Temperature is an important environmental cue that affects flowering time in *** MADS-box transcription factor FLOWERING LOCUS M(FLM)forms a heterodimeric complex with SHORT VEGETATIVE PHASE(SVP)and controls ambient temperature-responsive flowering in ***-βand FLM-δ,two major splice variants produced from the FLM locus,exert opposite effects on flowering,but the molecular mechanism by which the interaction between FLM isoforms and SVP affects temperature-responsive flowering remains poorly ***,we show that FLM-βand FLM-δplay important roles in modulating the temperature-dependent behavior,conformation,and stability of *** localization of SVP decreases as temperature ***-βis required for SVP nuclear translocation at low temperature,whereas SVP interacts with FLM-δmainly in the cytoplasm at high *** preferentially binds to FLM-βat low temperature in tobacco leaf *** shows high binding affinity to FLM-βat low temperature and to FLM-δat high *** undergoes similar structural changes in the interactions with FLM-βand FLM-δ;however,FLM-δlikely causes more pronounced conformational changes in the SVP ***-δcauses rapid degradation of SVP at high temperature,compared with FLM-β,possibly via *** of lysine 53 or lysine 165 in SVP causes increased abundance of SVP due to reduced ubiquitination of SVP and thus delays flowering at high *** findings suggest that temperature-dependent differential interactions between SVP and FLM isoforms modulate the temperature-responsive induction of flowering in Arabidopsis.

Atomic layer deposition （ALD） has become an essential deposition method for forming nanometer scale thin films in the microelectronics industry, and its applications have been extended to multi-component thin films, as well as to single metal oxide films. In order to investigate the development of the surface structure of ultra-thin film qualitatively as well as quantitatively, ALD processes are simulated with a molecular scale. For this simulation, the film materials are deposited on a imaginary substrate that consists of small lattice. The deposition behaviors are described by using random deposition （RD） model or random deposition with surface relaxation （RDSR） model as the ALD growth mode, and the proposed model was applied to the deposition of SrO-TiO2 thin films. Through this work, growth characteristics such as surface morphology, deposited film coverage can be predicted.

Relationship between intraocular pressure and parameters of obesity in ocular hypertension

AIM:To evaluate the correlation between intraocular pressure(IOP)and various obesity-related health factors in patients with ocular hypertension in ***:A total of 40850 subjects underwent age,sex,body weight,and height assessments and automated multiphasic tests,including non-contact tonometry,automated perimetry,fundus photography,systolic/diastolic blood pressure measurement,and evaluation of obesity-related health parameters such as obesity index,body mass index(BMI),a body shape index(ABSI),and waist-to-height ratio(WtHR).Subjects were divided into ocular hypertension group and normal IOP group according to IOP after matching of age and ***:Of 40850 participants,1515(3.7%)had ocular hypertension,and 1515 with normal IOP were selected as controls using propensity score *** mean IOP of control group was 15.3±2.3 mm Hg,compared with 23.3±1.6 mm Hg in ocular hypertension ***,obesity index,BMI,and WtHR in the ocular hypertension group were significantly higher than in the normal IOP group(P<0.001,P<0.001,P=0.009,P=0.002).IOP of ocular hypertension was positively correlated with obesity index(P=0.027)and BMI(P=0.016),whereas IOP of control was positively correlated with blood pressure(P<0.001,P=0.002),obesity index(P<0.001),BMI(P<0.001),and WHtR(P=0.002).Systolic blood pressure(β=0.022,P<0.001)and body weight(β=0.016,P=0.02)were precursors of IOP in normal subjects,but sex(male;β=-0.231,P=0.008)and obesity index(β=-0.007,P=0.017)were precursors of ocular hypertension according to multiple regression ***:Among various obesity-related health parameters,obesity index is the best indicator for further increase in IOP in ocular hypertension group.

The simultaneous measurement of the spatial profile and spectrum of laser-accelerated protons is important for further optimization of the beam qualities and *** report a detailed study regarding the underlying physics and regular procedure of such a measurement through the radioactivation of a stack composed of aluminum,copper,and CR-39 plates as well as radiochromic films(RCFs).After being radioactivated,the copper plates are placed on imaging plates(IPs)to detect the positrons emitted by the reaction products through contact *** spectrum and energy-dependent spatial profile of the protons are then obtained from the IPs and confirmed by the measured ones from the RCFs and CR-39 *** also discuss the detection range,influence of electrons,radiation safety,and spatial resolution of this ***,insights regarding the extension of the current method to online measurements and dynamic proton imaging are also provided.

The use of natural killer(NK)cells is a promising and safe immunotherapeutic approach in the field of cancer ***,combination treatments are required to enhance the effector functions and therapeutic efficacy of NK *** this study,we investigated the potential of daratumumab(Dara),bortezomib,and dexamethasone(Dvd)to augment the antitumor effects of NK cells in a multiple myeloma(MM)xenograft mouse *** cells were expanded and activated using the K562-OX40 ligand and membrane-bound IL-18 and IL-21 in the presence of IL-2 and IL-15 from peripheral blood mononuclear cells from MM patients.A human MM xenograft model was established using human RPMI8226-RFP-FLuc cells in NOD/SCID IL-2Rγnull(NSG)***-bearing mice were divided into six treatment groups:no treatment,expanded NK cells(eNKs),Dara,Dara+eNKs,Dvd,and Dvd+*** treatment strongly enhanced the cytotoxicity of eNKs by upregulating expression of NK cell activation ligands,downregulating expression of NK cell inhibitory ligands,and promoting antibody-dependent cellular *** combination of eNKs with Dvd significantly prolonged mouse survival and reduced the tumor burden and serum M-protein ***,Dvd pretreatment significantly increased eNK persistence and homing to MM *** findings suggest that Dvd treatment potentiates the antimyeloma effects of NK cells expanded and activated ex vivo by modulating immune responses in MM-bearing mice.

Nanovaccines are used as delivery platforms for antigens and adjuvants, which activate antigen-presenting cells (APCs) and enhance anticancer immune responses.1,2 Researchers have recently developed a self-assembled nanocomplex using a polysorbitol-co-PEI (PSPEI) polymer complexed with poly(I:C) (PIC). By binding to different surface proteins, this nanocomplex enhances the intracellular delivery of cargos and induces potent anticancer immune responses against melanoma cells.3 We have previously demonstrated that PD-1/PD-L1 blockade enhanced the efficacy of DC-based cancer immunotherapy.4,5 Moreover, the combination of nanomedicines and PD-L1 blockade has been reported to enhance CD8+ T cell activation and inhibit immunosuppressive cells within the tumor microenvironment.6 In the present study, we investigated the therapeutic efficacy of a combinatorial treatment comprising the immunoadjuvant nanocomplex PSPEI-PIC, a DC vaccine, and PD-L1 blockade in a murine colon cancer model.

Hepatitis B surface antigen (HBsAg) is produced and secreted through a complex mechanism that is still not fully understood. In clinical fields, HBsAg has long served as a qualitative diagnostic marker for hepatitis B virus infection. Notably, advances have been made in the development of quantitative HBsAg assays, which have allowed viral replication monitoring, and there is an opportunity to make maximal use of quantitative HBsAg to elucidate its role in clinical fields. Yet, it needs to be underscored that a further understanding of HBsAg, not only from clinical point of view but also from a virologic point of view, would enable us to deepen our insights, so that we could more widely expand and apply its utility. It is also important to be familiar with HBsAg variants and their clinical consequences in terms of immune escape mutants, issues resulting from overlap with corresponding mutation in the P gene, and detection problems for the HBsAg variants. In this article, we review current concepts and issues on the quantification of HBsAg titers with respect to their biologic nature, method principles, and clinically relevant topics.

Beta-nerve growth factor(β-NGF) is known to be a major leading cause of neuronal plasticity. To identify the possible action mechanisms of β-NGF gene therapy for sciatic nerve recovery, experimental dogs were randomly divided into control, pyridoxine, and pyridoxine + β-NGF groups. We observed chronological changes of morphology in the dorsal root ganglia in response to pyridoxine toxicity based on cresyl violet staining. The number of large neurons positive for cresyl violet was dramatically decreased after pyridoxine intoxication for 7 days in the dorsal root ganglia and the neuron number was gradually increased after pyridoxine withdrawal. In addition, we also investigated the effects of β-NGF gene therapy on neuronal plasticity in pyridoxine-induced neuropathic dogs. To accomplish this, tyrosine kinase receptor A(TrkA), βIII-tubulin and doublecortin(DCX) immunohistochemical staining was performed at 3 days after the last pyridoxine treatment. TrkA-immunoreactive neurons were dramatically decreased in the pyridoxine group compared to the control group, but strong TrkA immunoreactivity was observed in the small-sized dorsal root ganglia in this group. TrkA immunoreactivity in the dorsal root ganglia was similar between β-NGF and control groups. The numbers of βIII-tubulin-and DCX-immunoreactive cells decreased significantly in the pyridoxine group compared to the control group. However, the reduction of βIII-tubulin-and DCX-immunoreactive cells in the dorsal root ganglia in the β-NGF group was significantly ameliorated than that in the pyridoxine group. These results indicate that β-NGF gene therapy is a powerful treatment of pyridoxine-induced neuropathic damage by increasing the TrkA and DCX levels in the dorsal root ganglia. The experimental protocol was approved by the Institutional Animal Care and Use Committee(IACUC) of Seoul National University, South Korea(approval No. SNU-060623-1, SNU-091009-1) on June 23, 2006 and October 9, 2009, respectively.

Sojucktang (SJT) has long been used for the treatment of endometrial diseases in Korea. However, the mechanisms responsible for the SJT-induced apoptosis in endometrial cancer cells remain unclear. In the present study, SJT was demonstrated to show cytotoxic effect and induce apoptotic cell death via mitochondrial regulation in KLE endometrial cancer cells. Linderae Radix, Glycyrrhizae Radix, Zedoariae Rhizoma, Trogopterorum Faeces and Agelicae Gigantis Radix were found to be the potent constituent herbs of SJT to significantly decrease the viability of KLE cells by a tetra zolium salt (XTT) assay. Apoptotic bodies were observed in SJT-treated KLE cells by 4′-6-diamidino-2-phenylindole (DAPI) and TdT-mediated-dUTP nick-end labeling (TUNEL) assay. SJT also increased sub-G1 DNA contents of the cell cycle undergoing apoptosis in a dose-dependent manner. Furthermore, it was observed that SJT activated caspase-3 and cleaved poly (ADP-ribose) polymerase (PARP), and decreased mitochondrial membrane potential in a dose-dependent manner. Taken together, this study shows that SJT exerts anti-tumor activity against KLE endometrial cancer cells via mitochondrial dependent apoptosis induction.

Perforated duodenal ulcer presenting with massive hematochezia in a 30-month-old child

Peptic ulcer disease is uncommon in children and rarely suspected as a cause of abdominal complaints in this age group; the diagnosis is therefore made almost exclusively when complications develop. Peptic ulcer disease is usually not considered in the differential diagnosis of pediatric patients. We present the case of a 30-month-old boy with duodenal perforation due to a peptic ulcer without a known etiology. The patient was admitted through the emergency department due to severe hematochezia and ongoing anemia; he presented with neither abdominal pain nor abdominal distension. There were no medical problems, and no drugs, such as corticosteroids or nonsteroidal anti-inflammatory drugs, had been prescribed or administered recently. We tried to control the active bleeding by medical treatment including arterial embolization, but the active bleeding was not controlled. Finally, an exploratory laparotomy was performed. A discrete anterior perforation with active bleeding of the duodenal wall was found. After the operation, there were no complications and the patient recovered fully.
