Inhibiting SHP2 reduces glycolysis, promotes microglial M1 polarization, and alleviates secondary inflammation following spinal cord injury in a mouse model

Reducing the secondary inflammatory response, which is partly mediated by microglia, is a key focus in the treatment of spinal cord injury. Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.

Preparation and characterization of Rb-doped TiO_(2) powders for photocatalytic applications

TiO_(2)nanoparticles doped with different concentrations of Rb were prepared by sol–gel method.The crystal structures of samples were characterized by X-ray diffraction(XRD)and transmission electron microscopy(TEM).It was confirmed by the Rietveld refinement method that the as-prepared samples crystallize in anatase phase and Rb doping induces expansion and distortion of the crystal lattice.Compared with the pure TiO_(2),the Rb-doped nanoparticles have smaller crystal size and better dispersibility.Degradation rate of methylene blue was used to evaluate the photocatalytic activity of Rb–TiO_(2).It indicated that doping Rb into TiO_(2)can effectively improve the photocatalytic activity of TiO_(2)and the mechanisms were discussed in detail.When using the sample doped with 0.20%Rb,methylene blue degraded with the fastest rate(97%within 60 min).

Hematuria after nocturnal exercise of a man: A case report

BACKGROUND A man experienced multiple episodes of macroscopic hematuria following nocturnal exercise.Urinary stones and tumors were considered the two most likely causes.The patient had two hobbies:Consuming health care products in large quantities and engaging in late-night running.CASE SUMMARY Health care products contain a large amount of calcium phosphate,and we hypothesize that this could induce the formation of small phosphate stones.After exercise,the urinary system is abraded,resulting in bleeding.The patient was advised to stop using the health care products.Consequently,the aforementioned symptoms disappeared immediately.However,the patient resumed the above two habits one year later;correspondingly,the macroscopic hematuria reap-peared.CONCLUSION This finding further confirmed the above inference and allowed for a new avenue to determine the cause of the patient’s hematuria.

Decreased Nrf2 protein level and low sperm quality in intractable spermatocystitis

To investigate the molecular etiology of low sperm quality in patients with intractable spermatocystitis,spermatozoa samples from patients with persistent hematospermia undergoing transurethral seminal vesiculoscopy and healthy volunteers were utilized.Spermatozoa samples were collected from the seminal vesicles through transurethral seminal vesiculoscopy or by masturbation ejaculation.Sperm quality was analyzed by a WLJY-9000 color semen analysis system.Measurement of tumor necrosis factor alpha(TNFa)and interleukin-6(IL-6)in the seminal plasma was performed using enzyme-linked immunosorbent assay(ELISA).Measurement of H_(2)O_(2)in the seminal plasma was performed with a hydrogen peroxide kit.The protein levels of nuclear factor erythroid 2-related factor 2(Nrf2)and phosphorylated-Nrf2(p-Nrf2)were measured by western blot analysis and immunofluorescence assays.Low sperm quality parameters and increased levels of inflammatory cytokines(TNFa,IL-6,and H_(2)O_(2))in the seminal plasma were detected among the semen samples from the patients with persistent hematospermia.Nrf2 and p-Nrf2 were strongly expressed in the nucleus and periphery of human sperm cells,according to the results of the immunofluorescence assays.The protein levels of Nrf2 and p-Nrf2 were significantly lower in the spermatozoa samples from patients with persistent hematospermia than in those from healthy volunteers with normal sperm motility.The results suggested that Nrf2 signaling might play a role in the low sperm quality of patients with intractable spermatocystitis.

Investigation of the effect of filling level on the wear and vibration of a SAG mill by DEM

Semi-autogenous grinding(SAG)mill is widely used in the grinding process of coal and ores because of its strong applicability,large capacity and low consumption of grinding media.Therefore,the research of SAG mill is still of great important practical significance.Among those parameters of the SAG mill,the flling level is very important for it is not only directly related to the processing capacity of the mill,but also affects the operation cost and stability of mill.In this paper,combined with shear impact energy model(SIEM)and discrete element method(DEM),the particle collision energy spectrum was applied to analyze the influence of ore flling level and media flling level on the power draw of the mill,the impact energy on ore,and the wear of liner and media in a slice of an industrial SAG mill.It was found that increasing the ore flling level would increase energy consumption and reduce the energy efficiency and the wear of liner and grinding media.Furthermore,the vibration of mill was explored from the collision energy on liner.The results showed that the low frequency but high energy ollision between grinding media and liner was the main reason for liner wear and mill vibration.

Herein,we report a novel heterogeneous photocatalyst for radical-mediated oxidative oxysulfonylation of alkynes toβ-keto sulfones under environmentally benign conditions.The oxygen vacancy-rich semiconductor Nb2O5(labeled as OVs-N-Nb_(2)O_(5))could efficiently catalyze a wide range of alkynes,especially for those bearing electron-deficient substituents or internal alkynes,to their correspondingβ-keto sulfones in good to high yields with good tolerance of diverse functional groups under visible-light illumination.The late-stage modification of steroidal compounds and synthesis of bioactive molecules were also achieved via this procedure,highlighting its potential for practical applications.Meanwhile,the photocatalyst OVs-N-Nb_(2)O_(5) showed outstanding catalytic stability for successive recycles without appreciable loss in activity and selectivity.The critical role of oxygen vacancies on improving reaction activity and selectivity was clearly disclosed via control experiments and theoretical calculation.

Multiple Automatic Guided Vehicle(multi-AGVs)management systems provide an effective solution to ensuring stable operations of multi-AGVs in the same scenario,such as flexible manufacturing systems,warehouses,container terminals,etc.This type of systems need to balance the relationship among the resources of the system and solve the problems existing in the operation to make the system in line with the requirement of the administrator.The multi-AGVs management problem is a multi-objective,multi-constraint combinatorial optimization problem,which depends on the types of application scenarios.This article classifies and compares the research papers on multi-AGVs management in detail.Firstly,according to the different dimensions of the problem,the multi-AGVs management system is analyzed from three perspectives,namely,1)task dimensiondispatch,2)spatial dimension-path,and 3)time dimension-scheduling.The detailed comparison between the three dimensions and their respective solutions are discussed in detail as well.Secondly,according to their utility,the multi-AGVs management problems are divided into three categories:1)cost reduction,resource-oriented,2)efficiency improvement,problem-oriented,3)personalized demand,goal-oriented.The algorithm and methods of the different utility-oriented are analyzed and discussed.The related literature is summarized and corresponds to the composition of the multi-AGVs management system and the multi-AGVs management problems.Finally,according to the literature review,suggestions are made for further research.

An image processing method for feature extraction and dynamic tracking of particle clusters in CFBs

A new image processing method based on the high-speed camera is proposed to identify,locate,and track clusters.The instantaneous characteristic parameters of particle clusters in the riser of the circu-lating fluidized bed(CFB)can be acquired,such as solids holdup,vertical velocity,lateral displacement,aspect ratio and near-circularity.Experiments were carried out with glass bead particles,river sand particles and FCC particles.The time series of images of gas-solid flow in a CFB riser with a 100 mm x 25 mm cross-section and 3.2 m in length were obtained using high-speed cameras.The k-means++clustering algorithm is utilized to identify the clusters,centroid is applied to locate the clusters,and the cross-correlation algorithm is employed to track the specific clusters and number them to get the instantaneous characteristic parameters.The results illustrate that the shapes of clusters in the center area are closest to circle,moving upwards at a uniform speed,while the clusters in the side-wall area are mostly elongated or long chain-like,moving slowly downwards.In the transition area,the clusters are more complex,moving upwards at a constant speed,and having large lateral displacement.The results show that the image processing method used in this study is successful in acquiring the dynamic and structural parameters of the clusters simultaneously.

Immobilization of GO_(X) on PEG/fluoresence functionalized nanographene oxide to describe fluctuation of glucose level

Enzyme,produced and worked in all living things,could work as macromolecular biological catalysts in diverse biochemical processes with particular specificity,like glucose oxidase(GOX).The efficient use of enzyme properties has great importance in pharmaceutics and therapeutics.In this work,we could fabricate naive and effective electrochemical biosensors in the determination of glucose levels via utilizing GOX.Graphene oxide,as a water-soluble derivative of graphene,has shown great promise in a variety of biomedical applications including biosensors.Thus,we established a new-type special platform for GOX immobilization to perform its prosperities,in which nanographene oxide(nGO)was employed as an ideal base and poly(ethylene glycol)(PEG)was conjugated on the edge of nGO sheets to enhance its biocompatibility.Additionally,preferable functional dyes(Rhodamine B/fluorescein isothiocyanate)were also introduced to the platform.Enzyme-nanocomposites were then provided by locating GOX on the platform,i.e.,GOX@nGO-PEG-RhB and GOX@nGO-PEG-FITC.The microstructure and composite of platforms and enzyme-nanocomposites were confirmed by diverse characterizations.Finally,on account of corresponding cyclic voltammetric and typical ready-state amperometric curves,it was informed that GOX@nGO-PEG-RhB and GOX@nGO-PEG-FITC could effectively respond to the fluctuation of glucose level as electrochemical biosensor.The present work presents special platforms for the immobilization of enzymes like GOX and provides new-type biosensors in the detection of glucose levels.

General closed-form inverse kinematics for arbitrary three-joint subproblems based on the product of exponential model

The inverse kinematics problems of robots are usually decomposed into several Paden–Kahan subproblems based on the product of exponential model. However, the simple combination of subproblems cannot solve all the inverse kinematics problems, and there is no common approach to solve arbitrary three-joint subproblems in an arbitrary postural relationship. The novel algebraic geometric (NAG) methods that obtain the general closed-form inverse kinematics for all types of three-joint subproblems are presented in this paper. The geometric and algebraic constraints are used as the conditions precedent to solve the inverse kinematics of three-joint subproblems. The NAG methods can be applied in the inverse kinematics of three-joint subproblems in an arbitrary postural relationship. The inverse kinematics simulations of all three-joint subproblems are implemented, and simulation results indicating that the inverse solutions are consistent with the given joint angles validate the general closed-form inverse kinematics. Huaque III minimally invasive surgical robot is used as the experimental platform for the simulation, and a master–slave tracking experiment is conducted to verify the NAG methods. The simulation result shows the inverse solutions and six sets given joint angles are consistent. Additionally, the mean and maximum of the master–slave tracking experiment for the closed-form solution are 0.1486 and 0.4777 mm, respectively, while the mean and maximum of the master–slave tracking experiment for the compensation method are 0.3188 and 0.6394 mm, respectively. The experiments results demonstrate that the closed-form solution is superior to the compensation method. The results verify the proposed general closed-form inverse kinematics based on the NAG methods.
