Objective:To compare the efficacy of three different tissue stains,namely haematoxylin and eosin(H&E),periodic-acid Schiff(PAS)and immunohistochemical(1HC)stains for detection of Entamoeba histolytica(***)trophozoites in abscessed liver tissues of ***:Amoebic liver abscess was experimentally induced in a hamster by injecting 1×10~6of axenically cultured virulent *** trophozoites(HM1-IMSS strain)into the portal *** a week post-inoculation,the hamster was sacrificed and the liver tissue sections were stained with H&E,PAS and IHC stains to delect the amoebic ***:The three stains revealed tissue necrosis and amoebic trophozoites,but with varying clarity.H&E and PAS stained the trophozoites pink and magenta,respectively,however it was difficult to differentiate the stained trophozoites from the macrophages because of their similarity in size and *** the other hand,IHC stain revealed distinct brown appearance of the trophozoites in the infected liver ***:It can be concluded that out of the three *** is the best for identification of *** trophozoites in tissue sections.

The weak interface interaction and solid-solid phase transition have long been a conundrum for 1,3,5,7-tetranitro-1,3,5,7-tetraazacyclooctane(HMX)-based polymer-bonded explosives(PBX).A two-step strategy that involves the pretreatment of HMX to endow—OH groups on the surface via polyalcohol bonding agent modification and in situ coating with nitrate ester-containing polymer,was proposed to address the *** types of energetic polyether—glycidyl azide polymer(GAP)and nitrate modified GAP(GNP)were grafted onto HMX crystal based on isocyanate addition reaction bridged through neutral polymeric bonding agent(NPBA)*** morphology and structure of the HMX-based composites were characterized in detail and the core-shell structure was *** grafted polymers obviously enhanced the adhesion force between HMX crystals and fluoropolymer(F2314)*** to the interfacial reinforcement among the components,the two HMX-based composites exhibited a remarkable increment of phase transition peak temperature by 10.2°C and 19.6°C with no more than 1.5%shell content,***,the impact and friction sensitivity of the composites decreased significantly as a result of the barrier produced by the grafted *** findings will enhance the future prospects for the interface design of energetic composites aiming to solve the weak interface and safety concerns.

This study investigated the application of electrical resistance tomography(ERT)in characterizing the slurry spatial distribution in cemented granular materials(CGMs).For CGM formed by self-flow grouting,the voids in the accumulation are only partially filled and the bond strength is often limited,which results in difficulty in obtaining in situ samples for quality ***,it is usually infeasible to evaluate the grouting effect or monitor the slurry spatial distribution by a mechanical *** this research,the process of grouting cement paste into high alumina ceramic beads(HACB)accumulation is reliably monitored with *** shows that ERT results can be used to calculate the cement paste volume in the HACB accumulation,based on calibrating the saturation exponent n in Archie’s *** results support the feasibility of ERT as an imaging tool in CGM characterization and may provide guidance for engineering applications in the future.

Zeolite imidazole skeleton(ziF-8)is a promising option for self-cleaning of building exterior walls due to its large specific surface area,high antibacterial activity and low ***,it suffers from low antibacterial efficiency and yield under visible light *** address the issues,we developed the photocatalytic materials T-ziF-8-TDI(thermally treated-ziF-8-toluene 2,4-diisocyanate)by modifying ziF-8 with thermal oxygen sensitization and chemical *** results show that the yield of T-ziF-8-TDI photocatalytic antibacterial agent is increased to 11.5 times of that of T-ziF-8,while maintaining the crystal structure of T-ziF-8 and thermal stability up to 60℃.Furthermore,T-ziF-8-TDI exhibits extended optical response range to the nearinfrared region,significantly narrowed band gap,improved photogenerated elec tron-hole separation efficiency,reduced recombination rate,and excellent photocatalytic *** the concentration of antibacterial agent is 600 mg·L^(-1),the antibacterial rate of Escherichia coli(***)reaches 99.99%irradiated by visible light for30 min,and when the concentration of antibacterial agent is 200 mg·L^(-1),the antibacterial rate of Staphylococcus aureus(***)reaches 99.99%irradiated by visible light for 25 *** also analyzed the reasons in detail from the aspects of bacterial species and antibacterial mechanism,and proposed the antibacterial mechanism of·O_(2)^(-)and h^(+)as the main active *** findings suggest that T-ziF-8-TDI photocatalytic antibacterial agent has potential for use in self-cleaning of building exterior walls.

Objective Lindqvist-type polyoxometalates(POMs)exhibit potential antitumor *** study aimed to examine the effects of Lindqvist-type POMs against breast cancer and the underlying *** Using different cancer cell lines,the present study evaluated the antitumor activities of POM analogues that were modified at the body skeleton based on molybdenum-vanadium-centered negative oxygen ion polycondensations with different side *** colony formation assay,autophagy detection,mitochondrial observation,qRT-PCR,Western blotting,and animal model were used to evaluate the antitumor activities of POMs against breast cancer cells and the related *** MO-4,a Lindqvist-type POM linking a proline at its side strain,was selected for subsequent experiments due to its low half maximal inhibitory concentration in the inhibition of proliferation of breast cancer *** was found that MO-4 induced the apoptosis of multiple types of breast cancer ***,MO-4 activated intracellular mitophagy by elevating mitochondrial reactive oxygen species(ROS)levels and resulting in *** vivo,breast tumor growth and distant metastasis were significantly reduced following MO-4 *** Collectively,the results of the present study demonstrated that the novel Lindqvist-type POM MO-4 may exhibit potential in the treatment of breast cancer.

N6-methyladenosine(m^(6)A)plays an important role in embryogenesis,nuclear export,transcription splicing,and protein translation ***,we demonstrate a copper-free click chemistry-mediated assembly of single quantum dot(QD)nanosensor for accurately monitoring locus-specific m^(6)A in cancer *** m^(6)A-sensitive endoribonuclease MazF can digest the unmethylated A-RNA,and the intact m^(6)A-RNA then hybridizes with DNA probes a and b to produce a sandwich hybrid,initiating the click chemistry to generate probe a–b ligation product via first tandem ligation detection reaction(LDR)***,DNA probes c and d can hybridize with the probe a–b ligation product to generate the probe c–d ligation product via second LDR *** LDR cycles can be repeated through denaturation and annealing reaction to generate abundant biotin-/fluorophore-modified probe c–d ligation products that can easily assemble on the QD surface to induce distinct fluorescence resonance energy transfer(FRET)between QD and *** assay can be homogenously performed without the involvement of copper catalyst,m^(6)A-specific antibody,radioactive labeling,ligase enzyme,enzymatic reverse transcription,and next-generation ***,it can discriminate even 0.01% m^(6)A level in complex samples and accurately measure cellular m^(6)A-RNA expression,providing a promising avenue for clinical diagnostics and biomedical research.

Objective:To characterize the Entamoeba histolytica(***)antigen(s)recognized by moribound amoebic liver abscess ***:Crude soluble antigen of *** was probed with sera of moribund hamsters in 1D-and 2D-Westem blot *** antigenic protein was then sent for tandem mass spectrometry *** corresponding gene was cloned and expressed in Escherichia coli BL21-AI to produce the recombinant *** ADP-forming acetyl-CoA synthetase(EhACS)protein.A customised ELISA was developed to evaluate the sensitivity and specificity of the recombinant ***:A^75 kDa protein band with a pl value of 5.91-6.5 was found to be antigenic;and not detected by sera of hamsters in the control *** mass spectrometry analysis revealed the protein to be the 77 kDa *** ADP-forming acetyl-CoA synthetase(EhACS).The customised ELISA results revealed 100%sensitivity and 100%specificity when tested against infected(n=31)and control group hamsters(n=5)serum samples,***:This rinding suggested the significant role of EhACS as a biomarker for moribund hamsters with acute amoebic liver abscess(ALA)*** is deemed pertinent that future studies explore the potential roles of EhACS in better understanding the pathogenesis of ALA;and in the development of vaccine and diagnostic tests to control ALA in human populations.

Despite transosseous rotator cuff tear repair using sutures is widely accepted for tendon-bone fixation,the fibrocartilaginous enthesis regeneration is still hardly achieved with the traditional *** the present work,degradable magnesium(Mg)alloy wire was applied to suture supraspinatus tendon in a rat acute rotator cuff tear model with Vicryl Plus 4±0 absorbable suture as *** shoulder joint humerus-supraspinatus tendon complex specimens were retrieved at 4,8,and 12 weeks after *** Mg alloy suture groups showed better biomechanical properties in terms of ultimate load to *** observation showed that hyperplastic response of the scar tissue at the tendon-bone interface is progressively alleviated over time in the both Mg alloy suture and Vicryl suture *** the histological analysis,for Mg alloy suture groups,chondrocytes appear to proliferate at 4 weeks postoperatively,and the tendon-bone interface showed an orderly structural transition zone at 8 weeks *** collagenous fiber tended to be aligned and the tendon-bone interlocking structures apparently formed,where transitional structure from unmineralized fibrocartilage to mineralized fibrocartilage was closer to the native fibrocartilaginous *** vivo degradation of the magnesium alloy wire was completed within 12 *** results indicated that Mg alloy wire was promising as degradable suture with the potential to promotes fibrocartilaginous interface regeneration in rotator cuff repair.

Background Excessive backfat deposition lowering carcass grade is a major concern in the pig industry,especially in most breeds of obese type *** mechanisms involved in adipogenesis and fat accumulation in pigs remain *** 2-hydroxyisobutyrylation(Khib),is a novel protein post-translational modification(PTM),which play an important role in transcription,energy metabolism and metastasis of cancer cells,but its role in adipogenesis and fat accumulation has not been *** In this study,we first analyzed the modification levels of acetylation(Kac),Khib,crotonylation(Kcr)and succinylation(Ksu)of fibro-adipogenic progenitors(FAPs),myogenic precursors(Myo)and mesenchymal stem cells(MSCs)with varied differentiation potential,and found that only Khib modification in FAPs was significantly higher than that in ***,in parallel with its regulatory enzymes lysine acetyltransferase 5(KAT5)and histone deacetylase 2(HDAC2)protein levels,the Khib levels increased quadratically(P<0.01)during adipogenic differentiation of ***5 knockdown in FAPs inhibited adipogenic differentiation,while HDAC2 knockdown enhanced adipogenic *** also demonstrated that Khib modification favored to adipogenic differentiation and fat accumulation by comparing Khib levels in FAPs and backfat tissues both derived from obese-type pigs(Laiwu pigs)and lean-type pigs(Duroc pigs),***,the expression patterns of KAT5 and HDAC2 matched well to the degree of backfat accumulation in obese-and lean-type *** From the perspective of protein translational modification,we are the first to reveal the role of Khib in adipogenesis and fat deposition in pigs,and provided new clues for the improvement of fat accumulation and distribution as expected via genetic selection and nutritional strategy in obese-type pigs.
