Phytohemagglutinin(PHA)is a seed storage protein and a type of lectin originally discovered in the common bean(Phaseolus vulgaris)for its blood-agglutinating *** to its interactions with gut epithelia and digestive enzymes and its potential to trigger allergic reactions,PHA can lead to various symptoms in the human *** a result,it has been regarded as a signifcant antinutritional factor in beans and other *** several published works have summarized its structural,biochemical,and toxicological features,there is a scarcity of literature that reviews the detection,quantifcation,and reduction of PHA in beans,which is fundamental for the development of safer bean *** this review,we present a comprehensive analysis of traditional and innovative bio-sensing methods for measuring PHA,including the recently available ultrapure liquid chromatography–tandem mass spectrometry and emerging aptamer sensor-based techniques,while discussing their respective advantages and *** also revisit existing studies dedicated to creating PHA-depleted common bean varieties and explore the potential for reducing PHA content in beans without compromising their resistance to biotic ***,we offer insights into the potential for controlling PHA content using the latest biotechnologies and breeding ***,this review compiles rare and valuable information from studies that solely focuses on detection and depletion of PHA to shed light on and apply technological advancements in addressing potential food safety risks associated with the consumption of common beans.

Plant pathogens can alter the behavior of their insect vectors as well as their survival and *** African psyllid,Trioza erytreae,is one of the vectors of Huanglongbing,a citrus disease caused mainly by"Candidatus Liberibacter asiaticus"(CLas).The purpose of this study was to characterize the effects of CLas on the psyllid,*** using Citrus volkamerina plants as the study *** study focused more specifically on the CLas effects prior to and after its acquisition by the psyllid *** results did not support the hypothesis that CLas effects psyllid probing behavior prior to acquisition;few differences were observed between uninfected *** feeding on CLas-infected versus control *** the other hand,compared to psyllids that had completed their development on control plants,the ones that had completed their development on a CLas-infected plant exhibited changes in their behavior(greater velocity),physiology(smaller mass)and biochemistry(lower water and lipid content).Altogether,our results confirm the existence of a marked postacquisition effect on the vector locomotor behavior and a minor preacquisition effect of CLas on the vector behavior,which can be partially explained by physiological and biochemical changes.

Anoplophora glabripennis(Asian longhorn beetle,ALB)and Anoplophora chinensis(Citrus longhorn beetle,CLB)are native forest pests in China;they have become important international quarantine *** are found using the same Salix aureopendula host tree of Cixi,Zhejiang province,*** this host tree,we collected additional beetles that appeared to be morphologically intermediate between ALB and *** using a stereoscope,we observed that there were several bumps on the base of the elytra,which was inconsistent with ALB,which typically has a smooth elytral base,but was more like CLB,which has numerous short tubercles on the elytral *** their sympatry and intermediate morphology,we hypothesized that these may represent ALB×CLB *** studied the genomic profiles for 46 samples(ALB,CLB,and putative hybrids)using genotyping-by-sequencing(GBS)providing a reduced representation of the entire *** principal component analyses on the 163 GBS-derived single nucleotide polymorphism data,we found putative hybrids tightly clustered with ALB,but genetically distinct from the CLB ***,our initial hybrid hypothesis was not supported by genomic ***,while mating experiments between adult ALB and CLB were successful in 4 separate years(2017,2018,2020,and 2021),and oviposition behavior was observed,no progeny was *** employed population genomic analysis and biological hybridization experiments,we conclude that the putative hybrids represent newly discovered morphological variants within *** approach further confirmed the advantage of genome-wide information for Anoplophora species assignment in certain ambiguous classification cases.

Numerical Optimization by Finite Element Method of Stainless Steel/Glass-Epoxy Composite Bolted Joint under Tension and Compression

The aim of this study was to optimize the geometry and the design of metallic/composite single bolted joints subjected to tension-compression loading. For this purpose, it was necessary to evaluate the stress state in each component of the bolted join. The multi-material assembly was based on the principle of double lap bolted joint. It was composed of a symmetrical balanced woven glass-epoxy composite material plate fastened to two stainless sheets using a stainless pre-stressed bolt. In order to optimize the design and the geometry of the assembly, ten configurations were proposed and studied: a classical simple bolted joint, two joints with an insert (a BigHeadR insert and a stair one) embedded in the composite, two “waved” solutions, three symmetrical configurations composed of a succession of metallic and composites layers, without a sleeve, with one and with two sleeves, and two non-symmetrical constituted of metallic and composites layers associated with a stair-insert (one with a sleeve and one without). A tridimensional Finite Element Method (FEM) was used to model each configuration mentioned above. The FE models taked into account the different materials, the effects of contact between the different sheets of the assembly and the pre-stress in the bolt. The stress state was analyzed in the composite part. The concept of stress concentration factor was used in order to evaluate the stress increase in the highly stressed regions and to compare the ten configurations studied. For this purpose, three stress concentration factors were defined: one for a monotonic loading in tension, another for a monotonic loading in compression, and the third for a tension-compression cyclic loading. The results of the FEM computations showed that the use of alternative metallic and composite layers associated with two sleeves gived low values of stress concentration factors, smaller than 1.4. Background Extracorporeal membrane oxygenation(ECMO)for acute respiratory distress syndrome(ARDS)is systematically associated with decreased respiratory system compliance(CRS).It remains unclear whether transportation to the referral ECMO center,changes in ventilatory mode or settings to achieve ultra-protective ventilation,or the natural evolution of ARDS drives this change in respiratory ***,we assessed the precise moment when CRS decreases after ECMO cannulation and identified factors associated with decreased *** To rule out the effect of transportation and the different modes of ventilation on CRS,we conducted a retrospective,single-center,observational cohort study from January 2013 to May 2020,on 22 patients with severe ARDS requiring on-site ECMO and ventilated in pressure-controlled mode to achieve ultra-protective *** was assessed at different time points ranging from 12 h before ECMO cannulation to 72 h after ECMO *** primary outcome was the relative change in CRS between 3 h before and 3 h after ECMO *** secondary outcomes included variables associated with the relative changes in CRS within the first 3 h after ECMO cannulation and the relative changes in CRS at each time *** CRS decreased within the first 3 h after ECMO cannulation(−28.3%,95%confidence interval[CI]:−38.8 to−17.9,P<0.001),while the decrease was mild before and after these first 3 h after ECMO *** achieve ultra-protective ventilation,respiratory rate decreased in the mean by–13 breaths/min(95%CI:−15 to−11)and driving pressure by−8.3 cmH2O(95%CI:−11.2 to−5.3),resulting in decreased tidal volume by−3.3 mL/kg of predicted body weight(95%CI:−3.9 to−2.6)as compared to before ECMO cannulation(P<0.001 for all).Plateau pressure reduction,driving pressure reduction,and tidal volume reduction were significantly associated with decreased CRS after ECMO cannulation,whereas neither respiratory rate,positive end-expiratory pr

Introduction: While autograft bone is the gold standard for multilevel posterolateral lumbar fusion, bone substitutes and graft extenders such as allograft bone, ceramics and demineralized bone matrix (DBM) have been used to avoid the morbidity and insufficient quantity associated with harvesting autologous bone. The primary objective of this retrospective study was to determine whether, in patients with increased risk of operative nonunion related to multilevel fusion, adding DBM fibers to mineralized bone allograft resulted in better fusion than using allograft alone. The secondary objectives were to evaluate how adding DBM fibers affects functional disability, low back pain, intraoperative blood loss and the nonunion rate. Methods: This retrospective study involved a chart review of consecutive patients who underwent multilevel lumbar spinal fusion and were operated on by a single surgeon. The patients were divided into two groups: 14 patients received mineralized bone allograft (control group) and 14 patients received a combination of mineralized bone allograft and DBM (experimental group). Patients were reviewed at a mean of 16.4 ± 2.2 months after surgery at which point CT scans were analyzed to determine whether fusion had occurred;Oswestry disability index (ODI) and pain were also evaluated. Results: A mean of 5 levels [min 2, max 13] were fused in these patients. Posterolateral fusion as defined by the Lenke classification was not significantly different between groups. The experimental DBM group had a significantly better composite fusion score than the control group (P Discussion: Adding DBM fibers to allograft bone during multilevel posterolateral spinal fusion was safe and produced better composite fusion than using allograft only as an autograft extender.
