Real-time Quantitative RT-PCR for CT9 Level in Human Cancer

CT9 is a recently cloned cancer-testis antigen, which is a member of the bromodomain and extraterminal family. Each member of this protein family contains two N-terminal bromodomain motifs. We investigated the distribution of CT9 in different tissues and the possibility for it to be used as a potential therapeutic target in cancer treament. By using the real-time RT-PCR method and 18SrRNA as an internal standard, we analyzed the CT9 expression in several normal human tissues and in the tissues of patients suffering from cancer. The result of this study shows that the highest level of mRNA is only present in testis tissue because the CT9 expression has not been detected in other normal tissues. In 6 of 10 cases of gastric adenocarcinoma, in 3 of 10 cases of esophageal squamous cell carcinoma, in 2 of 9 cases of endometrial carcinoma and only in 1 of 12 cases of brain cancer, the low level expression of CT9 was detected. In none of the 12 cases of cervical squamous cell carcinoma, the expression of CT9 was detected. Since the high level expression of CT9 is only found in the normal testis tissue, but the low expression in cancer tissues, for example tissues of cervical squamous cell carcinoma, brain cancer, endometfial adenocarcinoma, esophageal squamous cell carcinoma, we conclude that CT9 cannot be used as a cancer therapeutic target molecule for cervical squamous cell carcinoma, brain cancer, endometrial adenocarcinoma, esophageal squamous cell carcinoma.

Effect of microenvironmental quantitative regulation on growth of Korean pine trees planted under secondary forest

Korean pine（Pinus koraiensis） and broadleaved mixed forest in Northeast China has been changed regressively into secondary forest with almost no *** Korean pine trees under the canopy of secondary forest is a feasible approach for recovering Korean pine and broadleaved mixed *** establishing an effective growth promotion method for under-canopy planted young Korean pine trees,two stands were selected as the experiment plots,Stand A（planted in 1989） and Stand B（planted in 1982）,and an experiment of microenvironment regulation was conducted relying mainly on Opening degree（K=1,K=1.5,K=2,CK） in *** results were shown that the adjustment had promoted growth of diameter and height of Korean pine planted in Stand A and Stand B,and had a significant influence on the growth rate of basal diameter,diameter at breast height and height in the two growth *** four years periodic increment of mean diameter and height of Korean pine planted in 1989 and in 1982 after regulation in K=1 level were 63.4%（D0） and 82.7%（H）,64.8%（D1.3） and 69.7%（H） higher than that of control *** regulation had significant influence on specific leaf area of Korean pine planted in 1989,and the current year specific leaf area（SLA） was lager than perennial year *** indexes of natural pruning capacity,normal form quotient and crown size was not significantly changed but shown a positive *** regulation scheme of Opening degree K=1 might be proper for adjusting the microenvironment of Korean pine trees planted under the canopy of secondary forest when the Korean pine trees were in the growth period of 15 to 26 years old in the experiment region.

This study explored the antibiofilm efficacy of 3,3’-diindolylmethane(DIM)on Staphylococcus aureus and its disinfection on common food-contact *** minimum biofilm inhibitory concentration(MBIC)of DIM on *** was 62.5μmol/L,while it did not impede the bacterial growth evaluated by growth curve and XTT reduction *** in the concentration range of 31.2-62.5μmol/L demonstrated a dose-dependent antibiofilm activity to ***,as confirmed by light microscopic(LM),confocal laser scanning microscopic(CLSM),and scanning electron microscopic(SEM)*** DIM of62.5μmol/L,the biomass of *** biofilm was significantly reduced by 97%and its average thickness by 58%(P<0.05).DIM of 62.5μmol/L inhibited the bacterial initial adhesion and proliferation,as well as cell motility;the release of extracellular DNA(eDNA)and extracellular polysaccharide(EPS)were reduced by 75%and 69%,*** exhibited a strong inhibition to *** biofilm formation on common food-contact surfaces,including 304 stainless steel,glass,and polyvinyl chloride(PVC)but not disperse the mature ***,our investigation identified DIM as a promising antibiofilm agent and its suitability to prevent the biofilm formation of *** on common food-contact surfaces utilized during food processing.

LyαEmission Enhancement Associated with Soft X-Ray Microflares

Lyα(Lyα,1216 A)is the strongest emission line in the solar ultraviolet *** the present work,we obtained a Lyαenhancement catalog covering flares larger than B1 class from the GOES/EUVS data during *** focused on the 242 B-class events which are less investigated,however,show non-negligible Lyαemission *** found that on average the Lyαpeak of B-class flares is 0.85%stronger than the *** the flare energetics,it is found that the weaker the soft X-ray(SXR)flare,the larger the ratio of the radiated energy in Lyαto *** the RHESSI data and multi-wavelength observations taken by SDO-AIA,we diagnose the thermal and non-thermal properties of several *** case studies show that the coincidence of the Lyαpeak with the SXR time-derivative peak is not a sufficient condition of the nonthermal property of a Lyα*** Lyαenhancement in the microflares may be caused by the nonthermal electron beams or/and thermal *** for typeⅢevents,we found that the delay of the Lyαpeak with respect to the SXR peak can be attributed to either the Lyαemission from a filament erupted or the cooling of the thermal plasma in flare ***,interestingly the Lyαemission from filaments can not only occur in the decay phase of the flare,but also in the preflare *** this case,the Lyαemission was originated from an erupted filament which probably initiated the flare.

Two novel two-dimensional(2D)supramolecular organic frameworks were fabricated in water based on the encapsulation-enhanced donor-acceptor interaction between the methyl viologen(MV)units,methoxy naphthyl(MN)units,and CB[8].The tetraphenylethylene(TPE)derivatives 1 with four MV units were employed as rigid building blocks and the two MN units modified oligoethylene glycol derivatives 2 and 3 served as flexible edges,*** obtained two SOFs have obvious sheet-like structures and exhibit fluorescence emission at 350-500 *** addition,these two SOFs were employed for the luminescent detection of Cr(Ⅵ)and Mn(Ⅶ)in aqueous solutions,and the detection limits of CrO_(4)^(2-),Cr_(2)_(O)_(7)^(2-),and MnO_(4)were calculated in a very low concentration range,indicating that these two SOFs can serve as a potential sensor for Cr(Ⅵ)and Mn(Ⅶ)detection in *** work constructs two SOFs in an aqueous solution through a facile method and further enriches the applications of SOFs.

Regulation of fertilization in male rats by CatSper2 knockdown

Interest in ion channels as drug targets for contraception has grown with the realization that certain ion channel subunits are located exclusively in sperm. Selective knockdown of ion channel subunits can lead to infertility without ill effects, and selective inhibitors and/ or openers of these ion channels could interfere with sperm function. In this study, in vivo electmporation （EP） and rete testis microinjection-mediated plasmid DNA were adopted to silence CatSper2 expression, which is essential in sperm hyperactivation. The results showed that high transfection efficiency and expression were achieved by plasmid DNA that was directly injected into the rete testis. As a result of the expression of CatSper2 being blocked, the treatment group showed significantly lower （P〈0.05） hyperactivation rate, fertilization rate in vitro, migration motility in viscoelastic solution and intracellular Ca2＋ peak. The low hyperactivation and fertilization rates lasted for 60 days. Meanwhile, analysis of the sperm survival rate and testis histology indicated that in vivo EP had no significant effect on the function of the testis, spermatogenesis or sperm activity. The present study demonstrated that it was feasible to achieve male contraception by silencing the expression of CatSper2, the key protein involved in sperm hvoeractivation.

The classification of low permeability-tight reservoirs is the premise of development. The deep reservoir of Shahejie 3 member contains rich low permeability-tight reserves, but the strong heterogeneity and complex micro pore structure make the main controlling factors subjective and the classification boundaries unclear. Therefore, a new indicator considering the interaction between fluid and rock named Threshold Flow Zone Indicator(TFZI) is proposed, it can be used as the main sequence of correlation analysis to screen the main controlling factors, and the clustering algorithm is optimized combined with probability distribution to determine the classification boundaries. The sorting coefficient, main throat radius, movable fluid saturation and displacement pressure are screened as the representative parameters for the following four key aspects: rock composition, microstructure, flow capacity and the interaction between rock and fluid. Compared with the traditional probability distribution and clustering algorithm, the boundary of the optimized clustering algorithm proposed in this paper is more *** classification results are consistent with sedimentary facies, oil levels and oil production *** method provides an important basis for the development of low permeability-tight reservoirs.

Hypoglossal motor neurons(HMNs) innervate tongue muscles and play key roles in a variety of physiological functions,including swallowing,mastication,suckling,vocalization,and *** of HMNs is associated with several diseases,such as obstructive sleep apnea(OSA) and sudden infant death *** A is a serious breathing disorder associated with the activity of HMNs during different sleep-wake *** the neural mechanisms by which the statedependent activities of HMNs are controlled may be helpful in providing a theoretical basis for effective therapy for ***,the presynaptic partners governing the activity of HMNs remain to be *** the present study,we used a cell-type-specific retrograde tracing system based on a modified rabies virus along with a Cre/loxP gene-expression strategy to map the whole-brain monosynaptic inputs to HMNs in *** identified 53 nuclei targeting HMNs from six brain regions:the amygdala,hypothalamus,midbrain,pons,medulla,and *** discovered that GAB Aergic neurons in the central amygdaloid nucleus,as well as calretinin neurons in the parasubthalamic nucleus,sent monosynaptic projections to *** addition,HMNs received direct inputs from several regions associated with respiration,such as the preBotzinger complex,parabrachial nucleus,nucleus of the solitary tract,and *** regions engaged in sleep-wake regulation(the parafacial zone,parabrachial nucleus,ventral medulla,sublaterodorsal tegmental nucleus,dorsal raphe nucleus,periaqueductal gray,and hypothalamus) also provided primary inputs to *** results contribute to further elucidating the neural circuits underlying disorders caused by the dysfunction of HMNs.
