Valorization of the Caragana waste via two-stage bioaugmentation: Optimizing nutrition composition, palatability, and microbial contaminant control

Caragana korshinskii kom. (CKK) waste, a common forestry byproduct in northwest of China, presents challenges in its transformation into alternative ruminant feed due to its initial nutritional limitations and unappealing palatability. Conventional strategies, such as ensiling and fungal- based solid-state fermentation (ssF) cannot effectively address this issue in practice. Herein, a two-stage bioaugmentation (TBA) process was devised, leveraging the benefits of ensiling and ssF. During the anaerobic ensiling phase, CKK waste was inoculated with Lactiplantibacillus plan- tarum LP1, effectively suppressing potential animal pathogens such as Aspergillus and Nocardiop- sis while enriching the material with potential probiotics like Pediococcus and Lactiplantibacillus , reaching an abundance of 95.7%. In the subsequent aerobic ssF stage, the ensiled CKK under- went inoculation with the white-rot fungus Irpex lacteus F17, which became enriched to 87.9%. Comprehensive multi-omics analysis identified Irpex as the key taxon, possessing an extensive redox enzyme system that led to the improvement in nutrient composition, reduction of astrin- gent phenolic substances, and mitigation of mycotoxins. As a result, the crude protein content of the CKK increased by 39.2%, while lignin, total phenolic substances, and tannic acid content de- creased by 24.4%, 52.2%, and 51.4%, respectively. The mycotoxin levels, including aflatoxin B1 , zearalenone, and vomitoxin, were rendered negligible, confirming the safety. Overall, this study demonstrates the TBA strategy can successfully transform challenging and unpalatable CKK waste into a nutrient-enriched and safe mycelium-based bioproduct, thereby enabling the valorization of a previously underutilized forestry resource as a promising alternative feed.

Effect of Preplant Fertilizer on Agronomic and Physiological Traits of soybean Cultivars from Different Breeding Programs

Understanding the changes in agronomic and physiological traits associated with yield genetic gain is important for soybean production and future breeding strategy. The objective of this study was to compare the older and modern cultivars to learn whether the yield improvements depend on preplant fertilizer or the plant productivity improvement, A set of older cultivars, with their modern counterparts derived from breeding programs in Liaoning and Ohio were evaluated for their agronomic and physiological traits under different fertilizer levels from 2004 to 2006. There was no improvement of response to N and P preplant fertilizer for genotypes. After more than 70 yr breeding, soybean breeders made some improvements in agronomic and physiological traits that contribute to yield increase. When compared to older cultivar, modern Liaoning and Ohio cultivars were shorter and more resistance to lodging, had greater leaf density, higher harvest index, more leaf area per plant, and greater photosynthetic rate, transpiration rate and stomatal conductance at the beginning of seed development. Ohio cultivars were more resistant to lodging as if selected for easy harvest by combine, even under high N and P preplant fertilizer level, which resulted in Ohio cultivars with higher and stable yield productivity.

sinocyclocheilus is a cave-dwelling cyprinid genus endemic to southwest *** species possess a conspicuous horn on their head,which has been suggested as a constructive troglomorphic trait but lacks substantial *** used non-invasive,high spatial resolution synchrotron X-ray microtomography to investigate the three-dimensional(3D)morphology of the horn of sinocyclocheilus hyalinus,one of eight such troglobiotic species.3D renderings demonstrated the osteological components,which were comprised of a rear wall comprised of the supraoccipital bone,a remaining frontal wall with numerous fenestrae,and the bottom continuous with the parietal and epiotic.A horn cavity occurred within the *** fenestrae in the frontal wall were continuous in the horn cavity and showed elaborate channeling,and were,connected to the cranial cavity by soft *** tentatively called this configuration the“otocornual connection”due to its anatomic and putative functional similarity to the otolateralic connection in clupeids and loricariids,which provide an indirect pathway to enhance perception of underwater sound *** study provides a functional morphology context for further histological and physiological investigations of such horn structures in sinocyclocheilus cavefish,and we suggest that the horn might enhance acoustic perception to compensate for visual loss in subterranean life,which warrants future physiological examination as lab-reared *** become available.

Dynamics of Nitrogen speciation in Horticultural soils in suburbs of shanghai,China

Dissolved organic nitrogen （DON） represents a significant pool of soluble nitrogen （N） in soil ecosystems. soil samples under three different horticultural management practices were collected from the Xiaxiyang Organic Vegetable and Fruit Farm, shanghai, China, to investigate the dynamics of N speciation during 2 months of aerobic incubation, to compare the effects of different soils on the mineralization of ^14C-labeled amino acids and peptides, and to determine which of the pathways in the decomposition and subsequent ammonification and nitrification of organic N represented a significant blockage in soil N supply. The dynamics of N speciation was found to be significantly affected by mineralization and immobilization. DON, total free amino acids, and NH^＋-N were maintained at very low levels and did not accumulate, whereas NO3^--N gradually accumulated in these soils. The conversion of insoluble organic N to low-molecular-weight （LMW） DON represented a main constraint to N supply, while conversions of LMW DON to NH4^＋-N and NH4^＋-N to NO3^--N did not. Free amino acids and peptides were rapidly mineralized in the soils by the microbial community and consequently did not accumulate in soil. Turnover rates of the additional amino acids and peptides were soil-dependent and generally followed the order of organic soil 〉 transitional soil 〉 conventional soil. The turnover of high-molecular-weight DON was very slow and represented the major DON loss. Further studies are needed to investigate the pathways and bottlenecks of organic N degradation.

Effects of Cu addition on microstructureand mechanical properties of as-cast Mg-6Znmagnesium alloy

The application of Mg-Zn binary alloys is restricted due to their developed dendritic microstructure and poor mechanical properties. In this study, an alloying method was used to improve the mechanical properties of Mg-Zn alloy. The Mg-6Zn magnesium alloys microalloyed with varying Cu content(0, 0.8, 1.5, 2.0 and 2.5wt.%) were fabricated by permanent mould casting, and the effects of Cu content on the microstructure and mechanical properties of as-cast Mg-6Zn alloys were studied using OM, sEM, XRD and tensile tests at room temperature. The obtained results show that the addition of Cu not only can refine the grains effectively, but also can modify the eutectic morphology and improve the mechanical properties of the alloys. The main phases of the studied alloys include α-Mg, MgZn_2, Mg_2Cu and CuMgZn. When the content of Cu exceeds 0.8wt.%, Mg_2Cu phase appears. Meanwhile, the eutectic morphology is modified into dendritic shape or lamellar structure, which has an adverse effect on the tensile properties. Furthermore, among the investigated alloys, the alloy containing 0.8% Cu shows an optimalultimate tensile strength of 196 MPa, while the alloy with 1.5wt.% Cu obtains an excellent elongation of 7.22%. The experimental alloys under different Cu contents show distinguishing fracture behaviors: the fracture of the alloy with 0.8wt.% Cu reveals a mixed mode of inter-granular and quasi-cleavage, while in other investigated alloys, the fracture behaviors are dominated by cleavage fracture.

Genome-Wide Transcriptional Analysis of Yield and Heterosis-Associated Genes in Maize (Zea mays L.)

Heterosis has contributed greatly to yield in maize, but the nature of its contribution is not completely clear. In this study, two strategies using whole-genome oligonucleotide microarrays were employed to identify differentially expressed genes （DEGs） associated with heterosis and yield. The analysis revealed 1 838 heterosis-associated genes （HAGs）, 265 yield- associated genes （YAGs）, and 85 yield heterosis-associated genes （YHAGs）. 37.1% of HAGs and 22.4% of YHAGs expressed additively. The remaining genes expressed non-additively, including those with high/low-parent dominance and over/under dominance, which were prevalent in this research. Pathway enrichment analysis and quantitative trait locus （QTL） co-mapping demonstrated that the metabolic pathways for energy and carbohydrates were the two main enriched pathways influencing heterosis and yield. Therefore, the DEGs participating in energy and carbohydrate metabolism were considered to contribute to heterosis and yield significantly. The investigation of potential groups of HAGs, YAGs, and YHAGs might provide valuable information for exploiting heterosis to improve yield in maize breeding. In addition, our results support the view that heterosis is contributed by multiple, complex molecular mechanisms.
