Pathological bone loss is caused by an imbalance between bone formation and *** bone microenvironments are hypoxic,and hypoxia-inducible factor (HIF) is known to play notable roles in bone ***,the relevant functions of HIF-2α are not well ***,we have shown that HIF-2α deficiency in mice enhances bone mass through its effects on the differentiation of osteoblasts and *** vitro analyses revealed that HIF-2α inhibits osteoblast differentiation by targeting Twist2 and stimulates RANKL-induced osteoclastogenesis via regulation of *** addition,HIF-2α appears to contribute to the crosstalk between osteoblasts and osteoclasts by directly targeting RANKL in osteoprogenitor *** performed with osteoblast- and osteoclast-specific conditional knockout mice supported a role of HIF-2α in this ***-2α deficiency alleviated ovariectomy-induced bone loss in mice,and specific inhibition of HIF-2α with ZINC04179524 significantly blocked RANKLmediated ***,our results suggest that HIF-2α functions as a catabolic regulator in bone remodeling,which is critical for the maintenance of bone homeostasis.

Demand for ammonia continues to increase to sustain the growing global *** direct electrochemical N2 reduction reaction(NRR)powered by renewable electricity offers a promising carbon-neutral and sustainable strategy for manufacturing NH3,yet achieving this remains a grand ***,we report a synergistic strategy to promote ambient NRR for ammonia production by tuning the Te vacancies(VTe)and surface hydrophobicity of two-dimensional TaTe_(2)*** NH3 faradic efficiency of up to 32.2%is attained at a mild overpotential,which is largely maintained even after 100 h of consecutive *** labeling validates that the N atoms of formed NH4+originate from *** situ X-ray diffraction indicates preservation of the crystalline structure of TaTe_(2)during *** density functional theory calculations reveal that the potential-determining step(PDS)is*NH_(2)+(H^(+)+e^(-))/NH3 on VTe-TaTe_(2)compared with that of*+N2+(H^(+)+e^(-))/*N-NH on TaTe_(2).We identify that the edge plane of TaTe_(2)and VTe serve as the main active sites for *** free energy change at PDS on VTe-TaTe_(2)is comparable with the values at the top of the NRR volcano plots on various transition metal surfaces.

AIM:To evaluate and compare the clinical outcomes of prophylactic submucosal saline-epinephrine injection and saline injection alone for large colon polyps by conventional polypectomy. METHODS:A prospective study was conducted from July 2003 to July 2004 at 11 tertiary endoscopic centers. Large colon polyps (> 10 mm in diameter) wererandomized to undergo endoscopic polypectomy with submucosal saline-epinephrine injection (epinephrine group) or normal saline injection (saline group). Endoscopic polypectomy was performed by the conventional snare method,and early ( 2 cm) and neoplastic polyps were independently and significantly associated with the presence of PPB. CONCLUSION:The prophylactic submucosal injection of diluted epinephrine does not appear to provide an additional advantage over the saline injection alone for the prevention of PPB.

Human gut microbiota,containing at least 100 trillion bacteria,resides in the mucosal surface of the human *** individual is host to a distinct set of at least 160 species in the *** collective microbial genome encodes 500 times more genes than the human genome,and so it is tempting to consider human genes as noise in the storm of microbial signals[1].Recent data suggest that,while microbial signals modulate crucial functions of the healthy human body,there are also accumulating data suggesting that many human diseases have their origin in distorted gut microbiota composition[2].

Cognitive development theories differ on how young students can meaningfully process new information and retain that information for future knowledge-building through scaffolding within their zone of proximal *** traditional theories like the cognitive load theory adhere to the rote memorization approach by categorizing students as passive learners and the teachers as initiators who provide information in a structured,often rigid format,to be stored and retrieved for future application using their working *** contrast,the more progressive theories,like constructivism,are premised on the belief that students should proactively initiate their own learning while teachers act more as *** current trend in government policy under ESSA is to embrace the latter approach in the classroom,which is also more inclusive of all types of students,especially neurodiverse ***,teachers can utilize the wider range of assistive technology tools to accommodate and support their students’unique learning styles.

本研究旨在克隆延边牛脂肪分化相关蛋白PLIN2基因完整CDS区,通过生物信息学分析PLIN2基因CDS区序列和蛋白质基本特性,探讨其在延边牛不同组织及前体脂肪细胞成脂过程中的表达规律。选取18月龄去势的延边牛为研究对象,利用RT-PCR和基因克隆获得延边牛PLIN2基因,与其他物种进行同源性比对及系统进化树构建,利用生物信息学方法预测PLIN2编码蛋白质的理化性质、潜在磷酸化位点及糖基化位点、二硫键分析、信号肽、亚细胞定位、蛋白高级结构,利用实时荧光定量PCR检测PLIN2基因在延边牛不同组织中的表达水平。结果显示,延边牛PLIN2基因CDS长1 353 bp,编码450个氨基酸;同源性比对发现延边牛PLIN2基因与黄牛、水牛、绵羊、山羊、野猪、人、小鼠和野鸡的同源性分别为100%、98.7%、96.7%、97.1%、85.5%、86.3%、47.3%和49.1%;系统进化树表明,延边牛与黄牛、水牛的亲缘关系最近,与野鸡的亲缘关系最远。PLIN2蛋白为不稳定蛋白,具有一定亲水性,存在61个潜在的磷酸化位点、16个O-糖基化潜在位点、12个N-糖基化潜在位点,存在2个二硫键,不存在信号肽,主要分布于细胞核中,细胞质和线粒体中有少量分布。PLIN2蛋白高级结构预测该蛋白是由α-螺旋、延伸链、无规则卷曲和β-转角组成,为混合型蛋白,通过无规则卷曲连接,以α-螺旋为主。实时荧光定量PCR结果显示,PLIN2基因在延边牛小肠组织中表达量最高,其次为背最长肌和肾脏组织。本研究结果可为进一步研究PLIN2基因的功能提供借鉴。

Several studies have demonstrated that the Chinese herb Gastrodia elata Blume can protect against amyloid beta-peptide （Ap）-induced cell death. To investigate the possible therapeutic effects of Gastrodia elata Blume on Alzheimer＇s disease, we established a rat model of AIzheimer＇s disease by injecting A325-35 into bilateral hippocampi. These rats were intragastrically administered 500 or 1 000 mg/kg Gastrodia elata Blume per day for 52 consecutive days. Morris water maze tests showed that Gastrodia elata Blume treatment significantly improved the spatial memory of Alzheimer＇s disease rats. Congo red staining revealed that Gastrodia elata Blume significantly reduced the number of amyloid deposits in the hippocampus of these rats. Western blot analysis showed that choline acetyltransferase expression in the medial septum and hippocampus was significantly increased by the treatment of Gastrodia elata Blume, while EIIman method showed significant decrease in the activity of acetylcholinesterase in all three regions （prefrontal cortex, medial septum and hippocampus）. These findings suggest that long-term administration of Gastrodia elata Blume has therapeutic potential for Alzheimer＇s disease.

C16 saturated fatty acid (Palmitic acid) is one of the most common dietary fatty acids which played an important role in the cellular biological functions. Palmitic acid (PA) was tested for potential inhibition of DNA topoisomerase I (topo I) and it exhibited inhibitory activity in the nanomolar range. Treatment of lung adenocarcinoma cell line A549 with PA resulted in a decrease in cell viability in a concentration-dependent manner, and PA showed cytotoxicity with an IC50 value of 150 μM. DNA fragmentation assay and caspase activity indicated that PA does not induce apoptotic cell death in A549 cells. Finally, we found that PA was able to cause an increase in autophagic flux in a time-dependent manner, evidenced by the accumulation of LC3 through monodansylcadaverine (MDC) staining. More importantly, inhibition of autophagy by blocking autophagosome formation via the inhibition of type III Phosphatidylinositol 3-kinases (PI-3K), by 3-Methyladenine (3-MA) was able to effectively suppress PA-induced autophagy. We showed that inhibition of autophagy sensitized the cells signal to PA-induced apoptosis, suggesting the pro-survival function of autophagy induced by PA. Taken together, results from this study reveal that PA as a topo I inhibitor induced autophagic cell death in A549 cells.

AIM:To investigate the risk factors and characteristics of hepatocellular carcinoma(HCC) in the patients with drug-resistant chronic hepatitis B(CHB).METHODS:A total of 432 patients with drug-resistant CHB were analyzed retrospectively from January 2004to December 2012. The patients were divided into two groups:the HCC group(n = 57) and the non-HCC group(n = 375). Two groups compared using logistic regression for various patients and viral characteristics in order to identify associated risk factors for ***,patient and tumor characteristics of HCC patients with na ve CHB(N group,n = 117) were compared to the HCC group(R group,n = 57) to identify any difference in HCC characteristics between ***:A significant difference was found for age,platelet count,alpha-fetoprotein(AFP),positivity of HBeAg,seroconversion rate of HBeAg,virologic response,the Child-Pugh score,presence of rtM204I,and the duration of antiviral treatment in non-HCC and HCC group. Cirrhosis,age(> 50 years),HBeAg(+),virologic non-responder status,and rtM204I mutants were independent risk factors for the development of HCC. The R group had lower serum C-reactive protein(CRP) and AFP levels,earlier stage tumors,and a shorter mean tumor surveillance period than the N group. However,the total follow-up duration was not significantly different between the two ***:13.2% of patients with drug-resistant CHB developed HCC. Age,cirrhosis,YIDD status,HBeAg status,and virologic response are associated with risk of HCC. Patients with drug-resistant CHB and these clinical factors may benefit from closer HCC surveillance.
