Broccoli sprout extract induces detoxification-related gene expression and attenuates acute liver injury

AIM: To investigate the effects of broccoli sprout extract(BSEx) on liver gene expression and acute liver injury in the ***: First, the effects of BSEx on liver gene expression were examined. Male rats were divided into two groups. The Control group was fed the AIN-76 diet, and the BSEx group was fed the AIN-76 diet containing BSEx. After a 10-d feeding period, rats were sacrificed and their livers were used for DNA microarray and realtime reverse transcription-polymerase chain reaction(RT-PCR) analyses. Next, the effects of BSEx on acute liver injury were examined. In experiments using acute liver injury models, 1000 mg/kg acetaminophen(APAP) or 350 mg/kg D-galactosamine(D-Gal N) was used to induce injury. These male rats were divided into four groups: Control, BSEx, Inducer(APAP or D-Gal N), and Inducer+BSEx. The feeding regimens were identical for the two analyses. Twenty-four hours following APAP administration via p.o. or D-Gal N administration via i.p., rats were sacrificed to determine serum aspartate transaminase(AST) and alanine transaminase(ALT) levels, hepatic glutathione(GSH) and thiobarbituric acid-reactive substances accumulation and glutathioneS-transferase(GST) activity. RESULTS: Microarray and real-time RT-PCR analyses revealed that BSEx upregulated the expression of genes related to detoxification and glutathione synthesis in normal rat liver. The levels of AST(70.91 ± 15.74 IU/m L vs 5614.41 ± 1997.83 IU/m L, P < 0.05) and ALT(11.78 ± 2.08 IU/m L vs 1297.71 ± 447.33 IU/m L, P < 0.05) were significantly suppressed in the APAP + BSEx group compared with the APAP group. The level of GSH(2.61 ± 0.75 nmol/g tissue vs 1.66 ± 0.59 nmol/g tissue, P < 0.05) and liver GST activity(93.19 ± 16.55 U/g tissue vs 51.90 ± 16.85 U/g tissue, P < 0.05) were significantly increased in the APAP + BSEx group compared with the APAP group. Daily Intake of a Commercially Available Vegetable Juice Has Beneficial Effects on Metabolic Syndrome-Related Biomarkers in Young, Healthy Japanese Adults

Adequate vegetable intake is important in reducing the risk of lifestyle-related diseases;however, intake amongst young Japanese adults is poor. This study aimed to assess the effect of daily intake of a commercially available vegetable juice on nutritional status and metabolic syndrome-related markers in healthy young adults. Fifty-one healthy adults (14 men, 37 women), age 20 - 22 years were recruited, and their food and nutritional intake was surveyed using an FFQ based on Food Groups. Subjects were divided into either a control group, who were not administered vegetable juice, or an intervention group, who consumed 200 mL vegetable juice daily for 2 months. Anthropometric data, serum carotenoid concentrations, blood and urinary parameters were measured at baseline and at 2 months. The average intake of vegetables in all subjects at baseline was approximately 140 g/day, less than half the dose recommended by the Japanese government. Total serum carotenoid concentration in the intervention group doubled from baseline after 2 months. In particular, serum concentrations of α-carotene, β-carotene, and lycopene were increased. In the intervention group, 24-hour urinary potassium excretion increased significantly, and 24-hour urinary sodium to potassium ratio decreased significantly. Waist circumference and systolic blood pressure also decreased significantly. Daily intake of a commercial vegetable juice for 2 months improved nutritional status in young adults with inadequate vegetable intake, shown by improved serum carotenoid and urine potassium levels. In addition, intake of vegetable juice may be beneficial in reducing the risk of metabolic syndrome.

POLYMER COLLOIDS FORMED BY POLYELECTROLYTE COMPLEXATION OF VINYL POLYMERS AND POLYSACCHARIDES IN AQUEOUS SOLUTION

The polyelectrolyte complex formed from the polyanion and polycation was studied by turbidimetry, static and electrophoretic light scattering, and elementary analysis. Sodium salts of polyacrylate （PA） and heparin （Hep） were chosen as the polyanion, and hydrochloric salts of poly（vinyl amine）（PVA） and chitosan （Chts） as the polycation. Although these vinyl polymers and polysaccharides have remarkably different backbone chemical structures and linear charge densities, all the four combinations PA-PVA, PA-Chts, Hep-PVA, and Hep-Chts provide almost stoichiometric polyelectrolyte complexes which are slightly charged owing to the adsorption of the excess polyelectrolyte component onto the neutral complex. The charges stabilize the complex colloids in aqueous solution of a non-stoichiometric mixture, and the aggregation number of the complex colloids increases with approaching to the stoichiometric mixing ratio. The mixing ratio dependence of the aggregation number for the four complexes is explained by the model proposed in the previous study.

Study on dynamic response of embedded long span corrugated steel culverts using scaled model shaking table tests and numerical analyses

A series of scaled-model shaking table tests and its simulation analyses using dynamic finite element method were performed to clarify the dynamic behaviors and the seismic stability of embedded corrugated steel culverts due to strong earth-quakes like the 1995 Hyogoken-nanbu earthquake. The dynamic strains of the embedded culvert models and the seismic soil pressure acting on the models due to sinusoidal and random strong motions were investigated. This study verified that the cor-rugated culvert model was subjected to dynamic horizontal forces (lateral seismic soil pressure) from the surrounding ground, which caused the large bending strains on the structure; and that the structures do not exceed the allowable plastic deformation and do not collapse completely during strong earthquake like Hyogoken-nanbu earthquake. The results obtained are useful for design and construction of embedded long span corrugated steel culverts in seismic regions.

Activity coefficients of NiO and CoO in CaO–Al_2O_3–SiO_2 slag and their application to the recycling of Ni–Co–Fe-based end-of-life superalloys via remelting

To design optimal pyrometallurgical processes for nickel and cobalt recycling, and more particularly for the end-of-life process of Ni-Co-Fe-based end-of-life（EoL） superalloys, knowledge of their activity coefficients in slags is essential. In this study, the activity coefficients of NiO and CoO in CaO-Al_2O_3-SiO_2 slag, a candidate slag used for the EoL superalloy remelting process, were measured using gas/slag/metal equilibrium experiments. These activity coefficients were then used to consider the recycling efficiency of nickel and cobalt by remelting EoL superalloys using CaO-Al_2O_3-SiO_2 slag. The activity coefficients of NiO and CoO in CaO-Al_2O_3-SiO_2 slag both show a positive deviation from Raoult＇s law, with values that vary from 1 to 5 depending on the change in basicity. The activity coefficients of NiO and CoO peak in the slag with a composition near B =（%CaO）/（%SiO_2） = 1, where B is the basicity. We observed that controlling the slag composition at approximately B = 1 effectively reduces the cobalt and nickel oxidation losses and promotes the oxidation removal of iron during the remelting process of EoL superalloys.

Electron scattering by Friedel oscillations in carbon nanotubes

Multi-walled carbon nanotube networks were confirmed to exhibit a linear decrease in resistivity with increasing temperature from 100 to above 400 *** linearity was explained using a defect scattering model that involved Friedel oscillations(that is,electron-electron interactions).The applicability of this model,which was originally proposed for graphene,to carbon nanotubes was assessed based on a comparison of various experimental *** in the slopes of the resistivity-temperature plots following the introduction of defects,as well as an effect of charge concentration on the slope were key predictions of this *** results obtained from few-walled carbon nanotube networks are also *** the literature,linear resistivity-temperature plots were obtained from other graphene derivatives,indicating that the linearity originates from the hexagonal symmetry of these *** present work also indicated a relationship between the appearance of linearity and negative magnetoresistance above 100 *** on a mechanism incorporating scattering in association with Friedel oscillations and conventional electron conduction models,the universality of resistivity-temperature plots obtained from carbon nanotube networks is introduced.
