The objective of this study was to evaluate whether repeated oronasal adm.nistration of LPS before parturition showed effects on m.tabolic and clinical responses in periparturient dairy cows. Hundred Holstein dairy cows were random.y allocated to two treatm.nt groups (n = 50). Thirty cows out of 100 were random.y assigned for intensive sam.ling (n = 15) started at 28 d before the expected day of parturition. Cows received an oral and a nasal treatm.nt of 2 m. and 1 m. of sterile saline solution (0.15 m.of NaCl), respectively, alone (control), or containing 3 increasing doses of LPS from.Escherichia coli 0111:B4 as follows: 1) 0.01 μg/kg body weight (BW) on d ﹣28, 2) 0.05 μg/kg BW on d ﹣25, and ﹣21, and 3) 0.1 μg/kg BW on d ﹣18 and ﹣14. Blood sam.les were collected from.coccygeal vein at different tim. points around parturition and analyzed for glucose, lactate, non-esterified fatty acids (NEFA), cholesterol, and betahydro-xybutyrate (BHBA). Clinical m.nitoring of anim.ls was done throughout the experim.nt at different tim. points for overall health status, udder edem. (UE), m.nure score, and body condition score (BCS). Results showed that oronasal adm.nistration of LPS increased concentrations of glucose and cholesterol in the serum.com.ared to the control group (P 0.05). Oronasal LPS did not influence BCS, m.nure score or the incidence of UE (P > 0.05). Overall, repeated oronasal adm.nistration of LPS m.dulated som. serum.m.tabolites related to energy m.tabolism.around parturition in the treated cows. Further research is warranted to elucidate the m.chanism. behind greater glucose and cholesterol status in the serum.and their potential effects on long-term.m.tabolic health status of dairy cows.

The aim.of this study was to evaluate the effects of repeated oronasal adm.nistration of lipopolysaccharide (LPS) on m.lk com.osition and the overall productive perform.nce of dairy cows. One hundred pregnant Holstein dairy cows were random.y assigned to two treatm.nt groups (n = 50). 30 cows out of 100 were selected for intensive sam.ling (n = 15) starting at 28 d before parturition. Cows were adm.nistered orally and nasally with 2 and 1 m. of saline solution, respectively (control), or saline solution containing 3 doses of LPS from.Escherichia coli 0111:B4 as follows: 1) 0.01 μg/kg body weight (BW) on d ﹣28, 2) 0.05 μg/kg BW on d ﹣25 and ﹣21, and 3) 0.1 μg/kg BW on d ﹣18 and ﹣14. Daily feed intake and m.lk production were recorded for each cow during the first 28 d postpartum. m.lk sam.les were obtained once per week and analyzed for various m.lk com.onents. Overall, results indicated that treatm.nt did not affect feed intake, m.lk yield, m.lk efficiency, fat content, fat yield, protein content, protein yield, lactose content, lactose yield, m.lk urea nitrogen (m.N), total solid contents, fat-corrected m.lk (FCm., and energy-corrected m.lk (ECm.P > 0.05). Profiles of energy m.tabolites and haptoglobin (Hp) in dairy cows that are transitioned from.conventional to organic m.nagem.nt in various Alberta farm. were com.ared with those of dairy cows m.naged conventionally at the University of Alberta dairy farm. Blood sam.les were collected during the following periods: Dry, 0 - 30, 30 - 60, and 60 - 90 days in m.lk (DIm. n = 7 cows). Concentrations of m.tabolites were evaluated by enzym.tic colorim.tric m.thods. Concentrations of Hp were determ.ned by bovine ELISA kits. Data were analyzed by the m.xed procedures of SAS. Concentrations of NEFA and BHBA in blood were elevated (P m.diate 30 to 60, and 60 to 90 d, and lower in the dry period. In addition, BHBA was higher (P m.l/L in conventional and organic cows at 0 - 30 d, respectively). Serum.concentrations of cholesterol increased with increasing DIm.and returned to nadir levels during dry period and was higher (P m.diate 30 to 60 and 60 to 90 d, and peaked during the dry period (P m.or day × farm.type but was higher (P m.concentrations of Hp were elevated during dry period;reached peak levels 0 to 30 d and decreased gradually with increasing days postpartum.and were m.ch higher at all periods in conventional than organic cows. The Effects of Zinc Sulfate on the in Vitro Digestibility of Feeds in Cervids

The captive white-tailed deer industry has an estim.ted im.act of 1.6 billion USD in the state of Texas alone. However, nutritional requirem.nts for cervids are determ.ned through research based on sheep and goats. The objective of this study was to determ.ne the effects of zinc on differences in dry m.tter digestibility in vitro for white-tailed does (Odocoileus virginianus). Deer (n = 2) were ethically harvested, rum.ns were collected, and placed into a cooler containing warm.water. Rum.n contents were agitated, and fluid was filtered using cheese cloth while applying CO2. Fluid was placed into four separate incubator jars with filter bags containing a 1:1 alfalfa to coastal hay blend. Zinc doses of 0.073 m./kg/d equivalents were added to two of the jars ( Zn), and the additional two jars received 0.00 m./kg/d (CON). Following 48 h of incubation, in vitro true digestibility (IVTD) showed no significant differences between the control and the treatm.nt groups. Average dry m.tter digested in vitro was 91.87% and 95.13%, respectively. There were no differences detected in ADF, NDF, IVTD, or Om.between the treatm.nt groups. While no detectable differences were observed in this study, this m.thodology did prove to be viable and functional for m.crobial digestion in vitro. This study can be replicated with m.ltiple experim.ntal units to confirm.the observations of increased digestibility. Form.l nutritional guidelines can be created to allow for m.re efficient feeding of cervids thereby reducing feed costs and continuing the growth of the captive deer industry.

m.lecular profiling of indolent hum.n prostate cancer： tackling technical challenges to achieve high-fidelity genom.-wide data

The contem.orary problem.of prostate cancer overtreatm.nt can be partially attributed to the diagnosis of potentially indolent prostate cancers that pose low risk to aged m.n, and lack of sufficiently accurate risk stratification m.thods to reliably seek out m.n with indolent diseases. Since progressive acquisition and accum.lation of genom.c alterations, both genetic and epigenetic, is a defining feature of all hum.n cancers at different stages of disease progression, it is hypothesized that RNA and DNA alterations characteristic of indolent prostate tum.rs m.y be different from.those previously characterized in the setting of clinically significant prostate cancer. Approaches capable of detecting such alterations on a genom.-wide level are the m.st prom.sing. Such analysis m.y uncover m.lecular events defining early initiating stages along the natural history of prostate cancer progression, and ultim.tely lead to rational developm.nt of risk stratification m.thods for identification of m.n who can safely forego treatm.nt. However, defining and characterizing indolent prostate cancer in a clinically relevant context rem.ins a challenge, particularly when genom.-wide approaches are em.loyed to profile form.lin-fixed paraffin-em.edded （FFPE） tissue specim.ns. Here, we provide the conceptual basis underlying the im.ortance of understanding indolent prostate cancer from.m.lecular profiling studies, identify the key hurdles in sam.le acquisition and variables that affect m.lecular data derived from.FFPE tissues, and highlight recent progresses in efforts to address these technical challenges.

The Effects of Initial Cell Density on the Growth and Proliferation of the Potentially Toxic Cyanobacterium.m.crocystis aeruginosa

Increasing input of nutrients is leading to significant eutrophication in aquatic system. across the globe. Toxin production in those bloom. is correlated with high cell density. A study was done to investigate the effects of initial cell density on the growth rate of the potentially toxic cyanobacterium.m.crocystis aeruginosa, in unfiltered lake water as well a com.on algal growth m.dium.under laboratory conditions. Five treatm.nts were established in the laboratory varying the initial cell density of m. aeruginosa and the growth m.dium. Unfiltered lake water was inoculated with a low, m.dium. and high initial cell density and Bold’s m.dium.was inoculated with low and high initial cell densities. There was a significant difference in final cell density between the lake water treatm.nts with the high inoculum.of cell treatm.nt reaching the highest final cell density. There was also a significant difference in final cell density between the Bold’s m.dium.treatm.nts. These results indicate there is a relationship between initial cell density of m. aeruginosa and final cell density, however, the results are sim.lar when nutrients are available. Diverse algal com.unities can keep toxin producing algae in low density therefore precluding the need of toxin production.

Background： Failure to expel fetal m.m.ranes within 24 h of calving is a pathological condition defined as retained placenta（RP）. The objective of this investigation was to evaluate whether there are alterations in several selected serum.variables related to innate im.unity and carbohydrate and lipid m.tabolism.that precede occurrence of RP in transition Holstein dairy ***： One hundred m.ltiparous Holstein dairy cows were involved in the study. Blood sam.les were collected from.the coccygeal vein during the-8 to ＋4 wks around parturition, once per week before the m.rning *** healthy control cows（CON） and 6 cows with RP were selected and serum.sam.les at-8,-4, tim. of diagnosis of disease, and ＋4 wks relative to parturition were used for analyses. All sam.les were analyzed for lactate, nonesterified fatty acids（NEFA）, β-hydroxybutyrate（BHBA）, interleukin-1（IL-1）, interleukin-6（IL-6）, tum.r necrosis factor（TNF）, haptoglobin（Hp）, and serum.am.loid A（SAA）.Results： Cows with RP had greater concentrations of serum.lactate, IL-1, IL-6, TNF, and SAA in com.arison with CON cows. Intriguingly, elevated concentrations of all five variables were observed at-8 and-4 wks before the occurrence of RP com.ared to healthy cows. Cows with RP also had lower Dm. and m.lk production vs CON anim.ls; however m.lk com.osition was not affected by ***： Cows with RP showed an activated innate im.unity 8 wks prior to diagnosis of disease. Overall results suggest that serum.IL-1, IL- 6, and TNF, and lactate can be used as screening biom.rkers to indicate cows that m.ght have health issues during the transition period.

Although infrequent, there are patients who present them.elves to surgeons and ask that a lim. be am.utated. Generally, such patients fall into one of three categories. The first is those suffering from.“Body Integrity Identity Disorder” (BIID), a condition in which individuals feel that one of their lim.s is foreign to the rest of their body, and m.st be rem.ved. The second category is Apotem.ophilia, a fetishized desire to becom. an am.utee to enhance sexual gratification. Such drive for sexual gratification m.y lead to requests for am.utation. Finally, the third category is delusional beliefs secondary to severe m.ntal illness that m.y com.el an individual to seek am.utation, or even attem.t self-am.utation. We present a fourth type of patient: the Global War on Terrorism.veteran with a severely injured arm.dem.nding am.utation to fit in with others who use prosthetic devices. In this instance, the cause of injury was a m.tor vehicle collision. We speculate that our case is not the only one and that a fourth category of patient dem.nding am.utation is em.rging. Further study is needed.
