ABC transporters are molecular machines which power the solute transport using ATP *** structural biology of ABC transporters has been exploding for the last few years,and this study explores timelines and trends for various attributes such as structural tools,resolution,fold,sources,and group *** study also evidences the significance of mammalian expression systems,advancements in structural biology tools,and the developing interest of group leaders across the world in the remarkably progressing *** field started in 2002 and bloomed in 2016,and COVID years were really productive to the ***,the study explores 337 structures of 58 unique ABC transporters deposited in the PDB database from which P-glycoprotein has the largest number of ***,62%of total structures are determined at the resolution of 3-4Åand 53%of structures belong to fold IV *** progressive advancements in the field,the field is shifting from prokaryotic to eukaryotic sources and X-ray crystallography to cryoelectron *** the nutshell,this study uniquely provides the detailed snapshot of the field of structural biology of ABC transporters with real-time data.

Recurrent intrathecal catheter-tip granuloma in a patient receiving high dose hydromorphone: A case report

Study Design: Case report. Patient Sample: A 42- year-old Caucasian male. Results: Catheter-tip granuloma formation is possible despite a normal catheter access study in patients with intrathecal (IT) infu- sion systems and its recurrence is possible after surgical excision. Increasing concentrations of hydromorphone from 50 mg/ml to 100 mg/ml without altering the daily dosage may have precipitated granuloma formation. Conclusions: In patients with previously implanted spinal catheters for intrathecal drug delivery, catheter access studies cannot be relied upon to rule out catheter-tip granulomas, which should be included in the differential diagnosis in patients with worsening clinical conditions or new onset neurologic symptoms. Caution should be exercised when increasing concentrations of intrathecal opioids while monitoring for signs or symptoms of intrathecal catheter- tip granuloma formation. Removal of previously implanted catheters and/or reductions in opioid concentrations may be necessary to prevent recurrent granuloma formations, which can occur quickly after surgical excision.

Degradation of Dyestuff Pollutant Sudan I Using Advanced Oxidation Process

Due to an increasing environmental pollution, a search for the cost effective treatment and disposal of the dyes from the textile effluents is getting more and more importance. Oxidation and reduction processes play important roles in the degradation treatments of the azo dyes. The latter process is more effective and in consequence its mechanism is also better understood. The mechanism of the oxidation processes, the intermediates involved in these reactions and their role in the effectiveness of the oxidative degradation of the azo dyes, viz, phenyl azo b-naphthol (PAN), Sudan I. On exposure to sunlight at 2 1/2 hours for various samples in different concentrations of PAN mixed with Fenton reagent, when the reactive intermediate?reacted with the colour, the pH vs. absorbance generally showed significant degradation in between pH 5 and 6. The results were compared with the same samples on exposure to uv-light of 254 nm and irradiated at 20 minutes. The degradation occurred in samples of relatively high concentrations, viz, 10-3 and 5 × 10-4 mol· dm-3 at near neutral pH 6 whereas. Low concentration samples such as 10-4 and 5 × 10-5 mol·dm-3 showed degradation towards more acidic range of pH 2 to 4. In advanced oxidation process (AOP), generally reactive, strongly oxidizing ·OH radicals play a main role in destruction of the dye molecules. The proposed mechanisms and the rate coefficients for the reactions of ·OH intermediates with the dye molecules and with model compounds are summarized.

Background Gemcitabine plus cisplatin is a standard treatment for stages IIIB and IV nonsmall cell lung cancer （NSCLC）. This randomized phase Ⅱ study evaluated a 3-week versus a 4-week schedule of gemcitabine-cisplatin as first line treatment for Chinese patients with advanced NSCLC. Methods Patients were randomized to receive cisplatin 75 mg/m^2 on day 1 plus either gemcitabine 1250 mg/m^2 on days 1 and 8 of a 21-day cycle （3-week group） or gemcitabine 1000 mg/m2 on days 1,8 and 15 of a 28-day cycle （4-week group）. Results One hundred patients were enrolled in this study. The response rate was 24% （12/51 patients） in the 3-week group and 27% （13/49 patients） in the 4-week group. There were no statistically significant differences between the two treatment groups in survival （hazard ratio： 1.19; 95% CI： 0.68-2.09） with a median survival of 12.1 months and 13.8 months in the 3-week group and the 4-week group respectively. The rate of grade 3/4 toxicity in the 3-week group was 55% compared with 86% in the 4-week group （P=0.001）. The difference in the incidence of grade 3/4 haematological toxicities did not reach statistical significance （3-week： 37%, 4-week： 57%）, however grade 3/4 drug related neutropenia （3-week： 27%, 4-week： 51%） and thrombocytopenia （3-week： 8%, 4-week： 31%） were significantly lower in the 3-week group. Grade 3/4 nonhaematological toxicities were less in the 3-week group （33% cf 63%; P=0.005）. Conclusions The differences in the efficacy endpoints were all in favour of the 4-week schedule of gemcitabine plus cisplatin, however these differences did not reach statistical significance. Fewer grade 3/4 toxicities were observed in the 3-week group compared with the 4-week group.

Mechanical and Dielectric Properties of InTe Crystals

The mechanical properties of indium telluride (InTe) crystals grown by the Bridgman technique were investigated at room temperature using a Vickers hardness tester. The microhardness is observed to vary nonlinearly with the applied load, 10 - 100 g. The cleaved ingots are found to have high value of microhardness (222.44 kg/mm2 at a load of25 g), which reflects an appreciable degree of strength due to their covalent bonding and homogeneity. The studies revealed that the dislocations in the grown crystals offered a resistance to fresh dislocations due to interaction. At higher loads, plastic deformation induces by slip, exhibiting a decrease in hardness from the peak value. The dielectric constant and dielectric loss of indium telluride crystals were evaluated in the frequency range, 1 kHz - 1 MHz for different temperatures (35oC - 140oC). The frequency dependence of AC conductivity was analyzed as a function of temperature. The effect of temperature and frequency on the dielectric response of InTe crystals has been assessed on their cleavage faces and the obtained results are discussed.

Background Distal pancreatic carcinoma is one of the most lethal cancers largely due to its high incidence of distant *** study aims to assess the prognostic value of splenic-vasculature involvement in resected distal pancreatic *** In this retrospective study,we collected the clinicopathologic information of 454 patients with pancreatic cancer and performed univariate and multivariate analyses to identify factors associated with progression-free survival(PFS)and overall survival(OS),with an emphasis on the prognostic value of splenic-artery and-vein *** Univariate analysis revealed that larger tumor size,non-intraductal papillary mucinous neoplasm(non-IPMN)-associated adenocarcinoma,poor differentiation,stage pT3,nodal metastasis,lymphovascular invasion,perineural invasion,and pathologic and radiographic evidence of splenic-vein invasion were significantly associated with shorter PFS and OS(all P<0.05).Multivariate analysis confirmed non-IPMN-associated adenocarcinoma,stage pT3,stage pN1-2,and postoperative adjuvant chemotherapy as independent risk factors for both PFS and OS,and larger tumor size and radiographic evidence of splenic-artery invasion as predictors of PFS *** Guidelines should be developed for a uniform approach with regard to the examination and reporting of the status of the splenic vasculature when dealing with distal-pancreatic-cancer specimens.
