3,4-Dihydroxy L-phenylalanine(L-DOPA)is considered a potent drug for the treatment of Parkinson *** and nutritional parameters where optimized by using Yarrowia lipolytica-NCIM 3450 to accomplished the highest production of *** of critical components were completed by using a Plackett–Burman design,while further optimization was carried out using the Box–Behnken *** optimized factor levels predicted by the model were pH 6.1,1.659 g L^(-1)yeast extract,1.491 g L^(-1)L-tyrosine and 0.0290 g L^(-1)*** predicted yield of L-DOPA with these levels was 1.319 g L^(-1),while actual yield obtained was 1.273 g L^(-1).the statistical analysis revealed that model is significant with F value 19.55 and R2 value *** process resulted in a 3.594-fold increase in the yield of LDOPA.L-DOPA was confirmed by HPtLC and HPLC ***,Yarrowia lipolytica-NCIM 3450 has potential to be a new source for the production of L-DOPA.

Fluorescently encoded microbeads are in demand for multiplexed applications in different *** to organic dye-based commercially available Luminex's x MAP technology, upconversion nanoparticles(UCNPs) are better alternatives due to their large antiStokes shift, photostability, nil background, and single wavelength excitation. Here, we developed a new multiplexed detection system using UCNPs for encoding poly(ethylene glycol) diacrylate(PEGDA) microbeads as well as for labeling reporter antibody. However, to prepare UCNPs-encoded microbeads, currently used swellingbased encapsulation leads to non-uniformity, which is undesirable for fluorescence-based multiplexing. Hence,we utilized droplet microfluidics to obtain encoded microbeads of uniform size, shape, and UCNPs distribution inside. Additionally, PEGDA microbeads lack functionality for probe antibodies conjugation on their *** to functionalize the surface of PEGDA microbeads(acrylic acid incorporation, polydopamine coating)reported thus far quench the fluorescence of UCNPs. Here,PEGDA microbeads surface was coated with silica followed by carboxyl modification without compromising the fluorescence intensity of UCNPs. In this study, droplet microfluidics-assisted UCNPs-encoded microbeads of uniform shape, size, and fluorescence were *** color codes were generated by mixing UCNPs emitting red and green colors at different ratios prior to encapsulation. UCNPs emitting blue color were used to label the reporter antibody. Probe antibodies were covalently immobilized on red UCNPs-encoded microbeads for specific capture of human serum albumin(HSA) as a model protein. the system was also demonstrated for multiplexed detection of both human C-reactive protein(hCRP) and HSA protein by immobilizing anti-h CRP antibodies on green UCNPs.

Microalgae can be cultivated for producing high-valued products through the production of enzymes to offset the cost of CO_(2) sequestration,providing financial *** viability of algae in the photobioreactor needs to be monitored to ensure biologically active live *** this study,we explored a simple fluorometry method for differentiation of live and dead algal cells in photobioreactors by fluorescein diacetate(FDA)and propidium iodide(PI)fluorescence *** stains fluorescent green to the living cells while PI stains the dead cells,allowing the discrimination of live and dead *** method was evaluated using two green algae and two strains of cyanobacteria grown in shake flasks and a continuously stirred *** method was found applicable for Chlorella pyrenoidosa and Synechococcus 7002 but was not applicable for the cultures of Scenedesmus dimorphus and Synechococcus elongatus *** conclude that FDA is a good stain for monitoring live algal cells in photobioreactors but its applicability to individual species of algae must be evaluated.

Phytochemical profiling of Artocarpus lakoocha Roxb. leaf methanol extract and its antioxidant, antimicrobial and antioxidative activities

Objective:to explore the phytochemical profile of Artocarpus lakoocha *** both qualitatively and quantitatively,and validate its role as a potent antioxidant and antimicrobial ***:Extraction and isolation of different compounds were done from the leaves of Artocarpus lakoocha based on solvent fractionation ***,quantitative and qualitative phytochemical profiling along with antioxidant,antimicrobial and antioxidative activities were tested following standard ***:Among the five fractions,methanol fraction of Artocarpus lakoocha exhibited higher content of phytochemical compounds[phenols=(3175.21±290.43)mg GAE/g dry extract,flavonoids=(1173.15±47.52)mg QE/g dry extract and tannins=(923.53±95.21)mg tAE/g dry extract]as compared to other *** methanol fraction showed the highest antioxidant activity in DPPH and ABtS radical scavenging assays with IC50 of(111.98±34.20)μg/mL and(138.26±0.66)μg/mL,respectively,and the best reduction potential with a value of(316.81±2.96)mg QE/g dry extract in reducing power *** was significant correlation between the amount of phytochemicals and antioxidant ***,the extract successfully protected Lambda phage DNA from damage at 5 and 6 mg/mL concentration and exhibited substantial bactericidal as well as fungicidal *** GC-MS analysis of methanol fraction of Artocarpus lakoocha revealed diethyl phthalate as the main phytochemical compound,along with 3,4-dihydroxymandelic acid,9-octyl eicosane and 7,8-didehydro-3-methoxy-17-methyl-6-methylene ***:the methanol fraction of Artocarpus lakoocha could be used as a potent antioxidant and antimicrobial agent for sustainable agriculture and pharmaceutical purposes.

Objective: to investigate the therapeutic efficacy of andrographolide, a plant derived compound, against chikungunya virus(CHIKV) infection. Methods: Using flow cytometry and immunoblotting assay, in vitro viral protein expression was studied in tHP-1 cells line. In Balb/c mouse neonates, viral RNA copy number was determined by real time PCR. Results:the results showed reduced CHIKV protein expression on andrographolide treatment in CHIKV-infected human peripheral blood mononuclear cells, Vero cells and tHP-I cell *** vivo, andrographolide treatment to CHIKV-infected neonates reduced viral RNA copy number. Further. andrographolide also increased cytotoxic t lymphocytes both in vitro and in vivo. Andrographolide also activated host innate immune pathways, viz., protein kinase *** eukaryotic initiation factor 2 α, retinoic acid inducible gene-Ⅰ and interferon regulatory factor 3/7, thereby increasing IFN-a secretion. CHIKV-induced nuclear factor κlight chain enhancer of activated B cells and tumor necrosis factor-a was also reduced on andrographolide treatment. Conclusion: Andrographolide inhibits CHIKV by suppressing viral protein expression and up-regulating host innate immunity and hence could be an effective therapeutic agent against CHIKV infection.

Development of a sensitive and rapid method for quantitation of (S)-(-)- and (R)-(+)-metoprolol in human plasma by chiral LC–ESI–MS/MS

A selective, sensitive and high throughput liquid chromatography-tandem mass spectro-metry （LC-ESI-MS/MS） method has been developed for separation and quantification of metoprolol enantiomers on a chiral Lux Amylose-2 （250 mm＆#215;4.6 mm, 5 mm） column. Solid phase extraction of （S）-（-）- and （R）-（t）-metoprolol and rac-metoprolol-d6 as an internal standard （IS） was achieved on Lichrosep DVB HL cartridges employing 200 mL human plasma. Both the analytes were chromatographically separated with a resolution factor of 2.24 using 15 mM ammonium acetate in water, pH 5.0 and 0.1% （v/v） diethyl amine in acetonitrile （50：50, v/v） as the mobile phase within 7.0 min. the precursor-product ion transitions for the enantiomers and IS were monitored in the multiple reaction monitoring and positive ionization mode. the method was validated over the concentration range of 0.500-500 ng/mL for both the enantiomers. Matrix effect was assessed by post-column analyte infusion experiment and the mean extraction recovery was greater than 94.0% for both the enantiomers at all quality control levels. the stability of analytes was evaluated in plasma and whole blood under different storage conditions. the method was successfully applied to a clinical study in 14 healthy volunteers after oral administration of 200 mg metoprolol tablet under fasting conditions. the assay reproducibility is shown by reanalysis of 68 incurred samples. the suitability of the developed method was assessed in comparison with＆amp;nbsp;different chromatographic methods developed for stereoselective analysis of metoprolol in biological matrices.
