Early selection of bread wheat genotypes using morphological and photosynthetic attributes conferring drought tolerance

Genetic diversity is the base of any genetic improvement breeding program aimed at stress *** variability among breeding materials is of primary importance in the achievements of a good crop ***,105 wheat genotypes were screened against drought stress using factorial completely randomized design at seedling stage to determine the genetic diversity and traits association conferring drought *** of variances revealed that all the studied parameters differed significantly among all genotypes,indicating the significance genetic variability existed among all genotypes for studied *** 10 best performance genotypes G1,G6,G11,G16,G21,G26,G39,G44,G51,and G61 were screened as drought tolerant,while five lowest performance genotypes G3,G77,G91,G98,and G105 were screened as drought *** length,chlorophyll a,chlorophyll b,and carotenoid contents were significantly correlated among themselves which exhibited the importance of these indices for rainfed areas in future wheat breeding *** length exhibited non-significant and negative association with other studied traits,and its selection seems not to be a promising criteria for this germplasm for drought *** performance genotypes under drought stress conditions will be useful in future wheat breeding program and early selection will be effective for developing high yielding and drought tolerant wheat varieties.

Pedigree and SSR Data Analysis Reveal Dominant Prevalence of Few Parents in Pedigrees of Pakistani Wheat Varieties

The international recognition of the importance of genetic diversity demands continuous estimation of genetic diversity of in hand population as test of its buffering capacity against all putative threats. Randomly selected Pakistani wheat varieties developed during 1965-1999 and 2000-2011 were evaluated on the basis of pedigree and SSR data. At 2nd and 3rd levels of pedigree, average occurrence of a parent per variety was 2.1 times. The dominating parents included BLUEBIRD, KALYANSONA and SIETE-CERROS-66, which were present in the pedigrees of 71.42%, 64.28%, and 58.57% varieties, respectively. The varieties INQLAB-91 and KIRAN-95 had the same pedigree and were genetically identical as revealed by SSR data. Similarly, varieties PAVON-76 and SOGHAT-90 also had the same parents in their pedigrees. This genetic similarity was also confirmed by SSR based cluster. The SSR based PC1 and PC2 showed narrow genetic diversity confirming the presence of few dominating parents. The results emphasize the inclusion of novel and genetically diverse parents in Pakistani wheat breeding programs to maintain broader genetic base of varieties/cultivars for buffering the effects of ever changing virulent pathogens and crop growth environments.

Family and/or Friends? Gene Mapping at Crossroads

Mapping gene(s) underlying a specific trait offers an opportunity to plant breeders to apply marker assisted selection. All gene mapping approaches except LD mapping use family based segregation populations developed by crossing two or more parents. These family based gene mapping approaches include simple interval mapping, composite interval mapping, multiple interval mapping and Bayesian mapping etc. Each approach has its own advantages and disadvantages based on type of population and underlying statistical model. Unlike family based approaches, LD mapping uses population of unrelated individuals which are like friends belonging to different family backgrounds. Relative pros and cons of family and friends based approaches make them complementary to each other. Family based approaches identify wide chromosomal region underlying the trait of interest with relatively lower markers density, and therefore, have low mapping resolution. Conversely, friends based LD mapping identifies chromosomal region of interest with higher resolution using higher marker density. The integration of family and friends based approaches addresses their respective pros and cons successfully to enhance mapping resolution for more valid application of marker assisted selection.

Stripe rust is a continuous threat to wheat crop all over the *** causes considerable yield losses in wheat crop every *** deployment of adult plant resistance(APR)genes in newly developing wheat cultivars is the most judicious strategy to combat this ***,we dissected the genetics underpinning stripe rust resistance in Pakistani wheat *** association panel of 94 spring wheat genotypes was phenotyped for two years to score the infestation of stripe rust on each accession and was scanned with 203 polymorphic *** on D’measure,linkage disequilibrium(LD)exhibited between loci distant up to 45 c ***-trait associations(MTAs)were determined using mixed linear model(MLM).Total 31 quantitative trait loci(QTLs)were observed on all 21 wheat *** QTLs were newly discovered as well as 19 QTLs and 35 previously reported Yr genes were validated in Pakistani wheat *** major QTLs were ***.2 AL and ***.3 BS(PVE,11.9%).Dissection of genes from the newly observed QTLs can provide new APR genes to improve genetic resources for APR resistance in wheat crop.

The Tale of Cotton Plant: From Wild Type to Domestication, Leading to Its Improvement by Genetic Transformation

Cotton is considered as a major cash crop of the world. It earns huge foreign exchange by its valuable products;fiber, lint, cotton seed oil, hull and a lot more. Being an important fiber crop, it earns huge foreign exchange by contributing to textile and seed oil industry. This review summarizes cotton biology, its diversity and domestication, genome assembly, constraints in its production and methods to improve cotton plant to fulfill the need of textile and oil industry. But cotton is facing enormous biotic and abiotic stresses with insect pests being most prominent. Massive destruction caused by insects needs to be controlled for maintaining fruitful cotton crop production. Conventional breeding approaches are limited to improving single trait and integrate stable genes within plant genome in approximately 7 - 8 years. Improved biotechnological procedures have paved new pathways to target genes specifically and improve cotton germplasm in lesser time than conventional breeding.

Genome Editing Tools: Need of the Current Era

Genome editing is considered as the most widely used approach of the present era. It had become a basic need of the current micro and molecular biological experiments. Gene engineering finds its widespread applications in medical, industry and agricultural sector. Unlike previous genetic engineering practices to insert or delete a part of genetic material at random place, genome editing allows the precise manipulation of DNA at a specific location. Zinc Finger Nucleases (ZFNs), Transcription Activator like Effector Nucleases (TALENs), Clustered Regularly Interspresed Short Palindromic repeats (CRISPR/Cas system) and meganucleases (recombinases) are the prime tools for editing an organism’s genome. Genome editing tools have an advantage to selectively delete or to integrate specific genes at specific loci. Use of recombinases for specifying site has further reduced time to integrate genes site specifically. Site specific gene stacking by the use of recombinases coupled with ZFNs, TALENs, or CRISPR/Cas genes have paved new pathways to target genes site specifically and to improve germplasm in lesser time than conventional breeding approaches.

Cotton is one of the most important fiber crops that plays a vital role in the textile *** production has been unstable over the years due to climate change induced biotic stresses such as insects,diseases,and weeds,as well as abiotic stresses including drought,salinity,heat,and *** breeding methods have been used to breed climate resilient cotton,but it requires a considerable amount of time to enhance crop tolerance to insect pests and changing climatic conditions.A promising strategy for improving tolerance against these stresses is genetic *** review article discusses the role of genetic engineering in cotton *** essential concepts and techniques include genome editing via clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(CRISPR-Cas9),overexpression of target genes,downregulation using RNA interference(RNAi),and virus-induced gene silencing(VIGS).Notably,the Agrobacterium-mediated transformation has made significant contributions to using these techniques for obtaining stable transgenic plants.

Kinetics of Acid Blue 1 Adsorption from Aqueous Solution by Carbonaceous Substrate Produced from Biotic Precursor

Adsorption of acid blue 1 from aqueous solution onto carbonaceous substrate produced from the wood of Paulownia tomentosa was investigated. The samples characterized by FTIR, SEM, EDS and XRD techniques, indicated that the surface functional groups like carboxyl, lactones or phenols and ethers have disappeared at high activation temperature （800 ℃） and as a result porous structure was developed that has a positive effect on the adsorption capacity. Bangham and parabolic diffusion models were applied to the kinetic adsorption data, which show that the adsorption of acid blue 1 was a diffusion controlled process. The reaction rate increased with the increase in temperatures of both the adsorption and activation. Thermodynamic parameters like △E^≠, △H^≠, △S^≠ and △G^≠ were calculated from the kinetic data. The negative values of △S^≠ reflected the decrease in the disorder of the system at the solid-solution interface during adsorption. Gibbs free energy （△G^≠）, representing the driving force for the affinity of dye for the carbon surface, increased with the increase in sample activation and the adsorption temperatures.

Characterization of hepatic progenitors from human fetal liver using CD34 as a hepatic progenitor marker

AIM: To enrich putative hepatic progenitors from the developing human fetal liver using CD34 as a marker. METHODS: Aborted fetuses of 13-20 wk were used for the isolation of liver cells. The cells were labeled with anti CD34; a marker used for isolating progenitor population and the cells were sorted using magnetic cell sorting. The positive fractions of cells were assessed for specific hepatic markers. Further, these cells were cultured in vitro for long term investigation. RESULTS: Flow cytometric and immunocytochemical analysis for alphafetoprotein (AFP) showed that the majority of the enriched CD34 positive cells were positive for AFP. Furthermore, these enriched cells proliferated in the long term and maintained hepatic characteristics in in vitro culture. CONCLUSION: The study shows that aborted human fetal liver is a potential source for isolation of hepatic progenitors for clinical applications. The study also demonstrates that CD34 can be a good marker for the enrichment of progenitor populations.

Helicobacter pylori and other Helicobacter species DNA in human bile samples from patients with various hepato-biliary diseases

AIM： To investigate the presence of Helicobacter species by nested PCR of 16S rRNA genes followed by the presence of Flelicobacter pylori （H pylori） 16S rRNA, ureA, cagA genes in bile obtained at endoscopic retrograde cholangio-pancreatography （ERCP） from 60 Indian subjects. METHODS： Sixty bile samples were obtained from patients diagnosed with various hepato-biliary diseases and control subjects at ERCP. PCR analysis was carried out using primers for Helicobacter genus 16S rRNA gene and H pylori （16S rRNA, urea and cagA） genes. Gastric H pylori status was also assessed from biopsies obtained at endoscopy from patients with various hepatobiliary diseases and controls. The control group mainly consisted of subjects with gastric disorders. Sequencing analysis was performed to confirm that PCR products with 16S rRNA and cagA primers were derived from H pylori. RESULTS No Helicobacters were grown in culture from the bile samples. Helicobacter DNA was detected in bile of 96.7% and 6.6% of groups Ⅰ and Ⅱ respectively. Ten from group Ⅰ were positive for 16S rRNA and ureA and 9 were positive for cagA gene. In contrast of the 2 from the control, 1 amplified with 16S rRNA, ureA and cagA primers used. The sequences of the 16S rRNA genes and cagA were 99% similar to Helicobacter pylori. CONCLUSION： Helicobacters are associated with the pathogenesis of various hepato-biliary disorders.
