Ovarian endometrioma（OE）, also known as “chocolate cysts,” is a cystic mass that develops in the ovaries due to endometriosis and is a common gynecological condition characterized by the growth of endometrial tissue outside the uterus, leading to symptoms such as dysmenorrhea, pelvic pain, and infertility. However, the precise molecular and cellular mechanisms driving this pathophysiology remain largely unknown, posing challenges for diagnosis and treatment. Here, we employed integrated single-cell transcriptomic profiling of over 52,000 individual cells from endometrial tissues of OE patients and healthy donors and identified twelve major cell populations. We identified notable alterations in cell type-specific proportions and molecular signatures associated with OE. Notably, the activation of IGFBP5+macrophages with pro-inflammatory properties, NK cell exhaustion, and aberrant proliferation of IQCG+and KLF2+epithelium are key features and may be the potential mechanisms underlying the pathogenesis of OE. Collectively, our data contribute to a better understanding of OE at the single cell level and may pave the way for the development of novel therapeutic strategies.

Surveillance recommendations for gastric cancer（GC） in current guidelines focused on advanced precancerous lesions and were based on precise diagnosis of severity/extent of baseline lesions. We aimed to develop a less endoscopy-related equipment-dependent risk-stratification tool, and assessed whether mild-precursor-lesion patients can be safely exempt from surveillance. In the multicenter communitybased cohort, 75,051 participants receiving baseline endoscopy were enrolled during 2015–2017 and followed-up until 2021. Cumulative incidence rates（CIRs） of GC for precancerous-conditions were calculated by Kaplan-Meier method and compared by Log-rank tests. Mixedeffects Cox regression models were used to detect potential factors for progression towards GC. A risk score was calculated as counts of selected factors. An independent cohort, including 26,586 participants was used for external validation. During a median follow-up of 6.25 years, CIRs of GC were 0.302%, 0.436%, and 4.756% for normal group, non-neoplastic（atrophic gastritis/intestinal metaplasia） and neoplastic lesions（low-grade/high-grade dysplasia）, respectively(Ptrendur predictors, including male, ≥60 years, smoking, and limited vegetable consumption, were selected for risk-stratification. High-risk patients（≥3 risk factors） with non-neoplastic lesions showed higher GC risks（adjusted HR=7.73, 95%CI: 4.29–13.92）, and their four-year CIR reached the one-year CIR of neoplastic lesions. Further categorizing non-neoplastic lesions by histological grade, both patients with moderate-to-severe lesions（aHR=3.07, 95%CI: 1.67–5.64） and high-risk patients with mild lesions（aHR=7.29, 95%CI: 3.58–14.86） showed higher risks. Consistent trends were observed in validation cohort. High-risk mild-precursor-lesion patients should receive surveillance within 3–5 years after baseline screening. Our study provides evidence on supplementing current guideline recommendations.

A lightweight road-assistant detection algorithm， EBD-YOLO， based on YOLOv5s is proposed to address the problems of high model complexity， computation cost， and difficulty in deployment on resource-limited embedded terminals in existing assisted driving detection algorithms. First， lightweight Transformer model EfficientViT was used as the backbone feature extraction network of YOLOv5s model to reduce network parameters and calculation costs. Secondly， a Focal-GIou Loss function is proposed for bounding box regression to accelerate model convergence and reduce loss. Thirdly， the feature pyramid structure is improved to a weighted bi-directional feature pyramid network (BiFPN) to enhance localization and semantic features. Then， a dynamic head framework is added to unify the attention mechanism with the object detection head to improve its performance. Finally， a Soft-CIou＿NMS algorithm is proposed in the post-processing stage to enhance occluded targets' localization and detection ability and reduce the missed detection rate. We conducted experiments on the KITTI and BDD100K datasets for autonomous driving， and the results showed that the EBD-YOLO model reduced in size by 38.4% and 37.2%， respectively. In comparison， the computational cost was reduced by 48.1%. As measured by mAP@0.5， the detection accuracy improved by 0.5% and 5.8%， respectively， and mAP@0.5:0.95 improved by 2.8% and 7%， respectively. These improvements satisfied the requirements for deployment on embedded terminals in cars.

Wetlands play a critical role in the global environment. The Middle Yangtze River Basin(MYRB), known for its abundant wetland resources, has experienced notable changes resulting from the complex interplay of environmental factors. Therefore, we investigated the spatiotemporal characteristics of wetland ecological quality in the MYRB from 2001 to *** the random forest(RF) regression algorithm and patch-generated land-use simulation(PLuS) model, we forecasted variations in wetland habitat quality and their determinants under the Shared Socioeconomic Pathway-Representative Concentration Pathway(SSPRCP) framework from 2035 to 2095. The main findings are as follows:(1) The RF algorithm was optimal for land-use and land-cover(LuLC) classification in the MYRB from 2001 to 2020,when notable changes were observed in water bodies and buildings. However, the forested area exhibited an increase and decrease of 3.9% and 1.2% under the SSP1-2.6 and SSP5-8.5 scenarios, respectively, whereas farmland exhibited a diminishing trend.(2) Wetlands were primarily concentrated in the central and eastern MYRB, with counties in the southwest exhibiting superior ecological-environmental quality from 2001 to 2020. Notably,wetland coverage revealed significantly high level, significant changes, frequent but relatively minor changes under the SSP1-2.6, SSP2-4.5, and SSP 5-8.5 scenarios, respectively.(3)Regions with lower habitat quality were primarily concentrated in urbanized areas characterized by frequent human activities, indicating a clear degradation in habitat quality across different scenarios. In conclusion, we established a foundational framework for future investigations into the eco-hydrological processes and ecosystem quality of watersheds.

Atherosclerosis(AS) is a prevalent condition associated with cardiovascular and cerebrovascular diseases, featuring lipid-laden plaque; however, the insufficient resolution and low sensitivity of traditional medical imaging techniques limit their application in lipid imaging. Currently, the second near-infrared(NIR-II) fluorescence imaging offers promising sensitivity and convenience. However, existing probes have limitations in terms of signal-to-background ratio, penetration depth, and tissue scattering. Herein, we use a molecular regulatory strategy to design a lipid-activatable probe Rh-965 with high specificity and NIR-II emission for improved imaging potential in deep tissues. The potential of Rh-965 for imaging plaque lipids in mice with AS is demonstrated by localizing carotid artery plaques using NIR-II imaging. The development of more stable lipid-activated NIR-II probes can significantly enhance atherosclerosis diagnosis and facilitate in vivo imaging in deeper tissues.
