MicroRNAs（miRNAs） are a class of highly conserved small non-coding RNA molecules that play a pivotal role in several cellular *** this study,miRNA and messenger RNA（mRNA） profiles were examined by Illumina microarray in mouse embryonic stem cells（ESCs） derived from parthenogenetic,androgenetic,and fertilized *** global analysis of miRNA-mRNA target pairs provided insight into the role of miRNAs in gene *** showed that a total of 125 miRNAs and 2394 mRNAs were differentially expressed between androgenetic ESCs（aESCs） and fertilized ESCs（fESCs）,a total of 42 miRNAs and 87 mRNAs were differentially expressed between parthenogenetic ESCs（pESCs） and fESCs,and a total of 99 miRNAs and 1788 mRNAs were differentially expressed between aESCs and *** addition,a total of 575,5 and 376 miRNA-mRNA target pairs were observed in aESCs ***,pESCs ***,and aESCs ***,***,15 known imprinted genes and 16 putative uniparentally expressed miRNAs with high expression levels were confirmed by both microarray and real-time ***,transfection of miRNA inhibitors was performed to validate the regulatory relationship between putative maternally expressed miRNAs and target mRNAs. Inhibition of miR-880 increased the expression of Peg3,Dyrklb,and Prrg2 mRNA,inhibition of miR-363 increased the expression of Nfat5 and Soatl mRNA,and inhibition of miR-883b-5p increased Nfat5,Tacstd2,and Ppapdc1 *** results warrant a functional study to fully understand the underlying regulation of genomic imprinting in early embryo development.

The journey to implement cancer genomic medicine(CGM)in oncology practice began in the 1980s,which is considered the dawn of genetic and genomic cancer *** the time,a variety of activating oncogenic alterations and their functional significance were unveiled in cancer cells,which led to the development of molecular targeted therapies in the 2000s and *** CGM is still a relatively new discipline and it is difficult to predict to what extent CGM will benefit the diverse pool of cancer patients,the National Cancer Center(NCC)of Japan has already contributed considerably to CGM advancement for the conquest of *** back at these past achievements of the NCC,we predict that the future of CGM will involve the following:1)A biobank of paired cancerous and non-cancerous tissues and cells from various cancer types and stages will be *** quantity and quality of these samples will be compatible with omics *** biobank samples will be linked to longitudinal clinical information.2)New technologies,such as whole-genome sequencing and artificial intelligence,will be introduced and new bioresources for functional and pharmacologic analyses(e.g.,a patient-derived xenograft library)will be systematically deployed.3)Fast and bidirectional translational research(bench-to-bedside and bedside-to-bench)performed by basic researchers and clinical investigators,preferably working alongside each other at the same institution,will be implemented;4)Close collaborations between academia,industry,regulatory bodies,and funding agencies will be established.5)There will be an investment in the other branch of CGM,personalized preventive medicine,based on the individual's genetic predisposition to cancer.

Analysis of proteomic profiling of mouse embryonic stem cells derived from fertilized, parthenogenetic and androgenetic blastocysts

Embryonic stem cells (ESCs) are derived from the inner cell mass (ICM) of preimplantation embryos. ESCs exhibit true pluripotency, i.e., the ability to differentiate into cells of all three germ layers in the developing embryo. We used 2-DE MALDI-TOF/TOF to identify differentially expressed proteins among three types of mouse embryonic stem cells (ESCs) derived from ferti-lized, parthenogenetic, and androgenetic (fESC, pESC and aESC, respectively) blastocysts. We detected more than 800 proteins on silver- stained gels of whole protein extracts from each type of ESC. Of these, 52 differentially expressed proteins were identified by the MALDI–TOF/TOF analyzer, including 32 (fESCs vs. pESCs), 28 (fESCs vs. aESCs) and 17 (pESCs vs. aESCs) prominent proteins with significantly higher or lower expression relative to the comparison cells. Among the 32 proteins from fESCs, 12 were significantly increased in expression and 20 were reduced in expression in fESCs com-pared with pESCs. Similarly, 10 of 28 and 8 of 17 proteins were more highly expressed in fESCs and pESCs compared with aESCs, respectively. In contrast, 18 of 28 and 9 of 17 proteins were reduced in expression in fESCs and pESCs compared with aESCs, respectively. Of the eight protein candidates in fESCs, four were in-creased and four were decreased in expression relative to both pESCs and aESCs in the 2-DE analysis. Differential expression of these pro-teins were confirmed by mRNA expression analysis using real time RT-PCR. For three pro-teins, ANXA5, CLIC1 and SRM, Western blot analysis corroborated the expression patterns indicated by the 2-DE results. ANXA5 and CLIC1 were increased in expression and SRM was de-creased in expression in fESCs compared with both pESCs and aESCs. The differentially ex-pressed proteins identified in the present study warrant further investigation towards the goal of their potential application in ESC therapy.

Design of Bridge Expansion Joints with Perforated Dowels Under Impact Loading

The expansion joints are expected to have movement capacity, bearing capacity for static and dynamic loading, water-tightness, low noise emission and traffic safety. In particular, the failure due to impact loading is the main reason for the observed damages. The problem of dynamic behavior of the expansion joints is so complex that we shall focus our attention on the impact factor for vehicle load that is governed by traffic impact. In order to overcome this difficulty, the cantilever-toothed aluminum joint (finger joint) is one of the promising joints under impact loading. In this study, from the viewpoint of design methodology, numerical studies for impact behavior were conducted for aluminum alloy expansion joints with perforated dowels. The design impact factor for the expansion joints with the perforated dowels against traffic impact loading was examined by using numerical simulations.

In design phases, expansion joints are required to have movement capacity, bearing capacity for static and dynamic loading, watertight, low noise emission and traffic safety. On the basis of the fact that failure due to dynamic loading is the main reason for the observed damages, attention is focused on the bearing capacity for dynamic loading governed by impact, because it differs from the static loading. In this study, from the viewpoint of durability, experimental studies for dynamic behavior were conducted for aluminium alloy expansion joints with perforated dowels. The validity of the perforated dowels against traffic impact loading was confirmed by both experimental and numerical studies.

Effect of acupuncture at acupoint of Shenshu (BL 23) on the bones of ovariectomized rats

OBJECTIVE: To investigate the effect of acupuncture at acupoint of Shenshu(BL 23) in ovariectomized rats and sham-operated ***: Female Sprague-Dawley rats were divided into the following four groups(n = 12 each):sham operation only(Sham), sham operation with acupuncture(Sham + ACP), ovariectomy only(OVX), and ovariectomy with acupuncture(OVX +ACP). Operations were performed at the 9 th week of age. Acupuncture of Shenshu(BL 23) was started at the 11 th week of age and conducted 3 times per week until the 26 th week of ***: Among ovariectomized rats, the acupuncture-treated rats had significantly lower bodyweights and cross-linked N-telopeptide of type 1 collagen levels in the urine after 3 weeks of acupuncture treatment, higher bone mineral density of the lumbar vertebra after 7 weeks, and smaller trabecular separation of the sixth lumbar vertebra,measured by micro-computed tomography, after17 weeks than control rats. These effects were not observed in sham-operated rats. Blood concentrations of estradiol were significantly higher in the OVX + ACP group than in the untreated group. Acupuncture treatment increased plasma testosterone levels in sham-operated rats, while this increase was not observed in OVX ***: Acupuncture of Shenshu(BL 23)prevents bone loss and structural changes by suppressing bone resorption.
