Effects of Ethephon and Abscisic Acid Application on Ripening-Related Genes in ‘Kohi' Kiwifruit (Actinidia chinensis) on the Vine

The effects of ethephon and abscisic acid(ABA) application on ripening-related genes of pre-harvest ‘Kohi' kiwifruit(Actinidia chinensis) were studied to clarify the possibility of the fruit ripening on the vine. The fruits were treated on the vine at mature stage with 250 μL·L^(-1)ethephon or 100 μmol·L^(-1)ABA, and the expression levels of chlorophyll synthase(Ac CLS), chlorophyllase1(Ac CLH1), polygalacturonase(Ac PG), expansin(Ac EXP),β-amylase(Acβ-AM), sucrose synthase(Ac SUSY), MADS-box SEPALLATA4/RIPENING INHIBITOR-like(Ac SEP4/RIN) and FRUITFUL-like(Ac TDR4/FUL) genes were analyzed. The expression levels of Ac PG, Ac EXP, Acβ-AM, and Ac SUSY increased in the ethephon-treated fruit, but those of Ac CLH1 at 6and 9 days after treatment and Ac CLS decreased. Moreover, the expression levels of Ac SEP4/RIN and Ac TDR4/FUL, the latter of which is associated with ethylene biosynthesis, were higher in the ethephon-treated fruit. The expression level of each gene in ABA-treated fruit was not significantly different from that of the untreated control. The results suggest that ethephon application increases the expression levels of Ac PG,Ac EXP, Acβ-AM, Ac SUSY, Ac SEP4/RIN, and Ac TDR4/FUL in ‘Kohi' kiwifruit on the vine.

AIM： To evaluate the role of positron emission tomography using 18F-fluorodeoxyglucose （FDG-PET） in the surgical management of patients with pancreatic cancer, including the diagnosis, staging, and selection of patients for the subsequent surgical treatment. METHODS： This study involved 53 patients with proven primary pancreatic cancer. The sensitivity of diagnosing the primary cancer was examined for FDG-PET CT, cytological examination of the bile or pancreatic juice, and the serum levels of carcinoembrionic antigens （CEA） and carbohydrate antigen 19-9 （CA29-9）. Next, the accuracy of staging was compared between FDG-PET and CT. Finally, FDG-PET was analyzed semiquantitatively using the standard uptake value （SUV）. The impact of the SUV on patient management was evaluated by examining the correlations between the SUV and the histological findings of cancer. RESULTS： The sensitivity of FDG-PET, CT, cytological examination of the bile or pancreatic juice, and the serum levels of CEA and CA19-9 were 92.5%, 88.7%, 46.4%, 37.7% and 69.8%, respectively. In staging, FDG-PET was superior to CT only in diagnosing distant disease （bone metastasis）. For local staging, the sensitivity of CT was better than that of FDG-PEr. The SUV did not correlate with the pTNM stage, grades, invasions to the vessels and nerve, or with the size of the tumor. However, there was a statistically significant difference （4.6 ± 2.9 vs 7.8 ± 4.5, P = 0.024） in the SUV between patients with respectable and unresectable disease. CONCLUSION： FDG-PET is thus considered to be useful in the diagnosis of pancreatic cancer. However, regarding the staging of the disease, FDG-PET is not considered to be a sufficiently accurate diagnostic modality. Although the SUV does not correlate with the patho-histological prognostic factors, it may be useful in selecting patients who should undergo subsequent surgical treatment.

It is highly important in Japan to choose a good site for wind turbines, because the spatial distribution of wind speed is quite complicated over steep complex terrain. We have been developing the unsteady numerical model called the RIAM-COMPACT (Research Institute for Applied Mechanics, Kyushu University, Computational Prediction of Airflow over Complex Terrain). The RIAM-COMPACT is based on the LES (Large-Eddy Simulation). The object domain of the RIAM-COMPACT is from several m to several km, and can predict the airflow and gas diffusion over complex terrain with high precision. In the present paper, the design wind speed evaluation technique in wind turbine installation point by using the mesoscale meteorological model and RIAM-COMPACT CFD model was proposed. The design wind speed to be used for designing WTGs can be calculated by multiplying the ratio of the mean wind speed at the hub-height to the mean upper-air wind speed at the inflow boundary, i.e., the fractional increase of the mean hub-height wind speed, by the reduction ratio, R. The fractional increase of the mean hub-height wind speed was evaluated using the CFD simulation results. This method was proposed as Approach 1 in the present paper. A value of 61.9 m/s was obtained for the final design wind speed, Uh, in Approach 1. In the evaluation procedure of the design wind speed in Approach 2, neither the above-mentioned reduction rate, R, nor an upper-air wind speed of 1.7 Vo, where Vo is the reference wind speed, was used. Instead, the value of the maximum wind speed which was obtained from the typhoon simulation for each of the investigated wind directions was adopted. When the design wind speed was evaluated using the 50-year recurrence value, the design wind speed was 48.3 m/s. When a somewhat conservative safety factor was applied, that is, when the 100 year recurrence value was used instead, the design wind speed was 52.9 m/s.

Case of colonic intussusception secondary to mobile cecum syndrome repaired by laparoscopic cecopexy using a barbed wound suture device

A 27-year-old man with recurrent right lower quadrant pain was admitted to our hospital. Ultrasonography and computed tomography examination of the abdomen revealed a target sign in the ascending colon, which was compatible with the diagnosis of cecal intussusception. The intussusception was spontaneously resolved at that time, but it relapsed 6 mo later. The patient underwent a successful colonoscopic disinvagination; there was no evidence of neoplastic or inflammatory lesions in the colon and terminal ileum. The patient underwent laparoscopic surgery for recurring cecal intussusception. During laparoscopy, we observed an unfixed cecum on the posterior peritoneum(i.e. a mobile cecum). Thus, we performed laparoscopic appendectomy and cecopexy with a lateral peritoneal flap using a barbed wound suture device. The patient's post-operative course was uneventful, and he continued to do well without recurrence at 10 mo after surgery. Laparoscopic cecopexy using a barbed wound suture device is a simple and reliable procedure that can be the treatment of choice for recurrent cecal intussusception associated with a mobile cecum.

Computational Investigation of the Causes of Wind Turbine Blade Damage at Japan’s Wind Farm in Complex Terrain

During the passage of Typhoon 0918 (Melor) over southern Honshu in Japan on 7 and 8 October 2009, strong winds with extremely high turbulence fluctuations were observed over Shirataki Mountain and the surrounding mountains in Shimonoseki, Yamaguchi Prefecture, Japan. These strong winds caused damage to wind turbine blades at the Shiratakiyama Wind Farm owned by Kinden Corporation. In order to investigate the causes of the blade damage, the airflow characteristics from the time of the incidences are first simulated in detail with the combined use of the WRF-ARW mesoscale meteorological model and the RIAM-COMPACT LES turbulence model (CFD model). Subsequently, in order to evaluate the wind pressure acting on the wind turbine blades, an airflow analysis is separately performed for the vicinity of the blades with the RANS turbulence model. Finally, the stress on the blades is investigated using the FEM with the RANS analysis results as the boundary conditions.
