Effects of albendazole,artesunate,praziquantel and miltefosine,on Opisthorchis viverrini cercariae and mature metacercariae

Objective:To explore larvicidal effects of anthelmintic drugs on Opisthorchis viverrini(***) for alternative approach to interrupting its cycle for developing a field-based control ***:The larvicidal activities of albendazole(A1),artesunate(Ar),praziquantel(Pzq) and miltefosine(Mf) on *** cercariae and mature metacercariae were *** concentrations(LC_(50) and LC_(95)) of these drugs were *** metacercariae previously exposed to various concentrations of the drugs were administered to *** were harvested 30 d post infection and worm recovery rates ***,Ar,Pzq and Mf produced morphological degeneration and induced shedding tails of cercariae after 24 h ***:The LC_(50) and LC_(95) of Al,Ar,Pzq and Mf on cercariae were 0.720 and 1.139,0.350 and 0.861,0.017 and 0.693,and 0.530 and 1.134 ppm,***_(50) and LC_(95) of Ar on mature metacercariae were 303.643 and 446.237 ppm and of Mf were 289.711 and 631.781 ppm,respectively but no lethal effect in Pzq-and Al-treated groups(up to 1 ppt).No worms were found in hamsters administered Pzq-treated *** adult worms from Al-treated metacercariae were significantly bigger in size compared to the control group(P<0.05).Fecundity and body width were greater in adults from Mf-treated metacercariae compared to the control group(P<0.05).Conclusions:The larvicidal effects of these drugs were high efficacy to *** cercariae but lesser efficacy to *** should be further studied with the eventual aim of developing a field-based control program.

Immunological evidence of Zika virus transmission in Thailand

Objective: To identify immunological evidence of Zika virus transmission in Thailand. Methods: To undertake a preliminary serosurvey of possible exposure to Zika virus, 21 serum samples from cohort of acute undifferentiated fever patients were examined for immunoreactivity to Zika, Dengue, Japanese encephalitis and Chikungunya envelope antigens by Western blot analysis. Results: Twenty of the 21 serum samples showed immunoreactivity to at least one of the antigens, with seven samples showing immunoreactivity to all antigens. Of particular note, two serum samples showed immunoreactivity only to Zika envelope antigen, with no immunoreactivity to other envelope antigens. Conclusions: This study presents the first evidence of Zika virus transmission in Thailand, although as yet the relationship between transmission and possible cases of Zika fever in Thailand requires further investigation.

The effect of carbon source and supporting material compositions on callus induction and plant regeneration from mature seeds of two indica rice varieties(RD6 and RD15)was studied in the present ***1 medium was used for callus induction from the sterilized *** then,three-week-old calli were transferred to four media(NBSl,NBS 2,NBM1 and NBM2)which were different in carbon sources and supporting materials for 3 *** of RD6 and RD15 varieties derived from the NBM2 medium(30 g Lmaltose as a carbon source and 2 g Lagar+4 g LPhytagelas a supporting material)showed the higher frequency for green spots and shoot regenerations when compared to the *** spot frequency was 91.67%and83.33%in RD6 and RD15 varieties,***,calli obtained from the same medium also showed maximum shoot regenerations in RD6 and RD15 varieties(75.00%and 66.67%,respectively).Well-developed plantlets were hardened and transferred to the soil.

Cytokine gene expression in human hepatocytes infected with dengue virus serotype 3 (strain-16562)

Liver is a site of viral replication and liver dysfunction is a characteristic of severe dengue infection. To understand these mechanisms, we analyzed the response of a hepatic cell linage, HepG2 to infection with dengue 3 virus (strain 16562). Steady state levels of mRNA accumulation were assessed for 14 genes involved in modulation of the host immune responses, at 6, 24 and 48 hpi, by quantitative reverse transcription real-time PCR (qRT-PCR). Fourteen genes showed altered expression upon infection with D3V including;cytokines/chemokines (IL-1β, IL-6, IL-8, RANTES, MCP-2, IL-2Rα and TGF-βIIIR), type I interferon (IFN-α and IFN-β), and pattern-recognition receptors (TLR3, TLR8, RIG-1, MDA5 and MyD88). Although these genes are associated with mechanism of innate immune response and anti-viral activity, their altered expression does not inhibit D3V (strain 16562) growth kinetics and virus yield in HepG2 cells. Gene expression in liver may explain pathological changes associated with dengue virus infection.

目的确定寨卡病毒在泰国传播的免疫学证据。方法对可能感染寨卡病毒的人群进行血清学调查,共对21名未明确病因的急性发热患者进行免疫学检查,检查内容包括寨卡病毒、登革热、日本脑炎和基孔肯雅热抗原免疫印迹分析。结果 21份血清样本中有20份显示至少存在一个抗原免疫反应阳性,7份样本显示所有抗原均呈阳性。其中两份血清样本仅显示寨卡病毒抗原阳性,而没有其它抗原检测阳性结果。结论本研究首次证实了寨卡病毒已经在泰国传播,但疑似病例如何感染寨卡病毒并传播尚需进一步调查。
