Fluoro-functionalized plant biomass adsorbent:Preparation and application in extraction of trace perfluorinated compounds from environmental water samples

Perfuorinated compounds(PFCs)are toxic and widely present in the environment,and therefore effective adsorbents are required to remove PFCs from environmental water.In the present study,a new type of fuorinated biomass materials was synthesized via an ingenious fuorosilanization reaction.These adsorbents were applied for the adsorption of 13typical PFCs,including perfuorocarboxylic acids(PFCAs)and perfuorosulfonic acids(PFSAs).By comparing their adsorption performance,Fluorinated cedar slag(FCS)was discovered to have the best absorption efficiency and enabled highly efficient enrichment of PFCs.The adsorption recovery of FCS with the investigated PFCs is greater than 90%under the optimal adsorption condition.Ascribed to the high affinity of F-F sorbent-sorbate interaction,FCS had good adsorption capacities of PFCs from aqueous solution,with the maximum adsorption capacity of 15.80 mg/g for PFOS and 10.71 mg/g for PFOA,respectively.Moreover,the adsorption time could be achieved in a short time(8 min).Using the FCS absorbent,an innovative FCS-solid phase extraction assisted with high performance liquid chromatography-electrospray-tandem mass spectrometry(FCS-SPE-HPLC-ESI-MS/MS)method was first developed to sensitively detect PFCs in the environmental water samples.The intra-day and inter-day recovery rates of the 13 compounds ranged from 90.7%-104.3%,with the RSD of 2.1%-4.7%(intra-day)and 2.5%-8.5%(inter-day),respectively.This research demonstrates the potential of the newly fuoro-functionalized plant biomass to adsorb PFCs from environmental water,with the advantages of high adsorption efficiencies,high antiinterference,easy operation and low economic cost.

Modelling smear effect of vertical drains using a diameter reduction method

Vertical drains are used to accelerate consolidation of clays in ground improvement projects.Smear zones exist around these drains,where permeability is reduced due to soil disturbance caused by the installation process.Hansbo solution is widely used in practice to consider the effects of drain discharge capacity and smear on the consolidation process.In this study,a computationally efficient diameter reduction method(DRM)obtained from the Hansbo solution is proposed to consider the smear effect without the need to model the smear zone physically.Validated by analytical and numerical results,a diameter reduction factor is analytically derived to reduce the diameter of the drain,while achieving similar solutions of pore pressure dissipation profile as the classical full model of the smear zone and drain.With the DRM,the excess pore pressure u obtained from the reduced drain in the original un-disturbed soil zone is accurate enough for practical applications in numerical models.Such performance of DRM is independent of soil material property.Results also show equally accurate performance of DRM under conditions of multi-layered soils and coupled radial-vertical groundwater flow.

Coniopteris simplex is a common component in many Jurassic floras.However,due to morphological variations in sterile fronds and incomplete preservation of reproductive organs,its classification has been controversial for a long time.Here,we present new material collected from the Middle Jurassic Yan’an Formation in the Ordos Basin,Inner Mongolia.The new material reveals important morphological characters,including hemi-dimorphic fronds,two forms of sterile pinnae,short-stalked sori and sporangia on fertile fronds,annulus of sporangium composed of about 30 cells,in situ rounded triangular spores with protruded suture,and the lips of trilete laesura encircled by one or two rows of small ostioles.Based on these observations,we emended the diagnosis of this common species.The spatio-temporal distribution of Coniopteris simplex shows that the iconic species was restricted to the paleolatitudes ranging from 19.5°N to 48.9°N in the Northern Hemisphere during the Early—Late Jurassic,mainly distributed in the tropical to paratropical zones,and the paleolongitude ranging from 9.6°E to 129.3°E,which may be limited by the opening of the Viking Corridor and the splitting of the Central Atlantic Ocean.

Auxin regulates flower and fruit abscission,but how developmental signals mediate auxin transport in abscission remains unclear.Here,we reveal the role of the transcription factor BEL1-LIKE HOMEODOMAIN11(SlBEL11)in regulating auxin transport during abscission in tomato(Solanum lycopersicum).SlBEL11 is highly expressed in the fruit abscission zone,and its expression increases during fruit development.Knockdown of SlBEL11 expression by RNA interference(RNAi)caused premature fruit drop at the breaker(Br)and 3d post-breaker(Br+3)stages of fruit development.Transcriptome and metabolome analysis of SlBEL11-RNAi lines revealed impaired flavonoid biosynthesis and decreased levels of most flavonoids,especially quercetin,which functions as an auxin transport inhibitor.This suggested that SlBEL11 prevents premature fruit abscission by modulating auxin efflux from fruits,which is crucial for the formation of an auxin response gradient.Indeed,quercetin treatment suppressed premature fruit drop in SlBEL11-RNAi plants.DNA affinity purification sequencing(DAP-seq)analysis indicated that SlBEL11 induced expression of the transcription factor gene SlMYB111 by directly binding to its promoter.Chromatin immunoprecipitation-quantitative polymerase chain reaction and electrophoretic mobility shift assay showed that S.lycopersicum MYELOBLASTOSIS VIRAL ONCOGENE HOMOLOG111(SlMYB111)induces the expression of the core flavonoid biosynthesis genes SlCHS1,SlCHI,SlF3H,and SlFLS by directly binding to their promoters.Our findings suggest that the SlBEL11-SlMYB111 module modulates flavonoid biosynthesis to fine-tune auxin efflux from fruits and thus maintain an auxin response gradient in the pedicel,thereby preventing premature fruit drop.

RNA interference(RNAi)is a powerful tool that post-transcriptionally silences target genes in eukaryotic cells.However,silencing efficacy varies greatly among different insect species.Recently,we met with little success when attempting to knock down genes in the mirid bug Apolygus lucorum via dsRNA injection.The disappearance of double-stranded RNA(dsRNA)could be a potential factor that restricts RNAi efficiency.Here,we found that dsRNA can be degraded in midgut fluids,and a dsRNase of A.lucorum(AldsRNase)was identified and characterized.Sequence alignment indicated that its 6 key amino acid residues and the Mg2+-binding site were similar to those of other insects’dsRNases.The signal peptide and endonuclease non-specific domain shared high sequence identity with the brown-winged green stinkbug Plautia stali dsRNase.AldsRNase showed high salivary gland and midgut expression and was continuously expressed through the whole life cycle,with peaks at the 4th instar ecdysis in the whole body.The purified AldsRNase protein obtained by heterologously expressed can rapidly degrade dsRNA.When comparing the substrate specificity of AldsRNase,3 specific substrates(dsRNA,small interfering RNA,and dsDNA)were all degraded,and the most efficient degradation is dsRNA.Subsequently,immunofluorescence revealed that AldsRNase was expressed in the cytoplasm of midgut cells.Through cloning and functional study of AldsRNase,the enzyme activity and substrate specificity of the recombinant protein,as well as the subcellular localization of nuclease,the reason for the disappearance of dsRNA was explained,which was useful in improving RNAi efficiency in A.lucorum and related species.

Chemotactic nanomotor for multimodal combined therapy of glioblastoma

Glioblastoma(GBM) is the most aggressive malignant brain tumor. Due to the infiltration and heterogeneity of GBM, the obstruction of the blood-brain barrier(BBB) and the unique immunosuppressive mechanism, it is hard to achieve significant effects of GBM treatment. Here, a kind of chemotactic nanomotor that loaded with glucose oxidase(GOx) and carboxylated cisplatin(Pt(IV)) prodrug on the L-arginine-derived polymer is proposed. The nanomotors are driven by catalysis of glucose decomposition and the positive chemotaxis towards the GBM microenvironment where inducible nitric oxide synthase and reactive oxygen species are highly expressed. This facilitates the BBB crossing and GBM targeting of the nanomotors. In addition, the released nitric oxide(NO) during propulsion as well as the loaded GOx and Pt(IV) can exert combined NO/starvation/chemotherapy. Meanwhile, it is able to induce and enhance the immune response through multiple pathways, thus better coping with the complexities of GBM treatment.

Assessment of Dust Aerosol Optical Depth and Shortwave Radiative Forcing over the Northwest Pacific Ocean in Spring Based on Satellite Observations

Dust aerosol optical depth (AOD) and its ac-companying shortwave radiative forcing (RF) are usually simulated by numerical models.Here,by using 9 months of Moderate Resolution Imaging Spectroradiometer (MODIS) aerosol product data in combination with Clouds and the Earth's Radiant Energy System Single Scanner Footprint (CERES/SSF) data,dust AOD and its shortwave RF were estimated over the cloud-free north-west (NW) Pacific Ocean in the springs of 2004,2005,and 2006.The results showed that in this region,the mean dust AOD and its shortwave RF were 0.10 and 5.51 W m 2,respectively.In order to validate the dust AOD de-rived by MODIS,results from the Goddard Global Ozone Chemistry Aerosol Radiation and Transport (GOCART) model were also used here.The correlation coefficient between the monthly averaged dust AOD derived by MODIS measurements and the model simulation results was approximately 0.53.Since the estimates of the dust AOD and its shortwave RF obtained in this study are based mainly on satellite data,they offer a good reference for numerical models.

Comprehensive analysis of the protein phosphatase 2A regulatory subunit B56ε in pan-cancer and its role and mechanism in hepatocellular carcinoma

BACKGROUND B56εis a regulatory subunit of the serine/threonine protein phosphatase 2A,which is abnormally expressed in tumors and regulates various tumor cell functions.At present,the application of B56εin pan-cancer lacks a comprehensive analysis,and its role and mechanism in hepatocellular carcinoma(HCC)are still unclear.The Cancer Genome Atlas,Genotype-Tissue Expression,Gene Expression Profiling Interactive Analysis,and Tumor Immune Estimation Resource databases were used to analyze B56εexpression,prognostic mutations,somatic copy number alterations,and tumor immune characteristics in 33 tumors.The relationships between B56εexpression levels and drug sensitivity,immuno-therapy,immune checkpoints,and human leukocyte antigen(HLA)-related genes were further analyzed.Gene Set Enrichment Analysis(GSEA)was performed to reveal the role of B56εin HCC.The Cell Counting Kit-8,plate cloning,wound healing,and transwell assays were conducted to assess the effects of B56εinterference on the malignant behavior of HCC cells.RESULTS In most tumors,B56εexpression was upregulated,and high B56εexpression was a risk factor for adrenocortical cancer,HCC,pancreatic adenocarcinoma,and pheochromocytoma and paraganglioma(all P<0.05).B56εexpression levels were correlated with a variety of immune cells,such as T helper 17 cells,B cells,and macro-phages.There was a positive correlation between B56εexpression levels with immune checkpoint genes and HLA-related genes(all P<0.05).The expression of B56εwas negatively correlated with the sensitivity of most chemotherapy drugs,but a small number showed a positive correlation(all P<0.05).GSEA analysis showed that B56εexpression was related to the cancer pathway,p53 downstream pathway,and interleukin-mediated signaling in HCC.Knockdown of B56εexpression in HCC cells inhibited the proliferation,migration,and invasion capacity of tumor cells.Core Tip:The expression of protein phosphatase 2A(PP2A)subunit B56εis up-regulated in most tumors,and its high expression is a risk factor for adrenocortical c

Glycine Attenuates Myocardial Fibrosis in Myocardial Infarction in Rats Partly through Modulating Signal Transducer and Activator of Transcription 3/Nuclear Factor-κB/Transforming Growth Factor-β axis

Objective: Inflammation and fibrosis are strongly associated with each other. Glycine is present in various traditional Chinese medicines and exhibits anti-inflammatory activity. However, the effects of glycine on myocardial fibrosis(MF) in rats with myocardial infarction(MI) have not been reported. The purpose of this study is to investigate the effects of glycine therapy on MF and comprehend its underlying mechanisms. Materials and Methods: Left anterior descending artery ligation-induced MI in Sprague Dawley rats was leveraged to assess the therapeutic effects of Glycine. Rats received either normal saline or glycine(0.5 mg/g bodyweight) for 7 days. Results: Glycine upregulated cardiac ejection fraction and fractional shortening to improve cardiac function, as evaluated by echocardiography. Histological and immunohistochemical analyses demonstrated that glycine could decrease inflammatory cell infiltration and alleviate collagen deposition. Western blotting revealed that nuclear factor-κB(NF-κB)-mediated inflammatory signaling was also downregulated by glycine treatment. The expression of signal transducer and activator of transcription 3(STAT3), tumor necrosis factor-α, and transforming growth factor-β(TGF-β) was decreased significantly in the glycine-treated group compared to the model group. Thus, glycine plays a protective role against myocardial ischemia and subsequent MF. Conclusion: The protective effects of glycine were achieved partly through STAT3/NF-κB/TGF-β signaling pathway.
