Comparison of the efficacy and safety of once-daily dosing and on-demand use of udenafil for type 2 diabetic patients with erectile dysfunction

We compared the efficacy and safety between once-daily dosing and on-demand use of udenafil for type 2 diabetic patients with erectile dysfunction （ED）. A multi-center, randomized, open-label, parallel-group, 12-week study was conducted. 161 patients who improved with on-demand 200 mg of udenafil according to Sexual Encounter Profile （SEP） diary Question 2 and 3 （Q2 and Q3） were randomized into 200 mg on-demand （n = 80） or 50 mg once-daily （n = 81） dosing groups for 8 weeks. The dosing period was followed by a 4-week treatment-free period. The primary efficacy endpoint was the change of the International Index of Erectile Function （IIEF） erectile function domain （EFD） score. The secondary efficacy endpoints included changes to the SEP diary Q2, Q3, IIEF Q3, Q4, other domains of IIEF, Global Assessment Question, and shift to the normal rate （EFD 〉 26）. Vascular endothelial markers were also assessed. The IIEF-EFD score of both groups improved after 8 weeks of treatment （P 〈 0.0001）. There was no statistically significant difference between two groups. Improvement was not maintained after the treatment-free follow-up period. Similar results were observed in the secondary efficacy endpoints. There was also no significant difference in vascular endothelial markers. Daily udenafil was well-tolerated, and there was no significant difference in the adverse drug reactions and adverse events between the two groups. Flushing and headache were the most frequent adverse events. Both regimens improved ED in diabetic patients and were well-tolerated. Further studies are needed to assess the effect of daily udenafil treatment in diabetic patients.

Ezetimibe improves hepatic steatosis in relation to autophagy in obese and diabetic rats

AIM: To investigate whether ezetimibe ameliorates hepatic steatosis and induces autophagy in a rat model of obesity and type 2 diabetes. METHODS: Male age-matched lean control LETO and obese and diabetic OLETF rats were administered either PBS or ezetimibe(10 mg/kg per day) via stomach gavage for 20 wk. Changes in weight gain and energy intake were regularly monitored. Blood and liver tissue were harvested after overnight fasting at the end of study. Histological assessment was performed in liver tissue. The concentrations of glucose, insulin, triglycerides(TG), free fatty acids(FFA), and total cholesterol(TC) in blood and TG, FFA, and TG in livertissue were measured. m RNA and protein abundance involved in autophagy was analyzed in the liver. To investigate the effect of ezetimibe on autophagy and reduction in hepatic fat accumulation, human Huh7 hepatocytes were incubated with ezetimibe(10 μmol/L) together with or without palmitic acid(PA, 0.5 mmol/L, 24 h). Transmission electron microscopy(TEM) was employed to demonstrate effect of ezetimibe on autophagy formation. Autophagic flux was measured with bafilomycin A1, an inhibitor of autophagy and following immunoblotting for autophagy-related protein ***: In the OLETF rats that received ezetimibe(10 mg/kg per day), liver weight were significantly decreased by 20% compared to OLETF control rats without changes in food intake and body weight(P < 0.05). Lipid parameters including TG, FFA, and TC in liver tissue of ezetimibe-administrated OLETF rats were dramatically decreased at least by 30% compared to OLETF controls(P < 0.01). The serum glucose, insulin, HOMA-IR, and lipid profiles were also improved by ezetimibe(P < 0.05). In addition, autophagy-related m RNA expression including ATG5, ATG6, and ATG7 and the protein level of microtubule-associated protein light chain 3(LC3) were significantly increased in the liver in rats that received ezetimibe(P < 0.05). Are prostatic calculi independent predictive factors of lower urinary tract symptoms？

We determined the correlation between prostatic calculi and lower urinary tract symptoms （LUTS）,as well as the predisposing factors of prostatic calculi. Of the 1 527 patients who presented at our clinic for LUTS,802underwent complete evaluations,including transrectal ultrasonography,voided bladder-3 specimen and international prostatic symptoms score （IPSS）. A total of 335 patients with prostatic calculi and 467 patients without prostatic calculi were divided into calculi and no calculi groups,respectively. Predictive factors of severe LUTS and prostatic calculi were determined using uni/multivariate analysis. The overall IPSS score was 15.7±9.2 and 14.1±9.1 in the calculi and no calculi group,respectively （P = 0.013）. The maximum flow rate was 12.1±6.9 and 14.2±8.2 mL s^（-1） in the calculi and no calculi group,respectively （P = 0.003）. On univariate analysis for predicting factors of severe LUTS,differences on age （P = 0.042）,prostatic calculi （P = 0.048） and prostatitis （P = 0.018） were statistically significant. However,on multivariate analysis,no factor was significant. On multivariate analysis for predisposing factors of prostatic calculi,differences on age （P〈0.001） and prostate volume （P = 0.001） were significant. To our knowledge,patients who have prostatic calculi complain of more severe LUTS. However,prostatic calculi are not an independent predictive factor of severe LUTS. Therefore,men with prostatic calculi have more severe LUTS not only because of prostatic calculi but also because of age and other factors. In addition,old age and large prostate volume are independent predisposing factors for prostatic calculi.

Comparison of Protein Profiles in Sputum between COPD and Acute Exacerbation of COPD

Background and objective: Chronic obstructive pulmonary disease (COPD) is characterized by progressive airflow limitation that is associated with an abnormal inflammatory response of the lung to noxious particles or gases. Cigarette smoking is the major risk factor for the development of COPD. This study evaluated the levels of cyclophilin B in sputa from patients with COPD and COPD with acute exacerbation (AECOPD). Materials and Methods: Two-dimensional electrophoresis was used for differential display proteomics. Western blotting was used to identify and quantify cyclophilin B in sputum from subjects with AECOPD and COPD. Results: Forty-nine protein spots differed in relative intensity between the AECOPD (n = 6) and COPD (n = 6) subjects. Twenty proteins showed increased expression in the sputum of AECOPD subjects, and 29 proteins were present at lower levels in AECOPD sputum compared with COPD sputum. One of these proteins was associated with cyclophilin B. Cyclophilin B concentrations were lower in sputum from subjects with COPD (n = 4) versus AECOPD (n = 4). Conclusion: The sputum proteomic analysis suggests that changes in various proteins are associated with the development of AECOPD.

Use of a Digital Image Correlation Technique for Measuring the Material Properties of Beetle Wing

Beetle wings are very specialized flight organs consisting of the veins and *** it is necessary from a bionic view to investigate the material properties of a beetle wing *** the present study,we have used a Digital Image Correlation (DIC) technique to measure the elastic modulus of a beetle wing *** were prepared by carefully cutting a beetle hind wing into 3.0 mm by 7.0 mm segments (the gage length was 5 mm).We used a scanning electron microscope for a precise measurement of the thickness of the beetle wing *** specimen was attached to a designed fixture to induce a uniform displacement by means of a *** used an ARAMISTM system based on the digital image correlation technique to measure the corresponding displacement of a *** thickness of the beetle wing varied at different points of the *** elastic modulus differed in relation to the membrane arrangement showing a structural anisotropy;the elastic modulus in the chordwise direction is approximately 2.65 GPa,which is three times larger than the elastic modulus in the spanwise direction of 0.84 *** a result,the digital image correlation-based ARAMIS system was suc- cessfully used to measure the elastic modulus of a beetle *** addition to membrane's elastic modulus,we considered the Poisson's ratio of the membrane and measured the elastic modulus of a vein using an Instron universal tensile *** result reveals the Poisson's ratio is nearly zero and the elastic modulus of a vein is about 11 GPa.

Statistical Downscaling for Multi-Model Ensemble Prediction of Summer Monsoon Rainfall in the Asia-Pacific Region Using Geopotential Height Field

The 21-yr ensemble predictions of model precipitation and circulation in the East Asian and western North Pacific （Asia-Pacific） summer monsoon region （0°-50°N, 100° 150°E） were evaluated in nine different AGCM, used in the Asia-Pacific Economic Cooperation Climate Center （APCC） multi-model ensemble seasonal prediction system. The analysis indicates that the precipitation anomaly patterns of model ensemble predictions are substantially different from the observed counterparts in this region, but the summer monsoon circulations are reasonably predicted. For example, all models can well produce the interannual variability of the western North Pacific monsoon index （WNPMI） defined by 850 hPa winds, but they failed to predict the relationship between WNPMI and precipitation anomalies. The interannual variability of the 500 hPa geopotential height （GPH） can be well predicted by the models in contrast to precipitation anomalies. On the basis of such model performances and the relationship between the interannual variations of 500 hPa GPH and precipitation anomalies, we developed a statistical scheme used to downscale the summer monsoon precipitation anomaly on the basis of EOF and singular value decomposition （SVD）. In this scheme, the three leading EOF modes of 500 hPa GPH anomaly fields predicted by the models are firstly corrected by the linear regression between the principal components in each model and observation, respectively. Then, the corrected model GPH is chosen as the predictor to downscale the precipitation anomaly field, which is assembled by the forecasted expansion coefficients of model 500 hPa GPH and the three leading SVD modes of observed precipitation anomaly corresponding to the prediction of model 500 hPa GPH during a 19-year training period. The cross-validated forecasts suggest that this downscaling scheme may have a potential to improve the forecast skill of the precipitation anomaly in the South China Sea, western North Pacific and the East Asia

Graphene electrode with tunable charge transport in thin-film transistors

Graphene, a single atomic layer of sp2-hybridized carbon, has immense potential as a transparent conducting material in electronic applications owing to its superior properties, including optical transparency and high conductivity. Particularly, the tunable work function of graphene enables the integration of graphene electrodes with various electronic devices. To achieve high performance in graphene-based devices, effective charge transport between the graphene electrode and the semiconducting material needs to be optimized; this is closely related to the modulation of the Schottky barrier （SB）. In this study, we investigate the ~nable charge transport properties as a function of graphene doping in n-channel thin-film transistors （TFTs） in terms of the electrical characteristics and low-frequency noise （LFN） behaviors. Alkali metal carbonates tuned the work function of graphene, resulting in a dramatic decrease in the SB and an improvement of the carrier injection in n-channel TFTs. The electrical performance of the TFTs was evaluated by extraction of the field-effect mobilities and ratio of contact resistance to total resistance. Furthermore, the level of contact noise created by the barrier height fluctuation and relative contribution of channel noise and contact noise in the TFTs was investigated by LFN measurements to demonstrate the ~nable charge transport. Our findings therefore provide new insights into the tunable charge transport mechanism in graphene-based devices and reveal the immense potential of graphene as electrodes in high performance flexible and transparent displays.

Impacts of energy feeds and supplemental protease on growth performance, nutrient digestibility, and gut health of pigs from 18 to 45 kg body weight

A total of 144 pigs with 18.4 ± 2.3 kg initial body weight(BW) at 6 wk of age were used in a 40-d trial to evaluate effects of protease(300,000 U/kg feed, BioResource International Inc., Durham, NC, USA) on growth performance, apparent ileal digestibility(AID) of nutrients, and gut health of pigs fed diets with sorghum. Pigs were randomly allotted to 4 treatments(12 pens per treatment, 3 pigs per pen) in a 2 x 2 factorial arrangement(corn or sorghum basal diets, and 0 or 0.05% protease as 2 factors) with sex and initial BW as blocks. Experimental period had phase 1(d 1 to 21) and phase 2(d 22 to 40). About 65%(phase 1) and 72%(phase 2) of cereal grains were used in corn or sorghum based diets. Both grains were ground to 400 μm. Body weight and feed intake were recorded weekly. On d 35, serum was collected to quantify tumor necrosis factor-a(TNF-a) and malondialdehyde(MDA). Titanium dioxide(0.3%) was added as an indigestible marker for an additional 4 d feeding. On d 40, 32 pigs(8 pigs per treatment)were euthanized to collect digesta from jejunum and ileum(for viscosity and AID), tissues(for morphology) and mucosa samples(for TNF-α and MDA) from duodenum, jejunum, and ileum. Replacing corn with sorghum in the diet increased(P < 0.05) overall average daily gain(from 756 to 787 g/day) and average daily feed intake(from 1,374 to 1,473 g/day), reduced(P < 0.05) overall gain:feed ratio(from0.553 to 0.537), and did not affect AID. Pigs fed diets with sorghum had lower(P < 0.05) MDA content in serum(from 14.61 to 6.48 μmol/L) and jejunum(from 1.42 to 0.91 μmol/g protein), and reduced(P < 0.05) villus height(from 492 to 396 μm) and crypt depth(from 310 to 257 μm) in jejunum. Dietary protease improved(P < 0.05) AID of crude protein(from 81.8% to 86.0%), decreased MDA level(from 1.20 to 0.98 μmol/g protein) in duodenum, and increased(P < 0.05) the ratio of villus height to crypt depth(from 1.08 to 1.21) in duodenum.
