Structure-based Comparative Analysis and Prediction of n-linked Glycosylation Sites in Evolutionarily Distant Eukaryotes

The asparagine-X-serine/threonine （nXS/T） motif, where X is any amino acid except proline, is the consensus motif for n-linked glycosylation. Significant numbers of high-resolution crystal structures of glycosylated proteins allow us to carry out structural analysis of the n-linked glycosylation sites （nGS）. Our analysis shows that there is enough structural information from diverse glycoproteins to allow the development of rules which can be used to predict nGS. A Python-based tool was developed to investigate asparagines implicated in n-glycosylation in five species： Homo sapiens, Mus musculus, Drosophila melanogaster, Arabidopsis thaliana and Saccharo- myees cerevisiae. Our analysis shows that 78 % of all asparagines of nXS/T motif involved in n-gly- cosylation are localized in the loop/turn conformation in the human proteome. Similar distribution was revealed for all the other species examined. Comparative analysis of the occurrence of nXS/T motifs not known to be glycosylated and their reverse sequence （S/TXn） shows a similar distribu- tion across the secondary structural elements, indicating that the nXS/T motif in itself is not bio- logically relevant. Based on our analysis, we have defined rules to determine nGS. Using machine learning methods based on these rules we can predict with 93% accuracy if a particular site will be glycosylated. If structural information is not available the tool uses structural prediction results resulting in 74% accuracy. The tool was used to identify glycosylation sites in 108 human proteins with structures and 2247 proteins without structures that have acquired nXS/T site/s due to non-synonymous variation. The tool, Structure Feature Analysis Tool （SFAT）, is freely available to the public at http：//***/tools/sfat.

A light weight and portable automatic linear scraping tool powered by an oscillating motor was developed to substitute the traditional hand scraping process. Its performance in terms of surface finish and material removal rate was compared with the commercially available angular scraping and hand scraping technique. Mild steel blocks finished by face milling were subjected to the three types of scraping. 2D surface profiles and the material removal rates of resulting surfaces were measured. The results point out to linear scraping as the best among the three scraping processes as the resulting surface parameters （Ra, Rq, Rpk, Rk and Rvk） are superior to the other two processes. Hence the resulting surface can give better tribological performance as it can be less susceptible to wear. The linear scraping tool was also found ergonomic better than hand scraping and angular scraping by reducing operator effort and increasing the scraping efficiency.

Effect of hydroalcoholic extract of Hibiscus rosa sinensis *** in experimental colitis in rats

Objective:To elucidate the ameliorative effect of liydrualcuholic extract of leaves of Hibiscus rosa sinensis(HRS) in acetic acid induced experimental colitis in male wistar ***: The animals were administered with 2 mL acetic acid(4%) via intra *** animals were divided into various treatment groups(n=6).Prednisolone was used as standard drug and HRS was administered at a dose of 50,100 and 200 mg/kg *** control group of animals received 1 mL of vehicle(distilled water).Ulcer area,ulcer index,spleen weight,colon weight to length ratio, macroscopic score,haemalological parameters,colonic superoxide dismutase(SOD),glutathione(GSH),myeloperoxidase(MPO),malondialdchyde(MDA),tumor necrosis factor-α(TnK- α), nitric oxide(nO) and histological changes were recorded after the treatment regimen of 11 days. Results:Intrarectal instillation of acetic acid caused enhanced ulcer area,ulcer index,spleen weight,colon weight to length ratio,colonic MPO,MDA,nO and TnF-α It caused significant decreased level of SOD and *** with HRS for 7 days exhibited significant effect in lowering of oxidative stress,colonic nO,TnF- α and elevation of SOD and GSH at a dose of 100 and 200 mg/kg in acetic acid induced ***:The present investigation demonstrates HRS is of potent therapeutic value in the amelioration of expcrimenlal colilis in laboralory animals by inhibiting the proinflammatory mediator like nO and TnK-α.

目的比较双对吻挤压（DK crush）和经典挤压技术治疗冠状动脉分叉病变的临床效果。方法311例真性分叉病变患者随机分入DK crush组（n=155）和经典挤压组（n=156），随访时间8个月。一级及二级终点分别为主要心脏不良事件（MACE，包括心肌梗死、心原性死亡和靶病变血运重建）和血管直径再狭窄及晚期丢失。结果DK crush组糖尿病患者较多。经典挤压组及DK crush组最终对吻扩张（FKBI）成功率分别为76％和100％（P〈0．001）。DK crush术式的不足包括造影剂用量大（P=0．04）、球囊数量多（P〈0．01）、手术时间长（P〈0．001），但是对吻扩张不满意率显著减少（27．6％比6．3％，P〈0．01）。临床随访率为100％，冠状动脉造影随访率为82％。经典挤压组累计再狭窄率为32．3％，而DK crush组为20．3％（P=0．01），经典挤压组分支血管再狭窄率高（24．4％比12．3％，P=0．01），而两组间主干血管再狭窄率差异无统计学意义。经典挤压组术后8个月时的累计MACE发生率为24．4％（FKBI失败组为35．9％，FKBI成功组为19．7％），显著高于DK crush组（11．4％，P=0．02）。经典挤压组血栓栓塞率为3．2％（FKBI失败组为5．1％，FKBI成功组为1．7％），而DK crush组为1．3％（P〉0．05）。经典挤压组术后8个月时无靶病变血运重建生存率为75．4％（FKBI失败组为71．2％，FKBI成功组为77．6％），而DK crush组为89．5％（P=0．002）。结论DKcrush可能是治疗冠状动脉分又病变的较佳术式。
