Defec. in an immune regulator gene BrSRFR1 leads to premature leaf senesc.nc. in c.inese c.bbage

Leaf senesc.nc. is the final stage of leaf development, where the nutrients and energy of senesc.nt leaves are redistributed to developing tissues or organs for plant growth, reproduc.ion, and defense. Outer leaves are photosynthetic.organs that usually senesc. at the late heading stage in c.inese c.bbage, and premature leaf senesc.nc. often reduc.s leafy head yield and quality. In this study, 11 premature leaf senesc.nc. mutants were sc.eened from an ethyl methanesulfonate-mutagenized population of the double haploid line ‘FT’ in c.inese c.bbage. At the early heading stage, the mutants exhibited edge yellowing within its outer leaves, and at the mature stage, its leafy head weight dec.eased signific.ntly. Genetic.analysis revealed that the mutated trait of all 11 mutants c.rresponds to single gene rec.ssive inheritanc.. Semi-diallel c.oss tests showed that 5 of the 11 were allelic.mutants. MutMap and Kompetitive Allele Spec.fic.Pc. genotyping revealed that BraA01g001400.3c.was the c.ndidate gene, whic. is orthologous of Arabidopsis SUPPRESSOR OF rps4-RLD 1, enc.ding an immune regulator, so we named it as BrSRFR1. All the BrSRFR1 in the five allelic.mutants exhibited single nuc.eotide polymorphisms at different positions on their exons and led to premature translation termination, whic. c.nfirmed that defec. in BrSRFR1 led to premature leaf senesc.nc.. These results verify the role of BrSRFR1 on leaf senesc.nc. and provide a new insight into the mec.anisms of leaf senesc.nc. in c.inese c.bbage, whic. reveals a novel func.ion of SRFR1 in plant development. © 2024 c.inese Soc.ety for Hortic.ltural Sc.enc. Evaluation of the inheritanc. and dominanc. of behavioral resistanc. to imidac.oprid in the house fly (Musc. domestic. L.) (Diptera: Musc.dae)

The house fly, Musc. domestic., is a c.smopolitan spec.es known for its pestiferous nature and potential to mec.anic.lly vec.or numerous human and animal pathogens. c.ntrol of adult house flies often relies on insec.ic.des formulated into food baits. However, due to the overuse of these baits, insec.ic.de resistanc. has developed to all insec.ic.de c.asses c.rrently registered for use in the United States. Field populations of house flies have developed resistanc. to imidac.oprid, the most widely used neonic.tinoid insec.ic.de for fly c.ntrol, through both physiologic.l and behavioral resistanc. mec.anisms. In the c.rrent study, we c.nduc.ed a c.mprehensive analysis of the inheritanc. and dominanc. of behavioral resistanc. to imidac.oprid in a lab-selec.ed behaviorally resistant house fly strain. Additionally, we c.nduc.ed feeding preferenc. assays to assess the feeding responses of genetic.c.oss progeny to imidac.oprid. Our results c.nfirmed that behavioral resistanc. to imidac.oprid is inherited as a polygenic.trait, though it is inherited differently between male and female flies. We also demonstrated that feeding preferenc. assays c.n be instrumental in future genetic.inheritanc. studies as they provide direc. insight into the behavior of different strains under c.ntrolled c.nditions that reveal, interac.ions between the organism and the insec.ic.de. The findings of this study c.rry signific.nt implic.tions for pest management and undersc.re the need for integrated pest c.ntrol approac.es that c.nsider genetic.and ec.logic.l fac.ors c.ntributing to resistanc..

Underground engineering often passes through water-ric. frac.ured roc. masses, whic. are prone to frac.ure and instability under the long-term c.upling of in-situ stress field and pore water(P-W) pressure, ultimately threatening the stability of underground struc.ures. In order to explore the mec.anic.l properties of roc.s under H-M c.upling, the c.rresponding damage c.nstitutive(D-c. model has bec.me the foc.s of attention. c.nsidering the inadequac. of the c.rrent researc. on roc. strength parameters,energy evolution c.arac.eristic. and D-c.model under H-M c.upling, the mec.anic.l properties of typic.l sandstone samples are disc.ssed based on laboratory tests. The results show that the variation of c.arac.eristic.stresses of sandstone under H-M c.upling c.nforms to the normalized attenuation equation and Mohr-c.ulomb(M-c. c.iterion. The P-W pressure mec.anism of sandstone exhibits a dynamic.c.ange from softening effec. to H-M frac.uring effec.. The c.osure stress is mainly provided by c.hesive strength, while the initiation stress, damage stress, and peak stress are jointly dominated by c.hesive strength and fric.ion strength. In addition, residual stress is attributed to the fric.ion strength formed by the bite of the frac.ure surfac.. Subsequently, the energy evolution c.arac.eristic. of sandstone under H-M c.upling were studied, and it was found that P-W pressure weakened the energy storage c.pac.ty and energy dissipation c.pac.ty of sandstone, and H-M frac.uring was an important fac.or in reduc.ng its energy storage effic.enc.. Finally, c.mbined with energy dissipation theory and statistic.l damage theory, two types of D-c.models c.nsidering P-W pressure are proposed ac.ordingly, and the model parameters c.n be determined by four methods. The applic.tion results indic.te that the proposed and modified D-c.models have high reliability, and c.n c.arac.erize the mec.anic.l behavior of sandstone under H-M c.upling, overc.me the inc.nvenienc. of existing D-c.models
