** objective of this study was to evaluate the stability of oil-in-water nanoemulsions loaded with two distinct types of chalcones at two different dosages,to propose a stable formulation with antimicrobial *** showed that nanoemulsions presented high encapsulation efficiency,low polydispersity index(PDI)and particle size below 200 nm,indicating that emulsification was a suitable method for nanoemulsion *** with higher dosages exhibited significant antimicrobial effects when compared to free chalcones and positive ***,B4AM nanoemulsions at higher dosages showed expressive activity against Salmonella minnesota,with a 420%greater inhibitory response compared to the free form and showing equivalence to the positive *** nanoemulsions showed excellent inhibitory activity at the highest dosage,equivalent to the positive control against *** and Staphylococcus *** greater number of conjugated bonds in CNM increased the antimicrobial activity in comparison with B4AM,and the formation of nanometric domains enhanced the bioavailability,being a promising alternative for antimicrobial applications.

Fluctuation of Mite Fauna Associated to Rice Culture （Oryza sativa L.： Poales, Poaceae） in Two Regions in the State of Rio Grande do Sul, Brazil

This survey had the aim to identify the mite fauna and mite ecologic interaction in rice culture, harvest 2010-2011, in Taquari and Cachoeirinha Counties, state of Rio Grande do Sul, Brazil. The mite population was biweekly evaluated on IRGA 424 and INTA PUITA CL cultivars in four areas where randomly sampled 20 plants/area. Fluctuation, ecologic indices and correlation between species and environmental factors were calculated. A total of 1,626 mites belonging to 14 species from 12 families were collected. Of the total specimens collected, 34.56% was on IRGA 424, in Taquari, 32.47% on IRGA 424-120 and 28.35% on IRGA 424-60, in Cachoeirinha and 4.61% on 1NTA PUIT,g, CL, in Taquari. Family Ascidae showed great richness, with four species, Lasioseius sp., Lasioseius sp., Proctolaelaps sp. and Cheiroseius sp.. Schizotetranychus oryzae Rossi de Simons （86.65%） was the phytophagous mite more abundant, while among the predators Neoseiulus paraibensis （Moraes and McMurtry）（6.88%） stood out. The population peaks of S. oryzae and N. paraibensis happened on March 2011. In all evaluated areas, the correlation between S. oryzae and N. paraibensis was positive, specially on IRGA 424-120 （r = 0.93, P = 0.006）.

Culex mosquitoes are primarily found in temperate and tropical regions worldwide where they play a crucial role as main vectors of filarial worms and arboviruses. In Recife, a northeast city in Brazil, high densities of Culex quinquefasciatus are often found in association with human populated areas. In marked contrast to another part of the city, field tests conducted in the neighborhood of Sitio dos Pintos showed that trapping of mosquitoes in skatole-baited ovitraps did not differ significantly from captures in control (water) traps. Thus, classical and molecular taxonomic approaches were used to analyze the Culex species circulating in Sitio dos Pintos. Results obtained from both approaches agreed on the cocirculation of Culex quinquefasciatus and Culex nigripalpus in three different areas of this neighborhood. What was initially considered as an unexpected failure of this lure turned out to be a more unsettling problem, that is, the first report in Recife of Culex nigripalpus, a vector of Venezuelan equine encephalitis virus and West Nile virus. Unplanned urbanization processes close to remnants of the Atlantic forest, such as observed in Sitio dos Pintos, may have contributed to the introduction of Cx. nigripalpus in urban areas.

Allelopathic Potential of Wheat on Sourgrass Resistant to Glyphosate

We aimed with this study to assay the allelopathic potential of wheat genotypes in inhibiting germination and initial growth of sourgrass (Digitaria insularis) resistant to the herbicide glyphosate. Two experiments were installed under greenhouse conditions: the first one aimed to assay the potential of wheat genotypes in inhibiting emergence and initial growth of two test plants, and the second aimed to evaluate the potential of those wheat genotypes which performed better at the first experiment, inhibiting the emergence and initial growth of sourgrass biotypes resistant to the herbicide glyphosate. Five doses of aqueous cool-extracted extract from 32 wheat genotypes were applied to tomato and cucumber (Experiment 1) aiming to identify the ones with higher inhibitory effect. The five most inhibitory wheat genotypes were selected for Experiment 2, where the same doses of extract were applied on sourgrass. For both experiments, shoot fresh and dry mass, as well as water content, were assessed. Sourgrass germination is affected by wheat extracts, but initial seedling growth seems not to be affected;wheat genotypes differ in terms of their allelopathic potential in inhibiting sourgrass germination;wheat presents little inhibitory effect on sourgrass.

From the estimation of the mass balance model, which can also be classified as “black box” model, it is possible to infer the impact of management on the system considered. This study aimed to evaluate water pollution generated by wastewater from a polyculture system of tilapia and shrimp and discuss the management employee and their relation to the quality of the effluent released. It used a pond measuring 1500 m2, average depth 1.6 meters, where 12 cages of 1 m3 populated with tilapia juveniles were installed 33 days after the shrimps’ population. The tilapia juveniles were distributed in densities from 200 to 400 fish per cubic meter, reaching the density of 2.4 fish per square meter within the total pond area. Shrimp post-larvae were released outside the cages within the pond area in a density of 3.3 organisms per square meter. Total density considering fish and shrimps was of 5.7 organisms per square meter in the pond area. Water samples were taken weekly in affluent and effluent of the pond (January-August/2009). The mass balance model was calculated from the difference between the estimated load for the output and input of the pond. The average flow rate was 4.46 L/s. The average loads nitrogen was 0.072 Kg/day (affluent) and 0.179 Kg/day (effluent) and phosphorus 0.0136 Kg/day (affluent) and 0.031 Kg/day (effluent). The mass balance resulted in mean values of 0.11 ± 0.06 Kg/day for total nitrogen and 0.017 ± 0.010 Kg/day for total phosphorus indicating that the system exported nutrients. The use of Best Management Practices (BMP) likes better feed and water management as a way to minimize nutrient export.

Technical Evaluation of Eye Drops Instillation： A Systematic Review

Objective： Assess the profile of eye drops in patients with eye diseases. Methods： Studies available in the scientific literature were identified without any time limits using the databases Embase-Medline, Scielo, Scopus and Web of Knowledge. The selected studies were compared with the following inclusion criteria：（i） if the study evaluated the eye drop instillation, （ii） if the study involved participation of patients with eye diseases. Results： Twelve studies met the inclusion criteria. Of them, 83.33% evaluated the technique of eye drop instillation in patients with glaucoma, 8.33% in patients with eye diseases and 8.33% in patients with cataract after undergoing surgery to correct. 41.66% of studies have chosen to record a video of patients to analyze the technique of instilling eye drops and 41.66% did not describe the/esearch location. Regarding the type of study, 75% had the design as prospective cross-sectional, 8.33% prospective open label study, 8.33% intervention study and 8.33% study called masked trial. Although studies evaluate the technique of eye drop instillation, only 8.33% describe in the article the reference in the literature used to evaluate patients. 50% of articles acknowledged some sort of bias or limitation. Conclusion： The limitations inherent in these types of studies should guide future research.
