Inflammatory mediator release by Brugia malayi from macrophages of susceptible host Mastomys coucha and THP-1 and RAW 264.7 cell lines

Objective:To investigate which life stage of the parasite has the ability to stimulate release of pro- or anti-inflammatory mediators from macrophages.Methods:The human macrophage/ monocyte cell line THP-1,the mouse macrophage cell line RAW 264.7 and naive peritoneal macrophages（PM） from the rodent host Mastomys coucha（M.coucha） were incubated at 37℃in 5%CO2 atmosphere with extracts of microfilariae（Mf）,third stage infective larvae（L3） and adult worms（Ad） of Brugia malayi.After 48 hr post exposure,IL-1β,IL-6,TNF-α,IL-10 and nitric oxide（NO） in cell-free supernatants were estimated.Results:Extracts of all the life stages of the parasite were capable of stimulating pro-（IL-1β,IL-6 and TNF-α） and anti-inflammatory （IL-10） cytokines in both the cell lines and peritoneal macrophages of M.coucha.Mf was the strongest stimulator of pro-inflammatory cytokines followed by L3 and Ad;however,Ad was a strong stimulator of IL-10 release.Mf was found to have potential to modulate LPS-induced NO release in RAW cells.Ad-induced NO release was concentration dependent with maximum at 20μg/mL in both RAW and PMs.Conclusions:The results show that parasites at all life stages were capable of stimulating pro-（IL-1β,IL-6 and TNF-α） and anti-inflammatory（IL-10） cytokines and NO release from macrophages of susceptible host M.coucha,human and mouse macrophage cell lines.Mf can suppress the LPS-induced NO release in RAW cells.The findings also show that the two cell lines may provide a convenient in vitro system for assaying parasite-induced inflammatory mediator release.

Role of Cartilage as a Graft Material for Tympanic Membrane and in Middle Ear Reconstruction

Objective: The work was planned to evaluate the results of cartilage graft in the surgical treatment of chronic suppurative otitis media. Study Design: A prospective study. Materials and Methods: The present study was a prospective study of tympanoplasties and tympanomastoid surgeries performed on 100 patients. The main outcome measures were both anatomical and functional in form of graft incorporation and postoperative hearing function. Results: Cartilage was used as tympanic membrane and/or ossicle graft in the cases. There were no immediate postoperative or long term complications of surgery except for 10 cases in which there was a failure of graft uptake. There was a 7.6-decibel (dB) improvement in mean air conduction threshold post-operatively. A mean closure of average air bone gap of 8.4 decibels (dB) was noted which was statistically significant. Conclusion: The cartilage graft is a very effective option for the repair of the conducting mechanism of the ear with good take-up rates, less chances of rejection or extrusion and very few significant complications. The thickness of cartilage creates stiffness that is more resistant than the fascia to the anatomic deformities caused by negative middle ear pressure thus improving the long term integrity of the graft.

Novel targeting approaches and signaling pathways of colorectal cancer: An insight

Colorectal cancer(CRC) is the third most common cancer of mortality in the world. Chemotherapy based treatment leads to innumerable side effects as it delivers the anticancer drug to both normal cells besides cancer cells. Sonic Hedgehog(SHH), Wnt wingless-type mouse mammary tumor virus/β-catenin, transforming growth factor-β/SMAD, epidermal growth factor receptor and Notch are the main signaling pathways involved in the progression of CRC. Targeted therapies necessitate information regarding the particular aberrant pathways. Advancements in gene therapies have resulted in the recognition of novel therapeutic targets related with these signal-transduction cascades. CRC is a stepwise process where mutations occur over the time and activation of oncogenes and deactivation of tissue suppressor genes takes place. Genetic changes which are responsible for the induction of carcinogenesis include loss of heterozygosity in tumor suppressor genes such as adenomatous polyposis coli, mutation or deletion of genes like p53 and K-ras. Therefore, many gene-therapy approaches like gene correction, virusdirected enzyme-prodrug therapy, immunogenetic manipulation and virotherapy are currently being explored. Development of novel strategies for the safe and effective delivery of drugs to the cancerous site is the need of the hour. This editorial accentuates different novel strategies with emphasis on gene therapy and immunotherapy for the management of CRC.

Biomass production from microalgae Chlorella grown in sewage,kitchen wastewater using industrial CO_(2)emissions:Comparative study

Microalgae cultivation using organic wastewater like kitchen wastewater(KWW)and sewage wastewater(SWW)are recommended for wastewater treatment along with simultaneous biomass production which is a cost effective strategy.KWW is rich in nutrients which might be an efficient wastewater feedstock for microalgae cultivation to achieve reasonably high biomass yield for energy generation.KWW has not been investigated extensively for microalgae cultivation till now because of high concentration of ammonia as it could inhibit algae growth.In order to make KWW suitable for microalgae growth,a common method is to dilute with water.Hence,in the present study,KWW was diluted with water at dilution ration 1:1 and compared with SWW for microalgae biomass growth,biomass yields and nutrient removal efficiency specifically using Chlorella.Both KWW and SWW showed highest biomass productivity in mixotrophic cultivation mode.The average biomass productivity in SWW was 0.6 g/L which was higher when compared to KWW.Fourier infrared spectroscopy(FTIR)of algal biomass revealed the presence of certain organic compound groups such as eOH,eCOOH,NH_(2),and ■O.It was concluded from the results that Chlorella was very effective in the treatment of KWW,SWW and industrial flue gas on the other hand the biomass obtained is a sustainable green energy source for biofuels.

Pseudomonas fluorescens-like bacteria from the stomach:A microbiological and molecular study

AIM:To characterize oxidase-and urease-producing bacterial isolates,grown aerobically,that originated from antral biopsies of patients suffering from acid peptic diseases.METHODS:A total of 258 antral biopsy specimens were subjected to isolation of bacteria followed by tests for oxidase and urease production,acid tolerance and aerobic growth.The selected isolates were further characterized by molecular techniques viz.amplifications for 16S rRNA using universal eubacterial and HSP60 gene specific primers.The amplicons were subjected to restriction analysis and partial sequencing.A phylogenetic tree was generated using unweighted pair group method with arithmetic mean(UPGMA) from evolutionary distance computed with bootstrap test of phylogeny.Assessment of acidity tolerance of bacteria isolated from antrum was performed using hydrochloric acid from 10-7 mol/L to 10-1 mol/L.RESULTS:Of the 258 antral biopsy specimens collected from patients,179(69.4%) were positive for urease production by rapid urease test and 31%(80/258) yielded typical Helicobacter pylori(H.pylori) after 5-7 d of incubation under a microaerophilic environment.A total of 240(93%) antral biopsies yielded homogeneous semi-translucent and small colonies after overnight incubation.The partial 16S rRNA sequences revealed that the isolates had 99% similarity with Pseudomonas species.A phylogenetic tree on the basis of 16S rRNA sequences denoted that JQ927226 and JQ927227 were likely to be related to Pseudomonas fluorescens(P.fluorescens).On the basis ofHSP60 sequences applied to the UPGMA phylogenetic tree,it was observed that isolated strains in an aerobic environment were likely to be P.fluorescens,and HSP60 sequences had more discriminatory potential rather than 16S rRNA sequences.Interestingly,this bacterium was acid tolerant for hours at low pH.Further,a total of 250(96.9%) genomic DNA samples of 258 biopsy specimens and DNA from 240 bacterial isolates were positive for the 613 bp amplicons by targeting P.fluorescens-specific conserved putative oute

Effect of Semecarpus anacardium fruits on reproductive function of male albino rats

Aim: To evaluate the effect of an ethanolic extract of Semecarpus anacardium fruits on spermatogenesis in albino rats. Methods: Male albino rats were fed with a 50 % ethanolic extract of Semecarpus anacardium fruit at 100 mg·kg-1.day-1, 200 mg·kg-1·day-1 and 300 mg·kg-1·day-1 for 60 days. Fertility test was performed after 60 days of treatment. Sperm motility and density were observed in the cauda epididymis. Biochemical and histological analyses of the blood and reproductive organs were done. Recovery of fertility was followed to evaluate the reversibility of drug action. Results: S. anacardium fruit extract administration resulted in spermatogenic arrest in albino rats. The sperm motility and density was reduced significantly. The RBC and WBC counts, haemoglobin, haematocrit, blood sugar and urea were found to be within the normal range in the whole blood. The protein, cholesterol and glycogen in the testes and the fructose in the seminal vesicle were significantly decreased after the treatment. The fruit extract feeding caused marked reduction in the number of primary spermatocytes, secondary spermatocytes and spermatids. The number of mature Leydig cells was also decreased and degenerating cells increased proportionately. Conclusion: 5. anacardium fruit extract causes spermatogenic arrest in albino rats.

Study of the distribution profile of piperidine alkaloids in various parts of Prosopis juliflora by the application of Direct Analysis in Real Time Mass Spectrometry(DART-MS)

Direct Analysis in Real Time Mass Spectrometry(DART-MS)was applied to identify and study the distribution profile of piperidine alkaloids in different parts of Prosopis juliflora,without isolation and separation of the compounds by standard chromatographic techniques.With the help of DART-MS,chemical fingerprint of raw plant parts were generated,which revealed the presence of piperidine alkaloids in leaf,pod and flower.A comparative study of the distribution pattern,showed variation in the presence and distribution of these alkaloids in various parts of P.juliflora.The leaves and pod displayed the largest alkaloid pattern with a total of 12 different alkaloids in each part,whereas only 4 alkaloids were present in flower.Alkaloids:julifloridine,prosopine,prosopinine and prosafrinine were ubiquitously distributed in all the alkaloid rich plant parts.Juliprosopine was pre-eminet alkaloid in leaf,whereas pod and flower displayed copious amounts of julifloridine.

Hydrometeorological assessments and suspended sediment delivery from a central Himalayan glacier in the upper Ganga basin

Integrated hydrometeorological investigations are not frequently available at a regional scale over a longer time period, especially near the terminus of Indian Himalayan glaciers. An integrated approach to the collection of hydrological data has major advantages for understanding the runoff generation mechanisms at basin scale, particularly when coupled with meteorological observations. The current study involves time series analysis of hydrometeorological records collected near the terminus of the Chorabari Glacier, for four consecutive ablation seasons(June-Sept.) 2009-2012. The analysis shows that variation in rainfall was higher(c= 0.9) at the same elevation over proximal sites, while the intensity of extreme rainfall events was 121-160 mm/d. The diurnal temperature range(DTR) has a tendency to reduce over the ablation season because of the onset of the Indian Summer Monsoon(ISM) and then further increases during the ISM withdrawal indicating humid-temperate conditions. The peak discharge(Qpeak) was found to be higher during July and August. Snow and glacier melt contributed 76% of the total suspended sediment transport during peak ISM months(July and August) reflecting seasonal evolution of the hydrologic conduits. The results indicate that Karakoram and western Himalayan glaciers produce comparatively low sediment yield compared to central Himalayan glaciers. The hydrological variations are depicted through flow duration curves(FDC) for meltwater discharge and sediment load. The flow corresponding to Q, Q, and Q(where Qx is the discharge that is exceeded x percent of the time referred to as % dependability) are 4.2, 3.7, and 2.8 m~3/s; and the corresponding dependability for suspended sediment loads(SSLs) are 409.0, 266.0, and 157.2 t/d, respectively. The daily SSL and discharge(Q) from 2009 to 2012 were used to develop a sediment rating curve(SSL = 39.55 × Q. Herbal remedies,vaccines and drugs for dengue fever:Emerging prevention and treatment strategies

Dengue fever is a disease that is caused by five dengue virus(DENV) serotypes. It is endemic in more than 128 countries. Millions of people are affected by this disease. But still, there is no specific treatment available till now to combat it. Some of the preventive measures using plant extracts showed certain promising ovicidal activity against Aedes aegypti. There are some vaccines as preventive measure and antiviral agents to cure the dengue fever, which are under clinical trials but their results have not been reported or approved yet. Dengvaxia is the only licensed vaccine to prevent dengue fever in some South American countries, but it is not approved by other regulatory authorities worldwide. Scientific research has been going on to find a permanent cure for dengue fever, but till now, no successful invention has been done. There are some general treatments for dengue patient by employing analgesics and fluid replacement; however, specific treatment is needed to overcome the lethal effect of dengue fever. Since this disease has affected millions of people and not much invention has been made in this field; therefore, advancement in dengue therapy is required for the safety and well-being of dengue patients. Herein we collectively describe the different chemotherapeutics agents, alternative methods like vaccines and antiviral agents to prevent and cure this lethal disease. Furthermore, the future perspective for the treatment of dengue is discussed in this review.

Resistance to second generation antiandrogens in prostate cancer: pathways and mechanisms

Androgen deprivation therapy targeting the androgens/androgen receptor(AR)signaling continues to be the mainstay treatment of advanced-stage prostate cancer.The use of second-generation antiandrogens,such as abiraterone acetate and enzalutamide,has improved the survival of prostate cancer patients;however,a majority of these patients progress to castration-resistant prostate cancer(CRPC).The mechanisms of resistance to antiandrogen treatments are complex,including specific mutations,alternative splicing,and amplification of oncogenic proteins resulting in dysregulation of various signaling pathways.In this review,we focus on the major mechanisms of acquired resistance to second generation antiandrogens,including AR-dependent and AR-independent resistance mechanisms as well as other resistance mechanisms leading to CRPC emergence.Evolving knowledge of resistance mechanisms to AR targeted treatments will lead to additional research on designing more effective therapies for advanced-stage prostate cancer.
