Effects of ethanol on the tonicity of corporal tissue and the intracellular Ca^2＋ concentration of human corporal smooth muscle cells

Heavy alcohol consumption is associated with an increased risk of erectile dysfunction （ED）; however, the acute effects of ethanol （EtOH） on penile tissue are not fully understood. We sought to investigate the effects of EtOH on corporal tissue tonicity, as well as the intracellular Ca^2＋ concentration （[Ca^2＋]i） and potassium channel activity of corporal smooth muscle. Strips of corpus cavernosum （CC） from rabbits were mounted in organ baths for isometric tension studies. Electrical field stimulation （EFS） was applied to strips precontracted with 10 μmol L^-1 phenylephrine as a control. EtOH was then added to the organ bath and incubated before EFS. The [Ca^2＋]i levels were monitored by the ratio of fura-2 fluorescence intensities using the fura-2 loading method. Single-channel and whole-cell currents were recorded by the conventional patch-clamp technique in short-term cultured smooth muscle cells from human CC tissue. The corpus cavernosal relaxant response of EFS was decreased in proportion to the concentration of EtOH. EtOH induced a sustained increase in [Ca^2＋]i in a dose-dependent manner, Extracellular application of EtOH significantly increased whole-cell K^＋ currents in a concentration-dependent manner （P 〈 0.05）. EtOH also increased the open probability in cell-attached patches; however, in inside-out patches, the application of EtOH to the intracellular aspect of the patches induced slight inhibition of Ca^2＋-activated potassium channel （KCa） activity. EtOH caused a dose-dependent increase in cavemosal tension by alterations to [Ca^2＋]i. Although EtOH did not affect KCa channels directly, it increased the channel activity by increasing [Ca^2＋]i. The increased corpus cavemosal tone caused by EtOH might be one of the mechanisms of ED after heavy drinking.

In this study, we investigated the effects of a combination of Ginkgo biloba extracts （GBE） and phosphodiesterase type 5 （PDE-5） inhibitors on the muscular tone of the corpus cavernosum and potassium channel activity of corporal smooth muscle cells. Strips of corpus cavernosum from male New Zealand white rabbits were mounted in organ baths for isometric tension studies. After contraction with 1 × 10^-5 mol I^-1 norepinephrine, GBE （0.01-1 mg ml^-1） and mirodenafil （0.01-100 nmol I^-1） were added together into the organ bath. In electrophysiological studies, whole-cell currents were recorded by the conventional patch-clamp technique in cultured smooth muscle cells of the human corpus cavernosum. The corpus cavemosum was relaxed in response to GBE in a dose-dependent manner （from 0.64%±8.35% at 0.01 mg ml^-1 to 52.28%±11.42% at 1 mg ml^-1）. After pre-treatment with 0.03 mg ml^-1 of GBE, the relaxant effects of mirodenafil were increased at all concentrations, After tetraethylammonium （TEA）（1 mmol I^-1） administration, the increased effects were inhibited （P〈0.01）. Extracellular administration of GBE increased the whole-cell K^＋ outward currents in a dose-dependent fashion. The increase of the outward current was inhibited by I mmol 1-1 TEA. These results suggest that GBE could increase the relaxant potency of mirodenafil even at a minimally effective dose. The K＋ flow through potassium channels might be one of the mechanisms involved in this synergistic relaxation.

^(18)F-fluoro-2-deoxyglucose uptake on PET CT and glucose transporter 1 expression in colorectal adenocarcinoma

AIM:To evaluate the correlation between the level of 18 F-fluoro-2-deoxyglucose (18 F-FDG) uptake and glucose transporter 1 (GLUT1) expression in colorectal adenocarcinoma (CRA).METHODS:Forty four patients with resected CRA and preoperative 18 F-FDG positron emission tomography computed tomography data were investigated in this *** of maximum standardized uptake value (SUVmax) of the lesion was made with GLUT1 expression by immunohistochemistry and various clinicopathologic factors including tumor volume,invasion depth,gross finding,and lymph node ***:SUVmax was 14.45 ± 7.0 in negative GLUT1 expression cases,15.51 ± 5.7 in weak GLUT1 expression cases,and 16.52 ± 6.8 in strong GLUT1 expression cases,and there was no correlation between between GLUT1 expression and *** was significantly correlated with tumor volume (P < 0.001).However,there was no significant differences in SUVmax and GLUT1 expression among other clinicopathologic ***:GLUT1 expression does not correlates significantly with 18 F-FDG uptake in CRA.18 F-FDG uptake was increased with tumor volume,which is statistically significant.

A granular cell tumor (GCT) is a benign neoplasm of unclear histogenesis that is generally believed to be of nerve sheath *** is not common and most often affects the tongue,skin and soft tissue,although it may occur anywhere in the *** experienced a case of GCT that arose in the cecum of a 55-yearold *** GCT was removed by laparoscopic *** addition to the tumor,endoscopic examination revealed the presence of a 5-mm-polyp in the descending colon and multiple tiny polyps in the sigmoid colon and *** examination demonstrated a cecal tumor 1.5 cm × 1.0 cm × 0.7 cm with a hard consistency;in cut sections,mixed cells with yellowish and whitish portions were *** tumor was located between the mucosa and subserosa,and was composed of plump histiocyte-like tumor cells with abundant granular eosinophilic cytoplasm,which were immunoreactive for S-100 protein,vimentin,neuron-specific enolase,inhibin-α and *** tumor showed extensive hyalinization and focal dystrophic *** profiles did not confirm any particular cell type for the histogenetic origin of the GCT,including a nerve sheath *** hyalinization and calcifi cation showing involution of tumor cells suggest benign clinical behavior of GCT.

UBVVI Surface Photometry of the Spiral Galaxy NGC 300 in the Sculptor Group

We present UBVI surface photometry over a 20.5' × 20.5' area of the late-type spiral galaxy NGC 300. We have derived isophotal maps, surface brightness profiles, ellipticity profiles, position angle profiles, and color profiles. By merging our I-band measurements with those of Boker et al. based on Hubble Space Telescope observations, we have obtained combined I-band surface brightness profiles for the region 0.02' < r < 500' and have decomposed the profiles into three components: a nucleus, a bulge, and an exponential disk.

In this paper we show a law of iterated logarithm for rescaled range statistics Q（n） for AR（1） model.

Dynamic simulation of universal spacer in Dynesys dynamic stabilization system for human vertebra

The aim of this study is to analyze the simulated behavior of universal spacer in Dynesys dynamic stabilization system inserted in human vertebra. Dynesys, so-called "Dynamic neutralization system for the spine", dynamic stabilization system is a new concept in the surgical treatment of lower back pain recently. Universal spacer used as flexible material is to stabilize the spine and the material property of universal spacer is polycarbonate urethane. Universal spacer may apply different kinematic behaviors at implanted level in vertebra. Spinal range of motion(SROM) of inter-vertebra with installed Dynesys dynamic stabilization system was studied using Adams+LifeMOD as simulation software package. The vertebra model was set up to closely resemble the in-vivo conditions. Inter-vertebra rotations were measured by post processor of Adams and compared with the intact values. SROMs of the flexion, extension, lateral bending, and axial rotation of human virtual models were measured, where three spinal fixation systems such as rigid system, Dynesys system, and fused system were installed. As a result, the value of SROM is decreased in flexion-extension and lateral bending when the spinal fixation system is implanted. The movement of Dynesys system is similar to that of intact model by allowing the movement of lumbar. This means that the Dynesys system is proved to be safe and effective in the treatment of unstable spinal condition.

Fatigue crack in large-scale tubular joints for offshore structures

Tubular members have been used for constructing offshore structures. Large-scale fatigue tests of welded tubular K-joints under the balanced in-plane bending brace were carried out to investigate the fatigue behavior of API 2W Gr.50 steel produced by POSCO. The experimental results were verified by numerical approaches and compared with the IIW, DnV RP-C203 and API RP 2A-WSD design curves. The test results based on the hot spot stress were in agreement with the design curves. The SCF factor for tubular K-joint was also obtained. The numerical parametric study of the K-joint (using the finite element program) was described and its results were compared with experimental results. The stress effects of various parameters including α, β, γ, τ and θ on the stress in the K-joint were investigated. The stress distribution for each parameter was introduced. The study showed that the maximum stress of the joint varied according to the variation of joint parameters.

Anti-integrin therapy for inflammatory bowel disease

In inflammatory bowel disease(IBD), tumor necro-sis factor plays an important role in mediating infla-mmation, but several other pathways are also involved in eliciting an inflammatory response. One such pathway is the invasion of the intestinal mucosa by leukocytes. Leukocytes within the systemic circulation move to sites of inflammation, and blocking this pathway could be an important treatment strategy for IBD. Anti-integrin therapy blocks the action of integrin on the surface of circulating immune cells and endothelial cell adhesion molecules, thereby inhibiting the interactions between leukocytes and intestinal blood vessels. Natalizumab, which acts on α4-integrin, was the first such drug to be approved for Crohn's disease, but its use is limited due to the risk of progressive multifocal leukoencephalopathy. Vedolizumab produces few systemic adverse effects because it acts on gut-trophic α4β7 integrin, and has been approved and is being used to treat IBD. Currently, several anti-integrin drugs, including etrolizumab, which acts on β7-integrin, and PF-00547569, which targets mucosal addressin cell adhesion molecule-1, are undergoing clinical trials and the results are being closely watched.

Hyperinsulinemic hypoglycemia due to diffuse nesidioblastosis in adults:A case report

Persistent hyperinsulinemic hypoglycemia is caused most commonly by an insulinoma in adults or by nesidioblastosis in neonates. In adults, nesidioblastosis is a rare disorder characterized by diffuse or disseminated proliferation of islet cells. We recently encountered a case of nesidioblastosis in an adult. A 71-year-old man was admitted due to intermittent general weakness, abdominal pain, and mild dyspnea. The patient underwent a subtotal gastrectomy for a gastric adenocarcinoma two years ago. After 5 d of admission, the patient showed symptoms of cold sweating, chilling, and hypotension 30 min after eating. Thereafter, he frequently showed similar symptoms accounting for hypoglycemia regardless of food consumption. Laboratory findings revealed a low fasting blood glucose level （25 mg/dL）, and a high insulin level （47 μIU/mL）. Selective intra-arterial calcium stimulation with hepatic venous sampling （ASVS） was performed to localize a mass and revealed an increased insulin level about fourfold that of the normal fasting level at 60 s in the splenic artery, which suggested the presence of an insulinoma in the tail of pancreas. A distal pancreatectomy was performed. Neither intraoperative exploration nor a frozen biopsy specimen detected any mass-forming lesion. On the histological examination, many of the islets were enlarged and irregularly shaped in all specimens, the arrangement of which was a Iobulated islet pattern. Cytologically, a considerable subpopulation of endocrine cells showed enlarged and hyperchromatic nuclei. By immunohistochemistry, the cells were identified as p-cells. These clinical, radiological, microscopic and immuno-histochemical findings are consistent with diffuse nesidioblastosis in adults.
