Objective to evaluate the chemical composition and effects of Artemisia vulgaris(AV)hydroalcoholic extract(HEAV)on breast cancer cells(MCF-7 and SKBR-3),chronic myeloid leukemia(K562)and NIH/3t3 *** Phytochemical analysis of HEAV was done by high-performance liquid chromatography-mass(HPLC)*** and cell death studies were performed using trypan blue and Annexin/FItC-7AAD,***-1(Fer-1)and necrostatin-1(Nec-1)were used to assess the mode of HEAV-induced cell death and acetoxymethylester(BAPtA-AM)was used to verify the involvement of cytosolic calcium in this *** calcium measurements were made using *** HEAV decreased the viability of MCF-7,SKBR-3 and K562 cells(Pthe viability of HEAV-treated K562 cells was reduced compared to HEAV-exposed fibroblasts(Ptreatment of K562 cells with HEAV induced cell death primarily by late apoptosis and necrosis in assays using annexin V-FItC/7-AAD(Pthe use of Nec-1 and Fer-1 increased the viability of K562 cells treated with HEAV relative to cells exposed to HEAV alone(Pthermore,BAPtA-AM,an intracellular Ca^(2+)chelator,decreased the number of non-viable cells treated with HEAV(Ptotoxic and activates several modalities of cell death,which are partially dependent on lysosomal release of Ca^(2+).these effects may be related to artemisinin and caffeoylquinic acids,the main compounds identified in HEAV.

Urban Soil Compaction Remediation by Shallow tillage and Compost in Hydroseeded Lawn

Construction activities often involve removal of topsoil and compaction of the exposed soil by heavy equipments. Such compacted soils with low organic matter can lead to low infiltration and poor vegetation establishment. the objective of this study was to investigate the efficacy of tillage (shallow till) and compost on soil physical and biological properties in a hydroseeded lawn as a post-construction best management practice for soil compaction remediation. the experimental site received a total of four land treatments in five replicated trials and it was hydroseeded with common Bermuda grass: 1) No tillage + Compost (Nt-C), 2) No tillage + No Compost (Nt-NC;control), 3) tillage + Compost (t-C), and 4) tillage + No Compost (t-NC). Bulk density (BD), infiltration rate (IR), and wet aggregate stability (WAS) in each plot were measured to assess soil physical properties while soil organic matter (SOM) and enzyme activity (β-glucosidase, acid-phosphatase, and alkaline-phosphatase) were measured for soil biological properties. Over a 15-months of monitoring period, the shallow tillage loosened the soil initially, but its effect on BD without compost was diminished to control plot level (Nt-NC) within 4 months after hydroseeding. Both tillage and compost led to an increase in IR, and it remained higher than control by 2 - 3 times throughout the observation period. the WAS and β-glucosidase activity decreased in tilled plot unless there was compost application. turfgrass showed greener leaves and aggregated roots in the compost-amended plots (Nt-C and t-C). Our results suggest that compost application plays a key role in improving soil physical and biological properties in hydroseeded lawns from construction sites.

this review highlights some established and some more contemporary mechanisms responsible for heart failure(HF)-induced skeletal muscle wasting and *** first describe the effects of HF on the relationship between protein synthesis and degradation rates,which determine muscle mass,the involvement of the satellite cells for continual muscle regeneration,and changes in myofiber calcium homeostasis linked to contractile *** then highlight key mechanistic effects of both aerobic and resistance exercise training on skeletal muscle in HF and outline its application as a beneficial ***,HF causes multiple impairments related to autophagy,anabolic-catabolic signaling,satellite cell proliferation,and calcium homeostasis,which together promote fiber atrophy,contractile dysfunction,and impaired *** both wasting and weakness are partly rescued by aerobic and resistance exercise training in HF,the effects of satellite cell dynamics remain poorly explored.

Development of synthetic bone substitutes has arisen as a major research interest in the need to find an alternative to autologous bone *** an ovine model,the present pre-clinical study presents a synthetic bone graft(BonelikeVR)in combination with a cellular system as an alternative for the regeneration of non-critical *** association of biomaterials and cell-based therapies is a promising strategy for bone tissue *** stem cells(MSCs)from human dental pulp have demonstrated both in vitro and in vivo to interact with diverse biomaterial systems and promote mineral deposition,aiming at the reconstruction of osseous ***,these cells can be found and isolated from many ***-critical bone defects were treated with BonelikeVR with or without MSCs obtained from the human dental *** showed that BonelikeVR and MSCs treated defects showed improved bone regeneration compared with the defects treated with BonelikeVR ***,it was observed that the biomaterial matrix was reabsorbed and gradually replaced by new bone during the healing *** therefore propose this combination as an efficient binomial strategy that promotes bone growth and vascularization in non-critical bone defects.

Since 2014,"Bring Your Own Data"workshops(BYODs)have been organised to inform people about the process and benefits of making resources Findable,Accessible,Interoperable,and Reusable(FAIR,and the FAIRification process).the BYOD workshops'content and format differ depending on their goal,context,and the background and needs of ***-focused BYODs educate domain experts on how to make their data FAIR to find new answers to research ***-focused BYODs promote the benefits of making data FAIR and instruct project managers and policy-makers on the characteristics of FAIRification ***-focused BYODs gather software developers and experts on FAIR to implement or improve software resources that are used to support ***,these BYODs intend to foster collaboration between different types of stakeholders involved in data management,curation,and reuse(*** experts,trainers,developers,data owners,data analysts,FAIR experts).the BYODs also serve as an opportunity to learn what kind of support for FAIRification is needed from different communities and to develop teaching materials based on practical examples and *** this paper,we detail the three different structures of the BYODs and describe examples of early BYODs related to plant breeding data,and rare disease registries and biobanks,which have shaped the structure of the *** discuss the latest insights into making BYODs more productive by leveraging our almost ten years of training experience in these workshops,including successes and encountered ***,we examine how the participants'feedback has motivated the research on FAIR,including the development of workflows and software.

Correlative CD4 and CD8 t-cell immunodominance in humans and mice:Implications for preclinical testing

Antigen-specific t-cell recognition is restricted by Major Histocompatibility Complex(MHC)molecules,and differences between CD4 and CD8 immunogenicity in humans and animal species used in preclinical vaccine testing are yet to be fully *** this study,we addressed this matter by analyzing experimentally identified epitopes based on published data curated in the Immune Epitopes DataBase(IEDB)*** first analyzed SARS-CoV-2 spike(S)and nucleoprotein(N),which are two common targets of the immune response and well studied in both human and mouse *** observed a weak but statistically significant correlation between human and H-2^(b)mouse t-cell responses(CD8 S specific(r=0.206,p=1.37×10^(−13));CD4 S specific(r=0.118,p=2.63×10^(−5))and N specific(r=0.179,p=2.55×10^(−4))).Due to intrinsic differences in MHC molecules across species,we also investigated the association between the immunodominance of common Human Leukocyte Antigen(HLA)alleles for which HLA transgenic mice are available,namely,A*02:01,B*07:02,DRB1*01:01,and DRB1*04:01,and found higher significant correlations for both CD8 and CD4(maximum r=0.702,p=1.36×10^(−31)and r=0.594,p=3.04−122,respectively).Our results further indicated that some regions are commonly immunogenic between humans and mice(either H-2^(b)or HLA transgenic)but that others are human ***,we noted a significant correlation between CD8 and CD4 S-(r=0.258,p=7.33×10^(21))and N-specific(r=0.369,p=2.43×10^(14))responses,suggesting that discrete protein subregions can be simultaneously recognized by t *** findings were confirmed in other viral systems,providing general guidance for the use of murine models to test t-cell immunogenicity of viral antigens destined for human use.

Cr（V）involvementin the toxicity pathway of testicular damage

OBJECtIVE:the functional integrity of the blood-testis barrier (BtB) in male mice exposed to Cr(V) was studied in order to clarify the mechanism underlying testicular injury.;MEtHODS:Adult male mice were subcutaneously injected repeated doses of 8.02 micromol (0.5 ml) of Cr/*** for 5 days. Animals receiving a similar volume of bis(hydroxyethyl)-aminotris(hydroxymethyl)methane buffer (Bt) were used as controls. the animals were sacrificed on day 6 and small fragments of seminiferous tubules, approximately 8-10 mm length, were incised and sutured at both ends. they were exposed in vitro to horseradish peroxidase-containing culture medium for 10 minutes. tissues were then fixed and processed for ultrastructural studies.;RESULtS:Controls and Cr(V)-treated group resulted in the uptake of the tracer by Sertoli cells. However, the major finding consisted in the permeability of the BtB only in the Cr(V)-group, as evidenced by the presence of the tracer within the junctions between the neighbouring Sertoli cells.;CONCLUSIONS:the BtB is disrupted in mice submitted to Cr(V). the permeability of the BtB is a crucial feature to be investigated for the understanding of lesions within the seminiferous tubule.

We study a subclass of the May–Leonard stochastic model with an arbitrary,even number of species,leading to the rise of two competing partnerships where individuals are *** carrying out a series of accurate numerical stochastic simulations,we show that alliances compete each other forming spatial domains bounded by interfaces of empty *** solve numerically the mean field equations associated with the stochastic model in one and two spatial *** demonstrate that the stationary interface profile presents topological properties which are related to the asymptotic spatial distribution of species of enemy alliances far away from the interface ***,we introduce a theoretical approach to model the formation of stable interfaces using spontaneous breaking of a discrete *** show that all the results provided by the soliton topological model,presented here for the very first time,are in agreement with the stochastic simulations and may be used as a tool for understanding the complex biodiversity in nature.

In recent years,our world has experienced significant disruptions due to the COviD-19 pandemic,and Russia's 2022 invasion of Ukraine,impacting human activities and the global *** paper explored air quality changes in Ukraine due to COVID-19,and Russia's invasion of Ukraine using on-demand with a what-you-see-is-what-you-get *** the cOVID-19 pandemic,strict quarantine policies in Ukraine led to a 2%reduction in tropospheric NO_(2) concentration before the lockdown and 4%during the lockdown *** like Kyiv,Donetsk,and Dnipro exhibited reductions of 5%,11%,and 16%,*** SO_(2) column concentration decreased by 6%before the lockdown and 2.5%during the lockdown period,except in high population density *** showed the highest reduction of 17%in SO_(2) concentration,while Donetsk and Dnipro exhibited an 11%***,during the Russian invasion,there was a significant increase in tropospheric NO_(2) concentration in heavily destroyed Kharkiv while most eastern regions experienced a *** total SO_(2) column was 48%higher before the war but reduced throughout the country after the war,except for in Kyiv and a few central *** findings can contribute to analyzing air pollution and building digital twin simulations for future reconstruction scenarios.
