Progress in blood biomarkers of subjective cognitive decline in preclinical Alzheimer’s disease

Alzheimer's disease(AD)is a neurodegenerative disease that gradually impairs cognitive functions.Recently,there has been a conceptual shift toward AD to view the disease as a continuum.Since AD is currently incurable,effective intervention to delay or prevent pathological cognitive decline may best target the early stages of symptomatic disease,such as subjective cognitive decline(SCD),in which cognitive function remains relatively intact.Diagnostic methods for identifying AD,such as cerebrospinal fluid biomarkers and positron emission tomography,are invasive and expensive.Therefore,it is imperative to develop blood biomarkers that are sensitive,less invasive,easier to access,and more cost effective for AD diagnosis.This review aimed to summarize the current data on whether individuals with SCD differ reliably and effectively in subjective and objective performances compared to cognitively normal elderly individuals,and to find one or more convenient and accessible blood biomarkers so that researchers can identify SCD patients with preclinical AD in the population as soon as possible.Owing to the heterogeneity and complicated pathogenesis of AD,it is difficult to make reliable diagnoses using only a single blood marker.This review provides an overview of the progress achieved to date with the use of SCD blood biomarkers in patients with preclinical AD,highlighting the key areas of application and current challenges.

Chloromethyl pivalate based electrolyte for non-aqueous lithium oxygen batteries

A novel electrolyte with chloromethyl pivalate （CP） used as solvent was first reported for non-aqueous lithium-oxygen （li-O2） batteries. Since there are no α-H atoms in the structure of CP, the CP based electrolyte in both superoxide radical solution and real lifO2 battery environment showed good chemical stability against superoxide radicals, which was confirmed by ^1H NMR and ^13C NMR measurements. Without a catalyst in the cathode of li-O2 batteries, the batteries showed high specific capacity and cycling stability.

Prospects and challenges of dynamic DNA nanostructures in biomedical applications

The physicochemical nature of DNA allows the assembly of highly predictable structures via several fabrication strategies, which have been applied to make breakthroughs in various fields. Moreover, DNA nanostructures are regarded as materials with excellent editability and biocompatibility for biomedical applications. The ongoing maintenance and release of new DNA structure design tools ease the work and make large and arbitrary DNA structures feasible for different applications. However, the nature of DNA nanostructures endows them with several stimulus-responsive mechanisms capable of responding to biomolecules, such as nucleic acids and proteins, as well as biophysical environmental parameters, such as temperature and p H. Via these mechanisms, stimulus-responsive dynamic DNA nanostructures have been applied in several biomedical settings, including basic research, active drug delivery, biosensor development, and tissue engineering. These applications have shown the versatility of dynamic DNA nanostructures, with unignorable merits that exceed those of their traditional counterparts, such as polymers and metal particles.However, there are stability, yield, exogenous DNA, and ethical considerations regarding their clinical translation. In this review, we first introduce the recent efforts and discoveries in DNA nanotechnology, highlighting the uses of dynamic DNA nanostructures in biomedical applications. Then, several dynamic DNA nanostructures are presented, and their typical biomedical applications,including their use as DNA aptamers, ion concentration/p H-sensitive DNA molecules, DNA nanostructures capable of strand displacement reactions, and protein-based dynamic DNA nanostructures, are discussed. Finally, the challenges regarding the biomedical applications of dynamic DNA nanostructures are discussed.

Hard tissue stability after guided bone regeneration: a comparison between digital titanium mesh and resorbable membrane

Guided bone regeneration(GBR)uses resorbable and non-resorbable membranes as biological barriers.This study compared the differences in hard tissue stability between GBR using evidence-based digital titanium mesh and resorbable collagen membranes during implant placement.A total of 40 patients(65 implant sites)were enrolled and divided into two groups:resorbable membrane and digital titanium mesh groups.The alveolar bone was analyzed at two-and three-dimensional levels using conebeam computed tomography and by reconstructing and superimposing the hard tissues at four time points:preoperatively,postoperatively,before second-stage surgery,and 1 year after loading.The use of digital titanium mesh showed less alveolar bone resorption in vertical and horizontal directions two-dimensionally before the second-stage surgery and 1 year after loading.Regarding volumetric stability,the percentage of resorption after 6 months of healing with resorbable membrane coverage reached 37.5%.However,it was only 23.4%with titanium mesh.Although postoperative bone volume was greater at all labial sites with resorbable membrane than with digital titanium mesh,after substantial bone resorption within 1 year of loading,the labial bone thickness at the upper part of implants was thinner with resorbable membrane than with digital titanium mesh.Furthermore,digital titanium meshes made according to ideal bone arch contour reduced soft tissue irritation,and the exposure rate was only 10%.Therefore,although both resorbable membrane and digital titanium mesh in GBR were able to successfully reconstruct the bone defect,digital titanium meshes were better at maintaining the hard tissue volume in the osteogenic space.

Brightness and lifetime Improved light-Emitting Diodes from Sr-Doped Quasi-Two-Dimensional Perovskite Layers

Green Perovskite light-Emitting Diodes(PeLEDs)have attracted wide attention for full spectrum displays.However,the inferior film morphology and luminescence property of quasi-two-dimensional(quasi-2D)perovskite layers limit the photoelectric property of the PeLEDs.In this paper,the effect of strontium(Sr)doped in quasi-2D perovskite layers is investigated to obtain a high-quality active layer.The morphologies and optical properties of Sr-doped quasi-2D perovskite films with different concentrations are studied.With the addition of strontium,more low-dimensional-layer perovskite phases(n D 2 and n D 3)appear in quasi-2D perovskite films,providing efficient intraband carrier funneling pathway and facilitating radiative recombination.The photoluminescence(PL)peak intensity of optimized Sr-doped quasi-2D perovskite layers increases 50%compared with the non-strontium counterpart.Moreover,green PeLEDs based on a Sr-doped quasi-2D perovskite layer reach a maximum luminance(Lmax)of 2943.77 cd/m^(2),which is three times of the control device.The electroluminescence(EL)peaks of Maximum External Quantum Efficiency(MEQE)and Lmax of Sr-doped PeLEDs exhibite a slight shift,indicating the excellent stability and performance of Sr-doped devices.The optimized device can continuously operate for 360 s at MEQE driving voltage,resulting in a half-lifetime of60 s,which is 3-fold greater than that of the control PeLEDs.

A tetrahedral framework nucleic acid based multifunctional nanocapsule for tumor prophylactic mRNA vaccination

Synthetic antigen-encoding mRNA plays an increasingly significant role in tumor vaccine technology owing to its antigen-specific immune-activation. However, its immune efficacy is challenged by inferior delivery efficiency and demand for suitable adjuvants. Here, we develop a novel mRNA nanovaccine based on a multifunctional nanocapsule, which is a dual-adjuvant formulation composed of cytosine-phosphateguanine motifs loaded tetrahedral framework nucleic acid(CpG-tFNA) and an immunopeptide murine β-defensin 2(mDF2β). This m RNA nanovaccine successfully achieves intracellular delivery, antigen expression and presentation of dendritic cells, and proliferation of antigen-specific T cells. In a tumor prophylactic vaccination model, it exerts an excellent inhibitory effect on lymphoma occurrence through cellular immunity. This mRNA nanovaccine has promising prophylactic applications in tumors and many other diseases.

High-speed impulsive stimulated Brillouin microscopy

Brillouin microscopy, which maps the elastic modulus from the frequency shift of scattered light, has evolved to a faster speed for the investigation of rapid biomechanical changes. Impulsive stimulated Brillouin scattering (ISBS)spectroscopy has the potential to speed up measurement through the resonant amplification interaction from pulsed excitation and time-domain continuous detection. However, significant progress has not been achieved due to the limitation in signal-to-noise ratio (SNR) and the corresponding need for excessive averaging to maintain high spectral precision. Moreover, the limited spatial resolution also hinders its application in mechanical imaging. Here, by scrutinizing the SNR model, we design a high-speed ISBS microscope through multi-parameter optimization including phase, reference power, and acquisition time. Leveraging this, with the further assistance of the Matrix Pencil method for data processing, three-dimensional mechanical images are mapped under multiple contrast mechanisms for a millimeter-scale polydimethylsiloxane pattern immersed in methanol, enabling the identification of these two transparent materials without any contact or labeling. Our experimental results demonstrate the capability to maintain high spectral precision and resolution at a sub-millisecond integration time for one pixel. With a two-order improvement in the speed and a tenfold improvement in the spatial resolution over the state-of-the-art systems, this method makes it possible for ISBS microscopes to sensitively investigate rapid mechanical changes in time and space.

980 nm Near-Infrared light-Emitting Diode Using All-Inorganic Perovskite Nanocrystals Doped with Ytterbium Ions

All-inorganic perovskite(CsPbX3)nanocrystals(NCs)have recently been widely investigated as versatile solution-processable light-emitting materials.Due to its wide-bandgap nature,the all-inorganic perovskite NC light-Emitting Diode(LED)is limited to the visible region(400-700 nm).A particularly difficult challenge lies in the practical application of perovskite NCs in the infrared-spectrum region.In this work,a 980 nm NIR all-inorganic perovskite NC LED is demonstrated,which is based on an efficient energy transfer from wide-bandgap materials(CsPbCl3 NCs)to ytterbium ions(Yb3+)as an NIR emitter doped in perovskite NCs.The optimized CsPbCl3 NC with 15 mol%Yb3+doping concentration has the strongest 980 nm photoluminescence(PL)peak,with a PL quantum yield of 63%.An inverted perovskite NC LED is fabricated with the structure of ITO/PEDOT:PSS/poly-TPD/CsPbCl3:15 mol%Yb3+NCs/TPBi/liF/Al.The LED has an External Quantum Efficiency(EQE)of 0.2%,a Full Width at Half Maximum(FWHM)of 47 nm,and a maximum luminescence of 182 cd/m?.The introduction of Yb3+doping in perovskite NCs makes it possible to expand its working wavelength to near-infrared band for next-generation light sources and shows potential applications for optoelectronic integration.
