Phenolics, fatty acids composition and biological activities of various extracts and fractions of Malaysian Aaptos aaptos

Objective: To investigate phenolics, fatty acids composition and biological activities of various extracts and fractions of Malaysian Aaptos aaptos. Methods: Fatty acid methyl ester was analyzed by gas chromatography-flame ionization detector. Antioxidant activity was determined using 2,2-diphenyl-picrylhydrazyl radical scavenging assay and total phenolics content by Folin-Ciocalteu procedure. Vero cells viability was evaluated using methyl thiazole tetrazolium and the inactivation of herpes simplex virus type 1 by neutral red uptake assay. p-Hydroxybenzamide isolated by column chromatography was characterized by utilizing nuclear magnetic resonance spectroscopy and electron impact mass spectrometry. Results: The chloroform, ethyl acetate and methanol extracts of Aaptos aaptos produced higher portions of straight-chain saturated fatty acid, while hexane extract mainly consisted of unsaturated fatty acid. The five majors of fatty acid methyl ester were identified as behenic acid, cis-10-heptadecenoic acid and cis-10-pentadecenoic acids, palmitic acid and tricosanoic acid. In addition, among all organic extracts, chloroform extract inactivated herpes simplex virus type 1 while exhibited weak cytotoxic activity against normal Vero cells and also exhibited strong cytotoxic activity on HL-60, MCF-7, K562, CEM-SS and WEHI-3 B cells. A phenolic compound, p-hydroxybenzamide was also isolated from the sponge. Conclusions: Aaptos aaptos could be a source to derive the potential antiviral and anticancer agents. However, further studies are needed to determine the mechanism involved in the process.

Malaysia's rapid economic and demographic development have placed negative pressure on its water supplies and the quality of the Juru River, which is close to the nation's capital and its major source of water. Healthy aquatic ecosystems are supported by physicochemical properties and biological diversity. This study evaluated the anthropogenic impacts on aquatic biodiversity, especially plankton, fish, and macrobenthos, as well as the water quality of the Juru River in the Penang area. Aquatic biodiversity and river water parameters were collected from ten sampling stations along the Juru River. Seven variables were used to assess the physicochemical environment: pH, temperature, total suspended solids (TSS), salinity, dissolved oxygen (DO), biochemical oxygen demand (BOD), and chemical oxygen demand. At each sampling station, the total number of plankton, fish, and macrobenthic taxa were counted and analyzed. The relationships between the physicochemical parameters and aquatic biodiversity were investigated with biotypological analysis, principal component analysis, hierarchical cluster analysis, and linear regression analysis. These analyses showed that the richness and diversity indices were generally influenced by salinity, temperature, TSS, BOD, and pH. The data obtained in this study supported the bioindicator concept. The findings, as they related to scientifically informed conservation, could serve as a model for Juru River management, as well as for river management throughout Malaysia and other tropical Asian countries.

Objective:To evaluate antioxidant and antimicrobial activities of stembark of Shorea kunstleri(***)together with analysis of phytochemical and total phenolic ***:Extraction was conducted with different solvent polarity of n-hexane,dichloromethane(DCM)and methanol by using Soxhlet *** phenolic content was determined using Folin-Ciocalteu *** radical scavenging activity and inhibition of lipid peroxidation were evaluated with DPPH radical scavenging and ferric thiocyanate assays,*** activities were performed using disc diffusion method,minimum inhibition concentration(MIC),minimum bactericidal concentration(MBC),and minimum fungicidal ***:*** stembark extracts revealed presence of steroids,terpenoids,saponins,flavonoids,and phenolic *** extract exhibited the highest total phenolic content and free radical scavenging activity resulting in phenolic content of(8.340±0.003)g GAE/100 g of extract and(95.90±1.07)%DPPH inhibition(IC_(50)value of 18.6μg/mL),*** thiocyanate assay of n-hexane,DCM,and methanol extracts indicated lipid peroxidation inhibitory activity of(74.20±0.35)%,(74.00±0.10)%,and(72.80±0.27)%,*** antimicrobial and antifungal tests,methanol extract showed inhibition against Staphylococcus aureus(***),Candida albicans,and Candida tropicalis with inhibition zones of 10-12,18-22,and 18-19 mm,*** MIC test of methanol extract showed highest inhibition against Candida albicans and ***(0.04 and 0.08 mg/mL,respectively)while DCM extract exhibited the highest activity towards Candida tropicalis(MIC value of 0.63 mg/mL).Taken together,MBC test of methanol extract strongly demonstrated bactericidal effect against *** with MBC value of 0.08 mg/***:The study demonstrated that stembark extracts of *** possessed antioxidant and antimicrobial properties.
